Interleukin- 17A as an Immunomodulatory Cytokine in Animal Health and Diseases: A Systematic Review.

Interleukin-17A (IL-17A), a pro-inflammatory cytokine primarily produced by Th17 cells, has emerged as a pivotal immunological mediator in both protective and pathological processes across different animal species in addition to human beings. Over the past decade, research has expanded significantly, elucidating IL-17A's roles in host defense, immune modulation, disease progression and vaccine induced immune responses. The present review focuses on recent advances in the understanding of IL-17A within the context of veterinary medicine. IL-17A has been implicated in protection from various dis

Phagocytosis of Mycobacterium fortuitum by Caprine Alveolar Macrophages Is Associated with iNOS and Pro-Inflammatory Markers Expression.

Mycobacterial diseases, including tuberculosis (TB), remain the major health and economic challenges in livestock, underscoring the need to characterise the innate immune mechanisms involved in early bacterial containment. Alveolar macrophages (AMs) are the first line of defence against inhaled mycobacteria, yet the functional links between activation, polarisation, and phagocytic capacity in caprine AMs remain poorly defined. In this study, we compared a pH-dependent live-cell fluorescence assay with a culture-based method to evaluate phagocytosis and clearance of Mycobacterium fortuitum unde

An ESAT-6-convergent prime-boost vaccination combining recombinant BCG expressing Mycobacterium marinum ESX-1 and ESAT-6/GLA-SE improves TB protection.

Although Bacille Calmette-Guérin (BCG) protects children against disseminated tuberculosis (TB), its limited efficacy against adult pulmonary TB underscores the need for improved vaccination strategies. We previously developed a recombinant BCG strain expressing the ESX-1 type VII secretion system of Mycobacterium marinum (BCG::ESX-1Mmar), which enhances immunogenicity through cytosolic immune signaling while maintaining low virulence. Here, we evaluated an ESAT-6-convergent prime-boost vaccination strategy in which mice were primed with ESX-1-competent BCG::ESX-1Mmar and subsequently boosted

Host Biomarkers in Tuberculosis.

Host-derived biomarkers are high-priority targets for development and could play important roles in diagnosis of various states of Mycobacterium tuberculosis (Mtb) infection and disease, prediction of outcomes, and might guide vaccine and drug regimen evaluation. We discuss the need for priority tuberculosis (TB) biomarkers, including markers of immune sensitization, prediction of progression, diagnosis of asymptomatic and symptomatic TB, including extrapulmonary TB, and treatment monitoring. The focus of the review is on ex vivo transcriptomic and proteomic markers as these are amenable to th

Genetic interference of distinctive Mycobacterium tuberculosis peptidoglycan modifications enhances β-lactam susceptibility and reveals expression-sensitive host immune dynamics.

The high mortality associated with tuberculosis (TB), alongside the lack of efficient therapeutics against emerging multidrug-resistant Mycobacterium tuberculosis (Mtb) strains, emphasizes the need for novel antitubercular targets. Mycobacterial peptidoglycan (PG), displaying characteristic modifications comprising the amidation of D-iso-glutamate (D-iGlu) and the N-glycolylation of muramic acid, is therefore a promising therapeutic target. The genes encoding the enzymes mediating these modifications (murT/gatD and namH) were silenced in Mtb using CRISPR interference (CRISPRi) to investigate t

New Therapeutic Cancer Vaccine with Lenalidomide Induces Potent T Cell Immunity and Inhibits HPV E6/E7-Expressing Tumor Growth in Mice.

Current prophylactic human papillomavirus (HPV) vaccines prevent infection but have limited therapeutic benefit in established HPV-driven malignancies. To address this unmet need, we developed VTX-067, a modular self-assembling therapeutic cancer vaccine consisting of a recombinant Mycobacterium tuberculosis heat shock protein 70-avidin fusion (MAV) noncovalently bound to biotinylated HPV E6/E7 MHC class I- and II-restricted peptide concatemers. MAV is produced in CHO cells with minimal endotoxin. Assembled VTX-067 is designed to elicit both CD4⁺ and CD8⁺ T-cell responses against HPV-associate

Pathological, microbiological and immunological performance of three tuberculosis vaccine candidates in goats.

Test and cull-based animal tuberculosis eradication programs have not succeeded in eliminating TB in various regions and implementing those programs is not feasible in others. Vaccination represents an additional control tool under study. Here we evaluated the immunologic, pathologic, and microbiological effects of heat-inactivated Mycobacterium bovis (HIMB), phage-inactivated Mycobacterium caprae (PIMC) and live Mycobacterium microti (LMM) subcutaneous vaccination in goats challenged endotracheally with M. caprae. All three vaccines were compatible with interferon-γ release assay diagnosis us

Superior protection against tuberculosis using heterologous mRNA nanoadjuvant vaccines.

Tuberculosis (TB) remains a major global health threat, underscoring the need for vaccines that surpass BCG efficacy. We developed QTAP-R, a novel mRNA-lipid nanoparticle (LNP) vaccine encoding Ag85B, Hsp70, and ESAT-6, to enhance immunity against Mycobacterium tuberculosis. QTAP efficiently encapsulated and delivered mRNA with high transfection efficiency and low cytotoxicity. In C57BL/6 mice, QTAP-R induced strong antigen-specific IgG and T-cell responses, including elevated CD4⁺ and CD8⁺ activation and increased polyfunctional cytokines (IFN-γ, TNF-α, IL-2, IL-17A). When combined with BCG (

Early immune dysregulation in Mtb/SIV co-infection resists cART treatment at the single-cell level.

