ETV4 as a potential target gene for alleviating proliferation metastasis of HCC.

BACKGROUND: Hepatocellular carcinoma (HCC) develops and progresses via the RAF/MERK/ERK signaling pathway. Sorafenib, a first-line therapy for advanced HCC, inhibits the RAF gene. However, RAF dimerization and ERK activation caused by RAF inhibition in HCC, and this paradoxical RAF/MERK/ERK activation is one of the reasons for Sorafenib resistance in liver cell carcinoma. ETV4 is an ERK downstream gene, hence inhibiting it may help to reduce treatment resistance in HCC. The human hepatoma cell line HepG2.2.15 was used as an HCC tumor cell model, while the HL-7702 cell line was used as a human

Endoscopic ultrasound in the management of complications related to cirrhosis- recent evidence.

Advanced endoscopic methods like endoscopic ultrasound (EUS) have recently been applied for the management of complications related to cirrhosis. This includes novel techniques to measure portal pressure gradients, esophageal, gastric, and ectopic varices management, and EUS-assisted treatment of liver tumors. Management of liver tumors has seen significant changes in recent years, and many minimally invasive procedures have been approved, especially in the management of liver cell carcinoma. These are specialized procedures and are performed only in patients with cirrhosis in specialized cent

The causal relationship between platelet indices and liver cancer: a bidirectional two-sample Mendelian randomization study.

BACKGROUND: Platelet indices are non-invasive biomarkers used to assess disease status. Observational studies have shown that platelet indices are related to liver cancer. However, the specific relationship between the two is still inconclusive. We conducted a Mendelian randomization (MR) study to investigate the causal relationship between platelet indices and liver cancer. METHODS: We identified 2 cancer outcomes (liver cell carcinoma, malignant neoplasm of liver and intrahepatic bile ducts) from genome-wide association studies (GWAS). We used inverse variance weighted (IVW) method as the m

Analyzing the gut liver axis: a dual role of the microbiome in the genesis, progression, and treatment of liver cell carcinoma.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality worldwide, and the poor prognosis highlights the pressing need for innovative therapeutic strategies. The gut-liver axis, a critical bidirectional pathway linking the gut microbiota to the liver, plays a pivotal role in HCC pathogenesis. This review systematically delineates current evidence on how gut dysbiosis, compromised intestinal barrier function, and resultant microbial metabolites (e.g., bacterially metabolized bile acids) drive hepatocarcinogenesis via specific signaling pathways, while also addressing

The combination of Schisandrin C and Luteolin synergistically attenuates hepatitis B virus infection via repressing HBV replication and promoting cGAS-STING pathway activation in macrophages.

BACKGROUND: HBV infection can result in severe liver diseases and is one of the primary causes of liver cell carcinoma-related mortality. Liuwei Wuling tablet (LWWL) is a traditional Chinese medicine formula, with a protecting liver and decreasing enzyme activity, usually used to treat chronic hepatitis B with NAs in clinic. However, its main active ingredients and mechanism of action have not been fully investigated. Hence, we aimed to screen the active ingredient and effective ingredient combinations from Liuwei Wuling tablet to explore the anti-herpatitis B virus activity and mechanism. ME

Causal effects of circulating glutamine on colitis, IBD, and digestive system cancers: a Mendelian randomisation study.

Backgrounds: There is growing evidence linking glutamine levels to the risk of gastrointestinal diseases, yet the presence of a causal relationship remains uncertain. In this study, we employed a Mendelian randomization (MR) approach to investigate potential causal associations between glutamine and colitis, inflammatory bowel disease (IBD), and digestive tumors. Methods: Genetic instrumental variables for glutamine exposure were identified from a genome-wide association study (GWAS) involving 114,751 participants. We pooled statistics from GWAS of gastrointestinal diseases in European populat

An optimized instrument variable selection approach to improve causality estimation in association studies.

Mendelian randomization (MR) is an emerging tool for inferring causality in genetic epidemiology. MR studies suffer bias from weak genetic instrument variables (IVs) and horizontal pleiotropy. We introduce a robust integrative framework strictly adhering with STROBE-MR guidelines to improve causality inference through MR studies. We implemented novel t-statistics-based criteria to improve the reliability of selected IVs followed by various MR methods. Further, we include sensitivity analyses to remove horizontal-pleiotropy bias. For functional validation, we perform enrichment analysis of iden

Obesity and risk for liver disease: a two-sample Mendelian randomisation study.

The associations between obesity and liver diseases are complex and diverse. To explore the causal relationships between obesity and liver diseases, we applied two-sample Mendelian randomisation (MR) and multivariable MR analysis. The data of exposures (BMI and WHRadjBMI) and outcomes (liver diseases and liver function biomarker) were obtained from the open genome-wide association study database. A two-sample MR study revealed that the genetically predicted BMI and WHRadjBMI were associated with non-alcoholic fatty liver disease, liver fibrosis and autoimmune hepatitis. Obesity was not associa

Synthesis, characterization, and antiproliferative evaluation of novel sorafenib analogs for the treatment of hepatocellular carcinoma.

Cancer is a disease triggered by an uncontrolled proliferation of a cluster of cells, typically originating from a single cell. Sorafenib, a widely utilized pharmaceutical, has limitations in clinical use due to pharmacokinetic challenges and the development of resistance mechanisms. This investigation aimed to synthesize new sorafenib analogs and evaluated their activity against HepG2 cell lines, specifically targeting hepatocellular carcinoma (HCC). Seven sorafenib analogs were synthesized and identified by Fourier-transform infrared spectroscopy and 1H-NMR spectra. Cytotoxicity of the analo

[Treatment of parenterally transmittable viral hepatitis].

The parenterally transmittable hepatitides B, D and C and their complications are a problem worldwide and also in Germany that should not be underestimated. Due to the estimated high gray area, a broad distribution, particularly by drug abuse, increasing prevalence due to immigration and a pandemic-related delay in the diagnostics, the identification of affected persons and therefore potentially infectious patients represents a great challenge for the healthcare system. Highly effective treatment concepts with practically no side effects and a tablet ingestion once daily are available for hepa
