Immunization of low-density lipoprotein receptor deficient (LDL-RD) mice with heat shock protein 65 (HSP-65) promotes early atherosclerosis.

Heat shock proteins are a family of approximately 25 highly conserved proteins upregulated in response to various forms of stress. They play an active role in the development autoimmune diseases in animals, and have been incriminated in human autoimmune diseases (i.e. rheumatoid arthritis, multiple sclerosis). It has been previously shown, that an induced immune response against Heat shock protein 65 (HSP-65) results in atherosclerotic lesions in normocholesterolemic rabbits. We have supported these findings showing that C57BL/6 mice immunized with HSP-65 and fed a high-fat diet develop enhanc

Vaccine adverse events in the new millennium: is there reason for concern?

As more and more infectious agents become targets for immunization programmes, the spectrum of adverse events linked to vaccines has been widening. Although some of these links are tenuous, relatively little is known about the immunopathogenesis of even the best characterized vaccine-associated adverse events (VAAEs). The range of possible use of active immunization is rapidly expanding to include vaccines against infectious diseases that require cellular responses to provide protection (e.g. tuberculosis, herpes viral infections), therapeutic vaccines for chronic infections (e.g. human immuno

Veterinary tuberculosis vaccine development.

Tuberculosis caused by Mycobacterium bovis in domestic livestock and wildlife is a significant problem in many countries worldwide. Wildlife reservoirs of tuberculosis confound programs for tuberculosis eradication from domestic livestock. Successful vaccination against tuberculosis in domestic animals or wildlife could contribute to tuberculosis eradication. Bacille Calmette-Guérin (BCG) has been used as the prototype vaccine for domestic livestock and wildlife. The majority of studies have been carried out with BCG-vaccinated animals challenged experimentally with M. bovis. Although protecti

Prior BCG vaccination improves survival of Gambian patients treated for pulmonary tuberculosis.

The protection provided by BCG against pulmonary tuberculosis ranges from nil to over 90%. While BCG protects against the more serious forms of tuberculosis, it is not known whether or not it protects patients with pulmonary tuberculosis from death. In a study designed to look at the effects of immunotherapy with M. vaccae as an adjunct to chemotherapy in 285 adult Gambian patients treated for proven pulmonary tuberculosis, we examined the association between the presence or absence of a BCG scar and mortality. The data showed that subjects who had a BCG scar were significantly younger than th

The development of wildlife control strategies for eradication of tuberculosis in cattle in Ireland.

Wildlife species, such as badgers, act as maintenance hosts for Mycobacterium bovis and contribute to the spread and persistence of tuberculosis in associated cattle populations. In areas in which there is a tuberculosis problem affecting a number of herds, the involvement of infected wildlife in the introduction of M. bovis infection into herds act as a constraint to eradication of the disease. Epidemiological evidence demonstrates a high prevalence of tuberculosis in badgers, and controlled studies involving comprehensive badger removal have shown that this strategy can serve to significantl

Latent Mycobacterium tuberculosis-persistence, patience, and winning by waiting.

Mycobacterium tuberculosis is a globally successful pathogen due to its ability to persist for long periods of time unrecognized by the human immune system. The panoply of genes that allows the organism to enter latency and then re-emerge later during endogenous reinfection are now being elucidated. Novel antimicrobials and vaccines will need to target these mycobacterial pathogenic mechanisms to suceed against tuberculosis.

Trends and determinants of immunisation coverage in India.

The 20th century has witnessed many important events in the control of infectious diseases that mostly affect children. In addition to the eradication of smallpox, the interruption of poliomyelitis transmission in many countries with a distinct possibility of its eradication by the turn of this century are some of the major achievements. Also, the rates of other vaccine preventable diseases such as measles, pertussis and diphtheria have gone down significantly. The discovery and use of vaccines have made it possible to save approximately 8 million deaths, annually. This is in addition to the r

New strategies against an old plague: genetically engineered tuberculosis vaccines.

The failure of BCG vaccination to control the global tuberculosis epidemic and the spread of multidrug resistance underline the need for a better vaccine. Recent advances in molecular microbiology, gene therapy, and, last but not least, immunobiology, provide a rational basis for the development of more efficient vaccines against tuberculosis. The complete sequencing of the M. tuberculosis genome marked a turning point in tuberculosis vaccine research. The advent of genetic vaccination with either naked DNA, live recombinant, or artificial vaccine vectors holds out great promise of more effici

Alteration of the calcium content in inner hair cells of the cochlea of the guinea pig after acute noise trauma with and without application of the organic calcium channel blocker diltiazem.

Calcium ions are known to be important to the process of signal transduction across the apical and basal sides of the inner hair cells. Calcium channel antagonists have been demonstrated by light microscopy to provide protection against acoustic trauma. To evaluate the protective effect of calcium channel blocker on the inner ear cells to noise exposure, the amount of the histochemical reaction products formed in the cytoplasm of the inner hair cells of the guinea pig after application of pyroantimonate was measured by an image processing system connected to an energy-filtering transmission el

Pediatric admission for vaccine preventable diseases: a 5-year survey from 1994 to 1998 in Aichi Prefecture.

Several infectious childhood diseases can be prevented by vaccination. A survey of hospital admissions for such diseases was conducted in Aichi Prefecture over 5 years beginning in 1994. A questionnaire was sent annually to hospitals with 100 or more beds to obtain information on pediatric patients requiring hospitalization for 10 vaccine preventable diseases. Information was obtained on 3,953 patients. Most admissions were for measles (49%), followed by mumps, chickenpox, pertussis, rubella, and tuberculosis. Over half of the patients were under 3 years old, with 20% aged under 12 months, 25%

[Policy program to minimize spread of infection. Prolonged cough may be a sign of tuberculosis].

