Beckwith-Wiedemann syndrome multiomic analysis of hepatoblastoma uncovers unique tumour heterogeneity and cellular landscapes, including transition cells leading to tumour formation.

BACKGROUND: Beckwith-Wiedemann syndrome (BWS) is an overgrowth and cancer predisposition syndrome caused by epigenetic alterations on chromosome 11p15 that predisposes children to multiple cancer types, including hepatoblastoma. Hepatoblastoma is heterogenous in nature, and the 11p15 changes that cause BWS can also be found as a somatic alteration in nonBWS hepatoblastomas, further adding complexity to this disease. METHODS: To understand the impact of the predisposition molecular cues in BWS hepatoblastoma, we interrogated BWS and nonBWS hepatoblastomas, as well as adjacent normal liver, usi

SNORD60-mediated 2'-O-methylation of KCP enhances ferroptosis sensitivity in hepatoblastoma.

Ferroptosis has increasingly emerged as a novel target for cancer therapy because of intrinsic or acquired ferroptosis vulnerabilities in cancers. Small nucleolar RNA (snoRNA)-mediated mRNA modifications play a critical role in regulating ferroptosis and supporting tumor cell adaptation. However, the function and mechanism of the box C/D snoRNA SNORD60 in hepatoblastoma (HB) remain poorly understood. Here, we found that SNORD60 expression was markedly downregulated in HB tissues, and restoring its expression inhibited HB cell proliferation and induced ferroptosis both in vitro and in vivo. Mec

Proteasome Targeting with Carfilzomib Induces Reactive Oxygen Species-Mediated Apoptosis in Hepatoblastoma.

Hepatoblastoma (HB) is the most common malignant liver tumor in children, yet therapeutic options remain largely confined to conventional chemotherapy. To identify novel therapeutic targets, we performed gene set enrichment analysis on three publicly available HB datasets and found consistent activation of the proteasome pathway, with marked overexpression of the β5 proteolytic subunit encoded by PSMB5. High PSMB5 expression was associated with poor survival in adult hepatocellular carcinoma, highlighting the proteasome as a candidate for therapeutic vulnerability. Targeting the β5 proteolytic

Prognostic Genes Linked to Asparagine Metabolism in Hepatocellular Carcinoma: Identification, Validation, and Regulatory Mechanisms Based on Transcriptome and Single-Cell RNA Sequencing.

Metabolic reprogramming is closely linked to tumor proliferation, invasion, and immune escape. Despite its central role in amino acid metabolism, the regulatory mechanisms of asparagine metabolism in hepatocellular carcinoma (HCC) progression remain poorly characterized. Rather than focusing on canonical metabolic genes, prognostic markers were identified from co-expression modules associated with asparagine metabolism signatures. Using the TCGA database and asparagine metabolism-related gene sets, a prognostic risk-scoring model was developed through differential expression analysis, univaria

Regulatory T Cells and Tumor-Associated Macrophages Colocalize in Pediatric Hepatoblastoma.

INTRODUCTION: Hepatoblastoma (HB) is the most common liver cancer in infants and children. Human HB tumor specimens were analyzed to identify a distinct population of tumor-associated macrophages (TAMs). We queried whether TAMs cooperate with other regulatory immune populations (e.g., regulatory T cells [Tregs]) to establish an immunoregulatory environment that promotes HB formation. METHODS: Immunohistochemistry was used to identify CD68+ TAMs and CD4+FoxP3+ Tregs in nine HB tumors and adjacent normal liver. After receiving neoadjuvant chemotherapy, regions of viable and nonviable tumor (tre

Improving image quality and diagnostic confidence for PRETEXT staging in pediatric hepatoblastoma using thin-slice and low-energy virtual monochromatic images in dual-energy CT with deep learning image reconstruction algorithm.

BACKGROUND: Hepatoblastoma is the most common pediatric hepatic tumor, for which surgery is the primary treatment option. The PRETEXT (Pretreatment Extent of Disease) staging system, based on CT images, is a crucial basis for surgical planning. Therefore, improving the image quality and diagnostic confidence of PRETEXT staging impacts the overall therapeutic outcomes in pediatric hepatoblastoma. OBJECTIVE: To investigate whether thin-slice 40 keV dual-energy CT (DECT) images combined with a deep learning image reconstruction (DLIR) algorithm can improve image quality and diagnostic confidence

The landscape of pediatric hepatocellular carcinoma and fibrolamellar carcinoma: A report from the Children's Hepatic tumors International Collaboration (CHIC).

BACKGROUND: Pediatric hepatocellular carcinoma (HCC) and fibrolamellar carcinoma (FLC) have historically been studied on prospective trials in aggregate with hepatoblastoma (HB). It is now recognized that both HCC and FLC have unique histologies, characteristic genomics, and a more aggressive clinical course than HB. It is anticipated that data analyzed from patients treated on previously conducted international trials will inform current and future approaches to the study of these rare diseases. METHODS: The Children's Hepatic tumors International Collaboration houses data from clinical tria

Neoplasms in prune belly syndrome: A novel case of early-onset metastatic bladder carcinoma and systematic review of reported tumors.