Using single-cell transcriptomics of bronchoalveolar lavage cells from Mtb/SIV co-infected rhesus macaques on cART, we reveal profound immune dysregulation during early SIV co-infection of latent tuberculosis. SIV induces a sharp decline in CD4+ T cells, NK, and NKT cells, with incomplete recovery of Mtb-specific TH1 effector responses despite viral suppression. Instead, a persistent TH17-skewed environment emerges, alongside sustained myeloid inflammation driven by Type I interferon signaling and pro-inflammatory regulators such as KLF6 and NFKB1. Ligand-receptor network analyses demonstrate

BVL3572S inhibits HisC and AlaA, exploiting vitamin B6 dependency to kill Mycobacterium tuberculosis.

Tuberculosis remains the leading cause of death from a single infectious agent, and rising multi-drug resistance in Mycobacterium tuberculosis (Mtb) underscores the urgent need for new antibiotics. Here, we characterize BVL3572S, a hydroxamic acid-containing compound that is bactericidal against Mtb. The primary targets of BVL3572S are the pyridoxal phosphate (PLP; active form of vitamin B6)-dependent aminotransferases HisC (Rv1600) and AlaA (Rv0337c; formerly AspC), simultaneously impacting L-histidine and L-alanine biosynthesis. X-ray crystallography revealed a covalent PLP-BVL3572S adduct w

Protective monoclonal antibody targeting mycobacterial HBHA blocks the interaction with the novel attachment co-receptor NRP1 to inhibit Mycobacterium bovis entry.

INTRODUCTION: The immune protection conferred by Bacillus Calmette-Guérin (BCG) is insufficient, underscoring the urgent need for novel strategies to control tuberculosis (TB). Although antibody-mediated responses targeting mycobacterial cell wall antigens have been observed during infection, the precise mechanism by which antibodies resist TB pathogens remains unclear. OBJECTIVES: This study aims to elucidate the molecular mechanism by which protective monoclonal antibodies (mAbs) against Mycobacterium bovis (M. bovis) cell wall antigens resist M. bovis infection. METHODS: In this study, we

Rpf-Toxo: A Preliminary Computationally Designed Dense Granule Antigen-Based Multi-Epitope Vaccine Against Toxoplasma gondii.

BACKGROUND: Toxoplasma gondii is a protozoan parasite of medical and veterinary importance causing abortion. While current therapies are limited to address the chronic phase of infection, effective prophylactic vaccines are warranted. OBJECTIVES: In this study, we applied a rational vaccine design approach centred around immunodominant dense granule antigens (GRAs). METHODS: In silico epitope mapping was performed on six GRAs (GRA15, GRA60, GRA76, GRA83, GRA-α and GRA-β). Using web servers, B-cell, cytotoxic T-lymphocyte and helper T-lymphocyte epitopes were predicted and then filtered for a

The roles of pre-season immunity, age, viral shedding, and community exposures in shaping influenza household transmission dynamics.

Protective immunity against influenza transmission remains poorly understood due to the high prevalence of undetected asymptomatic infections. To address this, we analyze 1518 individuals and 623 infection episodes from a three-year, rural and urban, South African household cohort study that combines pre-season serum collection with twice-weekly virological testing regardless of symptoms. By developing a subtype/lineage-specific household transmission model and incorporating time-resolved viral shedding kinetics for all observed infections, we disentangle household and community exposure, pre-

BCG immunisation through intranasal instillation protects mice against Streptococcus pneumoniae infection by enhancing alveolar macrophage activity.

Pneumonia remains a leading cause of morbidity and mortality worldwide, especially among young children, the elderly, and immunocompromised individuals. Streptococcus pneumoniae is the most common bacterial cause of community-acquired pneumonia, and the rise of antimicrobial resistance highlights the urgent need for innovative approaches to strengthen host defences. The Bacille Calmette-Guérin (BCG) vaccine, originally developed to prevent tuberculosis, has been shown to provide heterologous protection against unrelated pathogens, mediated by diverse innate and adaptive immune mechanisms. Computational identification of antigens and development of a multi-epitope vaccine for bacterial vaginosis caused by Gardnerella vaginalis.

Bacterial vaginosis (BV), a prevalent form of vaginal dysbiosis, is primarily caused by Gardnerella vaginalis. The increasing prevalence of BV has raised global health concerns, prompting the need for a vaccine that can enhance human immunity and mitigate BV transmission. The study developed a multi-epitope vaccine for BV infection using immunoinformatic methodologies. B-cell and T-cell epitopes were identified using the IEDB recommended server and NetCTL. Specifically, HTL epitopes were predicted against HLA-DR alleles (including DRB101:01, DRB103:05, and DRB104:04), while CTL epitopes were p

Targeting mycobacterial PPE65 protein to develop a multi-epitope vaccine candidate against tuberculosis.

Tuberculosis is a deadly infectious disease, and the current BCG vaccine being inconsistent in protecting against it, underscores the urgent need to develop better therapeutics. PPE65 is a surface-localized and highly immunogenic protein of Mycobacterium tuberculosis, and has been proposed as a potential vaccine target because of its ability to induce both humoral and cellular immunity. Hence, the present study outlines the systematic design and detailed computational and in vitro assessment of a novel multi-epitope vaccine against M. tuberculosis utilizing PPE65 protein as a core antigen.
