Advances in DNA nanostructures for pathogenic microorganisms.

DNA, the genetic material of life, encodes genetic information for most organisms, including humans. Its unique double-helix structure and base-pairing rules not only provide the foundation for the transmission of genetic information, but also offer a natural molecular tool for nanomaterial construction. DNA nanostructures, composed of DNA, possess advantages such as controllable structures, simple assembly, easily modified, and excellent biocompatibility. These features make them highly promising for applications in tissue engineering and biomedicine, such as providing new strategies to comba

Rates of Evolution Differ Between Cell Types Identified by Single-Cell RNAseq in Threespine Stickleback.

Rates of evolutionary change vary by gene. While some broad gene categories are highly conserved with little divergence over time, others undergo continuous selection pressure and are highly divergent among populations or species. But such comparative evolutionary studies rely on gene ontology categories that are rarely validated in the study's focal species. An alternative is to determine species-specific gene functions using single-cell RNA sequencing (scRNAseq) that identifies cell types and the characteristic genes that each expresses. Here, we combine scRNAseq with evolutionary genomics t

Impact of anthropogenic activities on the ecosystem and emergence of bat-borne zoonotic diseases.

About 70 % of zoonotic infectious diseases originate from wildlife reservoirs, particularly bats, primates, and rodents. Bats comprise about 20 % of all known mammal species worldwide and have been identified as reservoir and carrier hosts of various viral disease outbreaks. They also play a crucial role in viral adaptation and evolution. Given this inherent risk, this review focuses on how anthropogenic activities (habitat destruction, agricultural intensification, bushmeat hunting, and occupational exposure) and climate change are increasing the frequency and intensity of bat-human contact.

Plastic-Derived Pollutants as Emerging Modifiers of Viral Diseases.

Plastic pollutants, including phthalates, bisphenol A (BPA), per- and polyfluoroalkyl substances (PFAS), and microplastics (MPs), are increasingly recognized as emerging environmental cofactors that intersect with infectious disease dynamics. These compounds, once considered inert, can alter immune function, reshape host-pathogen interactions, and directly influence viral survival and transmission. In this review, we compile current evidence on the chemistry, environmental occurrence, and biological activity of major plastic-associated pollutants with emphasis on their role in viral infections

TMPRSS2 rs12329760 variant as a prognostic marker for prostate cancer progression.

BACKGROUND: TMPRSS2, a transmembrane serine protease and common partner in oncogenic TMPRSS2-ERG fusions, is regulated by androgen receptors and is implicated in prostate tumorigenesis. The nonsynonymous SNP rs12329760 (C > T; Val160Met) has been hypothesized to alter TMPRSS2 proteolytic activity, potentially enhancing AR signaling and contributing to aggressive tumor behavior. However, data on this variant in North African populations remain sparse. METHODS: A case-control study was conducted including 50 Moroccan men diagnosed with prostate cancer (PCa) and 50 matched controls. Genotyping o

Genomic and patient epidemiology of Streptococcus dysgalactiae subspecies equisimilis in Houston, Texas.

UNLABELLED: Many countries have reported increased human infections caused by Streptococcus dysgalactiae subspecies equisimilis (SDSE) in the last 15 years. However, there is scant molecular epidemiology data for SDSE in the United States, especially at the whole genome level. To address this knowledge deficit, we studied SDSE infections in a large health care system in the Houston, Texas, metroplex, a diverse population of 7.4 million. We used Illumina whole-genome sequencing to characterize 865 human isolates collected consecutively from unique patients with diverse infections in the Houston

Intranasal DNA nanocarrier vaccines with surface-patterned antigens enhance efficacy against respiratory syncytial virus.

Although injectable respiratory syncytial virus (RSV) pre-F vaccines are clinically established, effective intranasal alternatives remain elusive. Geometric and surface antigen display properties are critical for respiratory B cell activation, yet lack strategies for systematic optimization. Here we report a library of DNA nanocarriers with controlled dimensions and sizes, aiming to systemically evaluate the influence of geometric properties on intranasal retention. Taking advantage of the precise control on DNA nanocarriers and antigen functionalization, we organized the surface antigen patte

Predictive performance of weekly rectal surveillance cultures and extra-rectal colonization for carbapenem-resistant Enterobacterales infections in a resource-limited ICU with KPC and NDM co-circulation.

BACKGROUND: Carbapenem-resistant Enterobacterales (CRE) cause hard-to-treat infections. Rectal surveillance is widely used to detect carriers, but its predictive value where different carbapenemases co-circulate is unclear. We assessed whether CRE rectal and extra-rectal colonization predicts subsequent infection in an intensive care unit with concurrent KPC and NDM circulation. METHODS: We performed a retrospective observational study in a 60-bed adult intensive care unit in Buenos Aires, Argentina, from July 2016 to September 2019. All patients underwent weekly rectal surveillance, and extr

Human Genetic Analysis Reveals Circulating Alpha-1 Antitrypsin Level as a Protective Factor in Sepsis.

Sepsis is a dysregulated host response to infection and a leading cause of global mortality, yet effective targeted therapies remain lacking. Here, we applied a proteogenomic framework integrating large-scale human genetics with circulating proteomics to identify therapeutic targets. In a meta-analysis of genome-wide association studies of 60,314 sepsis cases and 1,464,733 controls, we identified four genome-wide significant loci, including a missense variant in SERPINA1, encoding alpha-1 antitrypsin (AAT), that was also associated with 30-day sepsis mortality in the UK Biobank. Impact of Pneumococcal Conjugate Vaccine Immunization on Nasopharyngeal Carriage of Streptococcus pneumoniae in Vaccinated Children Under 5 Years Old in El Salvador.