PURPOSE: Prune Belly Syndrome (PBS) is a rare congenital malformation complex characterized by cryptorchidism, urinary tract abnormalities and deficiency of the abdominal wall musculature. Although advances in neonatal and reconstructive care have substantially improved survival, the long-term risk of malignancy in this population remains poorly defined. We report a novel case of early-onset metastatic bladder carcinoma in a patient with PBS and perform a systematic review of all tumors described in association with the syndrome. METHODS: A systematic literature review was performed according

Incidental Hepatoblastoma in an Infant Who Underwent Liver Transplantation for Cryptogenic Liver Cirrhosis: A Case Report.

BACKGROUND: Cryptogenic liver cirrhosis in infancy is rare, and its coexistence with hepatoblastoma (HB) has not been previously documented. While HB generally arises in morphologically normal livers, the incidental discovery of HB within cirrhotic livers is exceptional. CASE PRESENTATION: We report a 6-month-old male infant with progressive cholestasis. Prenatal assessments indicated growth restriction and blood flow redistribution, and he was delivered preterm at 33 weeks and 6 days with a birth weight of 861 g (-4.1 SD). Despite extensive clinical, metabolic, infectious, and genetic evalua

Macrophage Polarization and Spatial Architecture Characterize the Tumor Immune Microenvironment and Prognosis in Hepatoblastoma.

Hepatoblastoma, the most common primary pediatric liver malignancy, exhibits resistance to contemporary treatments in a subset of patients. Understanding the tumor immune microenvironment is essential for the development of novel immuno-oncological therapies. Herein, the microenvironment of hepatoblastoma was characterized by mapping the spatial distribution of immune cells, identifying prognostic immune features, assessing chemotherapy-induced alterations, and exploring potential immunotherapeutic targets. Whole-slide multiplex immunofluorescence staining (n = 35) was performed on formalin-fi

Propolis components from stingless bees collected in Gia Lai, Vietnam, and their NRF2 activation.

Stingless bee propolis is a promising source of cytoprotective metabolites. However, its chemical and biological properties, as well as its ability to modulate the nuclear factor E2-related factor 2 (NRF2) pathway, a key regulator of antioxidant response element (ARE)-driven cytoprotective gene expression, have not been adequately characterised. Compounds (1-9) were isolated from Vietnamese stingless bee propolis and identified as three xanthones and six sesquiterpenes by NMR and HR-ESI-MS analyses. Xanthone 2 and sesquiterpenes 4-9 are reported for the first time from propolis. Real-time biol

Biallelic ARID1A Alterations : A Promising Novel Biomarker for Risk Stratification and Management in Pediatric Malignant Hepatocellular Tumors.

Somatic mutations and copy number alterations of the adenine-thymine (AT)-rich interactive domain-containing protein 1A ( ARID1A ), a tumor suppressor gene, are associated with poor prognosis in many cancers, including adult hepatocellular carcinomas (HCC). In this study, ARID1A protein expression, clinicopathologic features, and outcomes were investigated in 59 pediatric malignant hepatocellular tumors (2004 to 2022) relative to ARID1A mutation and copy number status. Additional studies were performed on the impact of the loss of ARID1A protein expression on genomic instability, immune-cell i

Mapping the Binding Landscape of Allosteric Inhibitor G6PDi-1 on Human G6PD.

Targeting the oxidative pentose phosphate pathway by inhibiting glucose-6-phosphate dehydrogenase (G6PD) is a promising anticancer strategy, yet clinically useful inhibitors remain unavailable. A key limitation is the lack of molecular insight into how allosteric and noncompetitive inhibitors perturb enzymatic activity, limiting rational optimization. Here, we investigated the mechanism of G6PDi-1, a potent, reversible, noncompetitive G6PD inhibitor, by combining biochemical assays with molecular dynamics simulations, Markov state model, and MM/PBSA binding energy calculations. Experimentally,

Hilar lymph node involvement in pediatric liver cancer and its impact on outcomes: A systematic review.

UNLABELLED: Management of liver tumors in children is well-defined, with clear treatment protocols established by organizations such as the International Society of Pediatric Oncology-Liver group (SIOPEL), the Children's Oncology Group (COG), and the Japanese Study Group for Pediatric Liver Tumors (JPLT). However, no structured approach exists for hilar lymph node (LN) resection. Surgical practices vary among centers, reflecting the adult setting. This systematic review aims to examine the current evidence on hilar lymphadenectomy in pediatric patients with primary liver tumors and to assess L

Biocompatible Sulfobetaine Polymer-Artemisinin Conjugates Inducing Ferroptosis in Cancer Cells: Synthesis by Mechanochemical Solid-State Polymerization and Characterization.

Artemisinin derivatives (ARTs) induce ferrous iron-dependent cell death (ferroptosis) by generating free radicals. Since the concentration of ferrous ions within cancer cells is high, the specific delivery of ARTs into cancer cells is advantageous for cancer therapy. Herein, we fabricate the biocompatible sulfobetaine polymer-ARTs conjugates through the mechanochemical solid-state copolymerization of sulfobetaine methacrylate (SBMA) and ARTs-conjugated monomer. A polymer conversion rate of more than 90% is achieved within 60 min of copolymerization. The number average molecular weight and hete
