Immunoinformatics-based design of a multi-epitope mRNA vaccine candidate targeting the major human-infecting hepatitis E virus genotypes 1-4.

BACKGROUND: Hepatitis E virus (HEV) is one of the leading causes of acute and chronic hepatitis among humans across the globe and is linked to severe diseases in immunocompromised individuals and pregnant women. Despite its substantial global burden, only one vaccine is licensed (in China and Pakistan), limiting its global accessibility. Furthermore, no globally approved mRNA vaccine against HEV is currently available, highlighting the need to develop strategies for alternative vaccines. METHODS: In this study, the amino acid sequences of ORF2 capsid proteins representative of HEV genotypes 1

Identification of G-Quadruplexes in Hepatitis E Virus Negative-Sense Genomic RNA as Potent Antiviral Targets.

Hepatitis E virus (HEV) causes significant global disease burden with no approved targeted therapies, highlighting the urgent need for innovative treatment strategies. G-quadruplexes (G4s), noncanonical nucleic acid structures formed by guanine-rich sequences, have emerged as important regulators of viral replication. Here, we identified two potential G4 sequences within HEV negative-sense genomic RNA. Circular dichroism spectroscopy confirmed their stable, parallel G4 structures, with structural stability enhanced by the G4-binding ligand pyridostatin (PDS). Using an EGFP reporter system, we

Reduced HAV IgG Seropositivity Among Unvaccinated People Living with HIV: The Weak Shield.

People living with HIV (PLWH) represent a high-risk population for hepatitis A virus (HAV) infection, with exposure risk equal to or higher than that of the general population, particularly within adult risk networks. Anti-HAV immunoglobulin G (IgG) serves as a neutralizing antibody and is considered a key marker of protective immunity against HAV infection. However, the serologic profile of anti-HAV IgG and IgM among unvaccinated PLWH remains insufficiently characterized, especially in South China. A total of 1232 unvaccinated adults were enrolled in the study, including 800 PLWH and 432 HIV-

SARS-CoV-2 Neutralization Breadth Is Shaped by Variant Exposure and Immune History.

Despite widespread COVID-19 vaccination, emerging SARS-CoV-2 variants continue to evade neutralizing responses. To determine how exposure history influences neutralizing breadth against SARS-CoV-2, plasma collected from 46 participants in the Clinical, Virological and Immunological COVID-19 study cohort (CVIC) was analyzed. Participants were categorized into six groups reflecting distinct SARS-CoV-2 exposure profiles: individuals infected with either Wuhan-Hu-1 or Delta variants, infection-naïve individuals who received two or four doses of mRNA vaccine (including a bivalent BA.4/5 booster), p

The complex interplay between hepatitis D virus and the interferon response.

Hepatitis D virus (HDV) infection represents a major global public health challenge and is responsible for the most severe form of chronic viral hepatitis. Affecting an estimated 12 million individuals worldwide, chronic hepatitis D is associated with rapid progression to advanced liver disease, including cirrhosis and hepatocellular carcinoma. Despite its clinical burden, therapeutic options for HDV remain limited. In this context, pegylated interferon alpha (PEG-IFN-α) continues to be the most widely used treatment globally, particularly outside Europe where access to newly approved therapie

Role of Oncoviruses in Cancer Progression and Emerging Phytochemical-Based Therapies from Medicinal Plants.

Oncoviruses continues to be an unattended cause of the global cancer load as they cause about 12-15 % of human malignancies in the world. The oncogenic infections that become persistent, such as Human Papillomavirus (HPV), Epstein-Barr Virus (EBV), Hepatitis B and C viruses (HBV, HCV), Human T cell Leukemia Virus-1 (HTLV-1), and Kaposi Sarcoma-associated Herpesvirus (KSHV), are associated with malignant transformation by continuous destabilization of host cell cycle regulation, immune surveillance, and metabolic homeostasis. These viruses abuse critical oncogenic signaling networks like PI3K/A

Update on the molecular and cellular biology of hepatitis E virus and therapeutic opportunities.

Hepatitis E virus (HEV) is an emerging zoonotic pathogen and the leading cause of acute viral hepatitis worldwide. Nevertheless, HEV remains largely underestimated and poorly controlled. HEV infects a broad range of host species, transmits through diverse routes and can cause chronic infections in immunocompromised individuals and severe hepatitis in pregnant women. Currently, there is no HEV-specific antiviral treatment available and the only licensed vaccine is restricted to a few countries. Taken together, this highlights its significance as a global health treat and underscores the urgent
