Lowering legal blood‑alcohol concentration limits for driving is an evidence‑based policy to prevent harms. In England and Wales, proposals to reduce the drink-drive limit from 80 mg to 50 mg alcohol per 100 ml of blood have generated competing narratives, including alcohol-industry claims about harm to rural pubs and public-health arguments emphasising road safety. We conducted a pre‑registered online experiment with adults in England and Wales (N = 495) to test how these narratives shape policy support. Participants read one of three news‑style articles framing the proposed reduction as alco

Ethanol represents the most abused drug worldwide, and among the most frequently evidenced in toxicological analysis on drug-induced/related deaths. When a blood sample is not available in a death characterized by severe trauma an alternative matrix is needed for an estimation of impairment. Presented is the correlation of postmortem blood and rectus abdominis muscle ethanol concentration. Ethanol concentrations in both blood (BAC) and muscle were determined by head space gas chromatography/mass spectrometry. Twenty-nine forensic cases with positive BAC were selected for the correlation study.

BACKGROUND: This Phase I randomized controlled trial aimed to assess the safety and tolerability of cannabidiol (CBD) and alcohol co-administration among moderate alcohol consumers in acute laboratory and chronic outpatient settings.
METHODS: Healthy adults ages 21-65 with moderate alcohol use completed three acute dosing sessions where they received either placebo, 50 mg CBD, or 100 mg CBD along with alcohol dosed to achieve peak blood alcohol concentrations (BAC) of 0.06 g/100 mL. Sessions were randomly ordered, and participants and outcome assessors were blinded to study drug during these

The direct blood-alcohol biomarker phosphatidylethanol (PEth), especially its most abundant analogue 1-palmitoyl-2-oleoyl-sn-phosphatidylethanol (PEth 16:0/18:1) has gained increasing relevance in clinical and forensic applications for assessing alcohol consumption. Accurate quantification of PEth is essential to reliably differentiate between abstinence, moderate alcohol consumption and excessive alcohol intake. Measurement accuracy of PEth 16:0/18:1 by well-established liquid chromatography-tandem mass spectrometry (LC-MS/MS) approaches such as multiple reaction monitoring (MRM) can be confo

BACKGROUND: Early-life stress constitutes a major risk factor for the development of neuropsychiatric and substance use disorders, exerting enduring effects on stress responsivity and emotional behavior. Maternal deprivation (MD) is a well-established model of early adversity that induces persistent neurobiological alterations. During adolescence, alcohol exposure presents an additional challenge that may interact with early-life stress, potentially influencing long-term adaptations in a sex-dependent manner. Among the systems involved, lipid signaling pathways, including the endocannabinoid s

Driving simulators are valuable for investigating driving impairment since ethical and safety considerations limit the ability to conduct such research on roads. Although simulators have generally demonstrated relative validity, alcohol is often referred to as a reference for impairment due to well-established legal limits and its known effects on driving ability. However, there is a relative lack of large-sample studies that systematically assess the sensitivity of established driving performance metrics to alcohol-induced impairment, alongside the application of rigorous methodological appro

BACKGROUND AND AIMS: Increased risk of alcohol dependence is recognised following bariatric surgery. Modified gastrointestinal anatomy and neurohormonal profiles lead to altered alcohol metabolism, peak blood alcohol concentrations and enhanced reward circuits and cravings. For patients with metabolic associated fatty liver disease, bariatric surgery has been shown to reduce liver fibrosis; however, the impact of bariatric surgery on alcohol consumption and consequent liver fibrosis has not been well-characterised using validated measures. This study examined the changes in alcohol consumption

BACKGROUND: Medical cannabis is now commonly prescribed for a range of chronic health conditions. Many medical cannabis products contain delta-9-tetrahydrocannabinol (THC), the intoxicating component in cannabis, though it is unclear whether these products produce impairment when used as prescribed and at therapeutic doses. With current Australian laws prohibiting driving with any amount of THC in one's system, this trial aims to generate novel data on the impact of prescribed medical cannabis on real-world driving performance to inform road safety policy.
METHODS: This is a two-phase trial,

BACKGROUND: Longitudinal excessive alcohol consumption has adverse effects on brain structure and functions. The immediate effects of alcohol consumption on the activity and functioning of different circuits in the brain are less known. This exploratory study aimed to examine the effects of acute alcohol exposure on cortical activity in healthy adults.
METHODS: The study included 34 adults, aged 29-36 years, who took part in the Youth and Alcohol Project's (YAP) alcohol exposure test. The participants' alcohol consumption was measured with the shortened version of the Alcohol Use Disorders Id

AIM: Sedative hypnotics can impair driving performance to a degree comparable to blood alcohol concentrations of 0.05%-0.08%. This study aimed to evaluate the residual effect of vornorexant, a novel orexin receptor antagonist, on next morning driving performance relative to a predefined clinically meaningful threshold.
METHODS: Participants received 10 and 20 mg of vornorexant and placebo for eight consecutive nights and zopiclone on Days 1 and 8 in a four-way crossover and double-blind manner. Driving performance at 9-h post-dose was assessed measuring the standard deviation of the lateral p

BACKGROUND: Clinical trials in neurological and psychiatric indications are hampered by poor measurement fidelity in currently used "standardized" rating scales. Digital, repeatable tests that can be remotely administered offer a more fine-grained understanding of the patient's clinical trajectory. Several such tools are being developed, but only a few have been validated in terms of their ability to discern and describe change over time-a critical element of clinical trials.
OBJECTIVE: Four cognitive tasks from a digital battery (delivered via tablet) are administered at high frequency follo

BACKGROUND: Alcohol-related driving fatalities persist as a public health issue in the United States. Recent studies have focused on the role of driver risk-taking in DUI occurrences and consequent driving fatalities. This laboratory study examined simulated driver risk-taking under an acute dose of alcohol and a placebo to test the degree to which individual differences in risk-taking were predicted by drivers' self-appraisal of their intoxication and the degree to which alcohol impaired their inhibitory control.
METHODS: Eighty young adult drivers (41 men and 39 women) received 0.65g/kg alc

BACKGROUND: Interoceptive processes may underlie maladaptive patterns of alcohol use. Bodily sensations experienced during alcohol intoxication could therefore reveal distinct mechanistic components relevant for addiction theory and research. Here, we apply novel tools to examine how intoxication impacts somatic awareness using bodily maps and a cardiac interoception task.
METHODS: In a double-blind, within-subjects, placebo-controlled study, social drinkers (n = 37, 17 female) were administered 0.4 g/kg of alcohol. We measured changes in self-reported bodily sensations during the ascending a

Alcohol consumption is known to affect dopamine (DA) release in the brain, with significant implications for understanding addiction and its neurobiological underpinnings. This meta-analysis examined the effects of acute alcohol administration on striatal DA release in healthy humans as measured with [11C]-raclopride positron emission tomography (PET). Oral alcohol administration was associated with a significant reduction in [11C]-raclopride binding potential (BPND) in the ventral striatum (Cohen's d = -0.76), indicative of increased DA release, particularly at lower blood alcohol concentrati