In a worldwide epidemiological perspective, Sweden is well favoured with an annual tuberculosis incidence of approximately six cases per 100,000 of the population. Neither the impact of the HIV pandemic nor the occurrence of multiresistant strains of Mycobacterium tuberculosis has yet become a major problem in the care of tuberculosis patients in Sweden. Only a few per cent of HIV patients have developed tuberculosis, and during the period, 1991-94, only one per cent of M. tuberculosis isolates in Sweden were resistant to such antimycobacterials as isoniazid and rifampicin. However, the epidem

Health and safety in clinical laboratory practice in Ibadan, Nigeria.

The economic implications of safety precautions in laboratory practice make compliance difficult especially for laboratories in countries with poor economies. However, economic factors are not the only determinants of a safe work environment. Worker's perception of health and safety also influence compliance with safety guidelines. This study was conducted to determine the attitude and practice of laboratory workers with regard to safety. A structured questionnaire was administered to laboratory workers in 10 clinical laboratories. The questionnaire inquired about the use of protective equipme

Immunological and endocrinological characteristics of tuberculosis that provide opportunities for immunotherapeutic intervention.

Immunity to tuberculosis requires a T helper 1 (Th1) response which can be compromised by excessive release of inflammatory cytokines or Th2 activity. Environmental saprophytes can protect against tuberculosis by inducing Th1 recognition of the common antigens, or make mice more susceptible to tuberculosis than unimmunized controls by evoking a Th2 response. A mixed Th1 + Th2 response increases the local toxicity of tumour necrosis factor alpha (TNF-alpha). Some saprophytes are potent immunogens. A killed preparation of Mycobacterium vaccae can cause systemic activation of spontaneously Th1 cy

Some effects of the rising case load of adult HIV-related disease on a hospital in Nairobi.

Increasing numbers of HIV-infected adults in Africa need hospital care. It remains unclear what impact this has on health care services or on how hospitals respond. The aim of this study was to describe the effects of a rising case load of adult HIV-related disease by comparing results from a prospective cross-sectional study of acute adult medical admissions to a government hospital in Nairobi conducted in 1992 with results from a previous study done in 1988 and 1989 in the same hospital, using the same study design and protocol. Data on age, gender, number admitted, length of stay, HIV statu

Induction of a type 1 immune response to a recombinant antigen from Mycobacterium tuberculosis expressed in Mycobacterium vaccae.

A 19-kDa lipoprotein from Mycobacterium tuberculosis was expressed as a recombinant antigen in the nonpathogenic mycobacterial host strain M. vaccae. Immunization of mice with the recombinant M. vaccae resulted in induction of a strong type 1 immune response to the 19-kDa antigen, characterized by immunoglobulin G2a (IgG2a) antibodies and gamma interferon (IFN-gamma) production by splenocytes. Immunization with the same antigen in incomplete Freund's adjuvant induced a strong IgG1 response with only low levels of IFN-gamma. Subsequent intravenous and aerosol challenges of immunized mice with v

Antibacterial vaccines: impact of antigen handling and immune response.

The emerging awareness that diseases caused by bacterial pathogens cannot be vanquished by chemotherapy alone has recalled interest in the generation of novel vaccines. Vaccines have proven their efficacy for the bacterial pathogens that are controlled by antibodies. In contrast, satisfactory vaccines are not available for intracellular bacteria that are under the control of T lymphocytes. This review describes the T cell populations that must be activated by successful vaccines against intracellular bacteria and discusses parameters that need to be fulfilled by protective T cell antigens. Use of the bacille Calmette-Guérin vaccination for the prevention of tuberculosis: renewed interest in an old vaccine.

The reemergence of tuberculosis, including the impact of HIV infection and multidrug-resistant tuberculosis, have renewed interest in the bacille Calmette-Guérin (BCG) vaccine. During the past 7 decades, numerous studies have shown variable efficacy of BCG vaccination, ranging from 0% to 80%. The BCG vaccine is more likely to prevent disseminated forms of tuberculosis in children than pulmonary tuberculosis in adolescents or adults. Bacille Calmette-Guérin vaccination is recommended in asymptomatic children with or at risk for HIV infection, but it rarely may cause disseminated BCG infection a

DNA for genetic vaccination and therapy.

DNA coding for an antigen can be directly injected into muscle or skin and stimulate an immune response against the expressed antigen. The genes expressed can be derived from pathogens (e.g. viruses or bacteria), and can either code for surface molecules, which are often the basis for conventional peptide vaccines, or from the more genetically stable internal proteins. The DNA mimics a real infection in that the antigens are produced intracellularly where they are correctly folded and where they can be presented to the immune system so that cytotoxic T cells are stimulated as a defense mechani

Immunological requirements for a subunit vaccine against tuberculosis.

Tuberculosis remains one of the most important threats to world health. Current vaccination and prevention strategies are inadequate and there is an urgent need for a new vaccine. The current vaccine bacille Calmette-Guérin (BCG), is unable to protect against re-activation of disease in later life and its efficacy varies tremendously in different human populations. An ideal replacement would be a non-living subunit vaccine that could impart protective efficacy greater than BCG but without its drawbacks. Before such a goal is achieved, however, there are many parameters that need to be examined
