Fat-containing liver lesions expanding the magnetic resonance imaging signature of hepatoblastoma.

BACKGROUND: Fat-containing liver lesions have been widely discussed in the radiology literature and include various benign and malignant lesions. However, data on the presence and imaging characteristics of intralesional fat in hepatoblastoma remain limited. OBJECTIVE: To investigate the presence of intralesional fat in hepatoblastoma using dual-echo magnetic resonance imaging (MRI) and to correlate imaging findings with histopathology. MATERIALS AND METHODS: This retrospective study included 22 pediatric patients with hepatoblastoma who underwent pretreatment MRI with a dual-echo gradient-r

Three-dimensional reconstruction-assisted preoperative assessment and surgical planning for pediatric tumors with complex vascular relationships: a report of two cases.

BACKGROUND: Pediatric abdominal tumors with complex anatomical relationships are characterized by substantial morphological heterogeneity. Although three-dimensional reconstruction is increasingly used in surgical planning, reports that define its problem-oriented role in children with major vascular involvement remain limited. When a lesion is closely associated with major abdominal vessels, preoperative assessment should focus not only on visualization but also on resectability, preservation of critical vasculature, and perioperative risk stratification. CASE PRESENTATION: In case 1, an upp

Preclinical Drug-Response Profiling Identifies BMI1 Inhibition as a Therapeutic Option for Hepatoblastoma.

Hepatoblastoma (HB), the most common pediatric liver cancer, exhibits marked variability in therapeutic response despite minimal genetic heterogeneity, implicating epigenetic regulation as a key driver of tumor behavior. Among these, polycomb repressor complexes (PRC) remain poorly explored as therapeutic targets. Integrative analysis of samples from patients with HB and public datasets identified BMI1, a core component of PRC1, as significantly upregulated, with high expression strongly associated with aggressive disease and poor survival. Functional screening of epigenetic inhibitors across

T-cell receptor-engineered T-cell therapy using a glypican-3-specific TCR derived from a hepatoblastoma vaccine responder for HLA-A2-positive glypican-3-expressing cancers.

Glypican-3 (GPC3) is highly expressed in most hepatocellular carcinomas (HCCs) and hepatoblastoma, shows minimal expression in non-tumor adult tissues, and is also detected in several other malignancies, making it an ideal target for anticancer immunotherapy. We previously demonstrated in clinical trials that GPC3 peptide vaccination can induce durable antitumor immunity in patients with HCC and pediatric hepatoblastoma and established human leukocyte antigen (HLA)-A∗02:01-restricted GPC3-specific cytotoxic T lymphocyte (CTL) clones. In this study, we isolated high-avidity T-cell receptors (TC

Integrative transcriptomic and CRISPR dependency analysis identifies hepatoblastoma-specific essential genes and actionable vulnerabilities.

BACKGROUND: Hepatoblastoma (HB) is the most common primary liver malignancy in childhood, yet its molecular determinants, functional dependencies, and therapeutic vulnerabilities remain incompletely characterized. Integrative analyses combining transcriptomic profiling with functional genomic datasets provide a strategy to identify essential genes, biomarkers predictive of tumor behavior and treatment response. METHODS: Differential expression analysis comparing HB tumors with normal liver was processed on training cohort. These genes were integrated with DepMap CRISPR-Cas9 dependency scores

Global landscape of liver disease burden in 1990-2021.

Liver diseases remain a major global contributor to disability-adjusted life years (DALYs). Using Global Burden of Disease 2021 data, we applied a three-tier framework linking total liver diseases to acute hepatitis, cirrhosis, liver cancer, and their main etiologies. In 2021, liver diseases caused 309.72 million new cases, 1.98 million deaths, and 63.53 million DALYs worldwide. Although global age-standardized rates declined, death and DALY rates increased in High-income North America and Australasia, with steatosis- and alcohol-related liver diseases accounting for a growing share of the bur

Total vascular exclusion in pediatric hepatectomy: reducing hemorrhage without added morbidity.

BACKGROUND: Major hepatectomy in children carries distinct risks and challenges, with intraoperative hemorrhage being a critical determinant of morbidity and survival. Total vascular exclusion (TVE) provides complete vascular control, but its use in adults is associated with high morbidity. Evidence for pediatric hepatectomy is lacking. METHODS: A single-center study of all children (<18 years) undergoing major hepatectomy between 2013 and 2023 was performed. Patients were grouped by TVE use. Primary outcomes were major complications (Clavien-Madadi ≥ III) and posthepatectomy liver dysfunctio

A robust dynamic 3D liver spheroid platform for (sub)chronic toxicity assessment: Metabolic characterization of HepG2 spheroids as a case study.

In vitro assessment of (sub)chronic chemical exposure requires liver models that maintain stable functionality during long-term cultivation and whose baseline behavior is well characterized. HepG2 cells remain widely used in toxicology due to their robustness and reproducibility; however, their hepatoblastoma origin confers persistent tumor‑associated traits. Although three‑dimensional (3D) HepG2 spheroids provide greater physiological relevance than two‑dimensional (2D) cultures, their baseline behavior during prolonged dynamic cultivation remains insufficiently characterized, limiting confid

Associating Liver Partition and Portal Vein Ligation for Staged Hepatectomy (ALPPS) in Children with Advanced Hepatoblastoma-Lessons from a Case Series and Literature Review.

BACKGROUND: Liver transplantation is the standard treatment for children with advanced hepato-blastoma when complete resection is not feasible. However, transplantation may be contraindicated because of persistent metastatic disease, severe comorbidities, poor clinical condition, or donor-related limitations. Associating Liver Partition and Portal Vein Ligation for Staged Hepatectomy (ALPPS) has emerged as a potential rescue strategy in highly selected patients. This study evaluated the feasibility, safety, and oncological outcomes of ALPPS in pediatric hepatoblastoma. METHODS: A retrospectiv

SMAP1 promotes hepatoblastoma cell proliferation by modulating the C-Kit-activated ERK/MAPK pathway.

Hepatoblastoma (HB) is the most common pediatric liver malignancy. We investigated the role of SMAP1 in HB proliferation and its mechanism. Sixty-seven HB tissues were analyzed for SMAP1 expression via immunohistochemistry and correlated with clinical outcomes. qRT-PCR quantified SMAP1 mRNA in HB cell lines versus normal hepatocytes. SMAP1 was knocked down (siRNA/lentivirus) or overexpressed (plasmid) in HB cells; proliferation was assessed by CCK-8 and colony formation. A nude-mouse xenograft model was used to evaluate tumorigenicity. Western blotting determined C-Kit and ERK/MAPK pathway act