BACKGROUND: Following pneumococcal conjugate vaccine (PCV) introduction in El Salvador, data on pneumococcal nasopharyngeal (NP) carriage in children are limited. This study assessed NP carriage of Streptococcus pneumoniae in healthy Salvadoran children aged 1-4 years to evaluate the impact of different vaccine schedules (PCV10, PCV13, or PCV10 plus PCV13). METHODS: In this analytical cross-sectional study, 380 fully vaccinated children attending well-child visits were enrolled over 1 year. Nasopharyngeal swabs were collected to determine S. pneumoniae carriage prevalence, serotype distributi

High frequency of the HLA-G*01:05N null allele in Beninese populations and its potential impact on reduced soluble HLA-G expression.

Human Leucocyte Antigen (HLA)-G, a non-classical HLA class Ib molecule, is involved in feto-maternal immune tolerance. Among HLA-G alleles, HLA-G*01:05N is defined by a cytosine deletion (ΔC) at exon 3, which causes a premature stop codon at position 189 in exon 4, disrupting the translation of HLA-G1, HLA-G4, and HLA-G5 isoforms. The modulation of HLA-G expression in carriers of the HLA-G*01:05N allele could have important immune and pathologic consequences. We investigated the HLA-G*01:05N allele frequency in two Beninese populations and evaluated its association with HLA-G plasma levels. HL

The dynamics of hepatitis B transmission with treatment and vaccination: A case study in the Ili region of Xinjiang, China, by a physics-informed neural networks approach.

Hepatitis B virus (HBV) infection remains a leading cause of cirrhosis and hepatocellular carcinoma worldwide, and long-term control relies on sustained immunization and effective reduction of transmission. In high-burden settings, vaccination programs have expanded substantially, but there still exist differences in the immune response effect after vaccination and the phenomenon of post-vaccination immunity decline. In this paper, we propose a model incorporating the vaccination of newborns and susceptible and imperfect vaccination, and study how the two factors affect the transmission dynami

Injectable dual-affinity self-assembling peptide hydrogels for local controlled delivery of therapeutic antibodies.

Therapeutic monoclonal antibodies have revolutionized the treatment of cancers, infectious diseases and immune disorders; however, their efficacy is compromised by limitations associated with systemic administration, including systemic toxicity, poor patient compliance and inadequate drug concentration at pathological sites. Hydrogels are promising carriers for localized antibody delivery, but conventional formulations fail to simultaneously address the dual challenges of uncontrolled antibody release and insufficient hydrogel retention at the target site. Herein, leveraging a peptide co-assem

Association of HLA class II haplotypes with antibody concentrations after diphtheria-tetanus acellular pertussis booster vaccination in four age groups of Finnish participants.

BACKGROUND: Despite widespread use of whole-cell (wP) and acellular (aP) pertussis vaccines, outbreaks persist in many countries. Polymorphisms in HLA class II molecules, which present antigens to CD4+ T cells, may play a vital role in vaccine responses. Emerging evidence suggests that HLA-DR/DQ variants can significantly influence wP vaccine responses. This study investigated the possible association of HLA class II haplotypes with antibody concentrations after aP booster vaccination. MATERIALS AND METHODS: Healthy Finnish children (7-10y, n = 37), adolescents (11-15y, n = 37), young adults

Serotype-specific pneumococcal invasiveness: a global meta-analysis of paired estimates of disease incidence and carriage prevalence.

BACKGROUND: Serotype-specific estimates of pneumococcal invasiveness used in pneumococcal carriage transmission models to predict changes in disease incidence post-vaccination are largely derived from high-income settings. We conducted a systematic review of carriage prevalence and invasive pneumococcal disease (IPD) incidence to calculate case-carrier ratios (CCRs) in different income settings. METHODS: We conducted a systematic search of MEDLINE, Embase, and Global Health databases on March 14, 2022, to identify publications on pneumococcal carriage prevalence or IPD incidence; we requested

BACKGROUND & AIMS: Hepatology is experiencing major shifts in disease etiologies and is raising demand for multidisciplinary care. However, a globally harmonized framework defining core training content remains lacking. We aimed to develop a globally informed expert consensus framework for the content of core hepatology training, to provide a structured foundation for curriculum development. METHODS: A two-round modified Delphi using the RAND/UCLA Appropriateness Method was conducted. A comprehensive list of curriculum items was developed from international training standards, refined by a st

Single unscreened carrier triggered ICU outbreak of a KPC-producing Klebsiella pneumoniae which acquired in vivo resistance to ceftazidime-avibactam, and cefiderocol.

BACKGROUND: Carbapenemase-producing Enterobacterales are a major cause of healthcare-associated outbreaks in intensive care units (ICUs), where unrecognized carriers can drive silent transmission. We report an ICU outbreak caused by KPC-producing Klebsiella pneumoniae and the within-host emergence of resistance to ceftazidime-avibactam and cefiderocol in the index case. METHODS: Four ICU patients with five K. pneumoniae isolates identified between July and August 2023 were investigated. Phenotypic, genomic and functional analyses were performed to determine the clonal relatedness of the isola

Beyond the tip of the iceberg: quantifying the odds of multidrug-resistance carriers for receiving post-acute follow-up rehabilitation after hospital stay - a real-world study using German data.

INTRODUCTION: Health equity means equal opportunities for all patients. While follow-up rehabilitation (German: Anschlussheilbehandlung (AHB)) improves patient-outcomes and reduces societal burdens of disease, patients colonised or infected with multidrug-resistant organisms (MDRO-carriers) have reportedly been denied admission by post-acute care (PAC) providers in Germany. This might however only be the tip of an 'iceberg', since carriers might drop out during earlier application processes. In this study, we quantify MDRO associated dropouts and model the odds of dropping out during different
