Educational Materials for Health-Related Discrete Choice Experiments: A Theory-Driven Approach.

BACKGROUND: To make informed choices in health-related discrete choice experiments (DCEs), respondents need to understand the choice scenarios, alternatives, attributes and attribute levels presented to them. Respondent populations are often highly diverse in knowledge and experience, leading to varying informational needs that make it challenging to design educational materials that sufficiently inform all respondents without disengaging others. At present, however, structured guidance on developing DCE educational materials is lacking. Drawing on the instructional design literature, this pap

Matrix Metalloproteinase-8 Deficiency Alters the Balance Between Viral Clearance and Immunopathology in H1N1 Infection.

Matrix metalloproteinase-8 (MMP-8) regulates inflammatory responses, but its contributions to the pathogenesis of influenza A viral (IAV) disease are not known. Plasma MMP-8 levels were increased 30-fold in subjects infected with H1N1 or seasonal IAV and were inversely associated with the PaO2/FiO2 ratio, suggesting potential value as a candidate marker of lung injury severity that requires validation in larger cohorts. MMP-8 levels were elevated in the lungs of H1N1-infected wild-type (WT) mice and localized to airway epithelial cells, macrophages, and polymorphonuclear neutrophils (PMNs). H1

H5 influenza virus mRNA-lipid nanoparticle (LNP) vaccination elicits adaptive immune responses in Holstein calves.

Highly pathogenic avian influenza (HPAI) clade 2.3.4.4b H5N1 circulated widely in lactating dairy cows in the United States following the initial detection in 2024. Due to the critical need for intervention strategies for this outbreak, we evaluated antibody and cellular immune responses of a clade 2.3.4.4b H5 mRNA-LNP vaccine in calves. We found that the H5 mRNA-LNP vaccine induced a robust antibody and CD8+ T cell-mediated immune response and conferred protection against clade 2.3.4.4b H5N1 infection.IMPORTANCEIt is important to develop vaccines that limit clade 2.3.4.4b H5N1 infection in ca

A multivalent mRNA-lipid nanoparticle vaccine containing eight hemagglutinin antigens elicited broad neutralizing antibody responses and protected against influenza A virus challenge in swine.

The diversity within H1 and H3 subtype influenza A viruses (IAVs) in swine prevents effective vaccine control approaches with inactivated whole-virus vaccines. We addressed the challenge of controlling co-circulating hemagglutinin (HA) clades of swine IAV with the development of a multivalent messenger RNA (mRNA)-lipid nanoparticle (LNP) vaccine expressing eight HA proteins to maximize genetic coverage. We applied a computational approach to select eight HA genes that represented 95% of the observed IAVs detected in the United States between 2022 and 2025. Piglets were vaccinated and boosted i

A digitally immune-optimized influenza vaccine broadly neutralizes swine and human H1N1 influenza viruses and protects from heterologous challenge.

UNLABELLED: Influenza A virus (IAV) zoonotic transmission and continuous evolution in multiple host species heighten the risk of emerging novel strains at the human-animal interface. Digitally immune-optimized hemagglutinin (HA), neuraminidase (NA), and matrix-2 (M2) vaccine antigens were engineered and combined as a single DNA expression construct (DVX-H1N1) to maximize immune responses to human seasonal, pandemic, and zoonotic H1N1 IAVs. Immunization of mice with DVX antigens induced broad neutralizing antibody responses to human- and swine-origin H1N1 isolates. The DVX-H1N1 candidate was th

Cross-clade protection against human H5N1 isolates by a clade 2.3.4.4b HA mRNA vaccine in mice.

The recent spread of clade 2.3.4.4b H5N1 high pathogenicity avian influenza (HPAI) viruses into mammals, despite limited human infections to date, has raised serious public health concerns and highlights the need for rapidly deployable vaccines, such as mRNA-based platforms, to strengthen pandemic preparedness. Here, we evaluated the effectiveness of an mRNA vaccine encoding the HA of NIID-002, a WHO-recommended H5 candidate vaccine virus (CVV) derived from A/Ezo red fox/Hokkaido/1/2022 (clade 2.3.4.4b) against A/Texas/37/2024 (TX37; clade 2.3.4.4b) and A/Cambodia/2311257/2023 (Cam23; clade 2.

The Role of Enoxaparin in Influenza Virus Infections and its Therapeutic Implications.

Frequent emergence of respiratory viruses with pandemic potential, like severe acute respiratory syndrome coronavirus 2 or influenza, underscores the need for broad-spectrum prophylaxis. Existing vaccines show reduced efficacy against newly emerged variants, and the ongoing risk of new outbreaks highlights the importance of alternative strategies to prevent infection and viral transmission. As respiratory viruses primarily enter through the nose, formulations targeting the nasal epithelium are attractive candidates to neutralize pathogens and thus prevent or minimize infection. Bridging Genomics and Clinical Medicine: RSVrecon Enhances RSV Surveillance With Automated Genotyping and Clinically Important Mutation Reporting.

BACKGROUND: Respiratory Syncytial Virus (RSV) causes significant respiratory infections, particularly in young children and elderly adults. Genetic variations in the fusion (F) protein can reduce the efficacy of vaccination and monoclonal antibody treatments, emphasizing the need for genomic surveillance of this virus. MOTIVATION: Current pipelines for RSV genome assembly focus on sequence reconstruction but often lack features for detecting genotypes, clinically relevant mutations, or presenting results in formats that are suitable for clinical researchers. RESULTS: We introduce RSVrecon, a

Maternal-Fetal Impact of Respiratory Viral Infections: Immune Mechanisms and Clinical Outcomes.

Pregnancy is a unique immunological state characterized by physiological adaptations that support fetal growth and development. These immune changes, however, can leave pregnant women more vulnerable to severe outcomes from infections, including those caused by respiratory viruses. Respiratory viral infection during pregnancy can increase maternal morbidity and the likelihood of developing long-lasting neurodevelopmental consequences in the offspring, including autism spectrum disorder, schizophrenia, and other behavioral conditions, particularly in male offspring. Some respiratory viruses can

Immune history confers antibody- and T cell-dependent cross-protection against highly pathogenic avian influenza H5N1 viruses.

The outbreak of highly pathogenic avian H5 influenza (HPAI) clade 2.3.4.4b in cattle has spread across the United States. Mice with pre-existing immunity to H1N1 virus or with a live-attenuated influenza vaccine showed protection against a lethal bovine-derived HPAI H5N1 viral challenge. Notably, ferrets with mixed immunity also demonstrated protection against a feline-derived H5N1 virus, independent of cross-reactive neutralization titers, but antibodies to whole virus were observed. To investigate protective factors, we conducted T cell epitope mapping using published H1N1 viral sequences an

Dual impact of squalene-adjuvanted influenza vaccine on immunity and glucose homeostasis in obese mice.

Obesity is a known risk factor for impaired immune responses and reduced vaccine efficacy. Individuals with obesity-related metabolic disorders face heightened vulnerability to influenza, underscoring the urgent need to optimize vaccine strategies for this population. We investigated the immunological and metabolic outcomes of different influenza vaccine formulations in a diet-induced obese (DIO) mouse model. Specifically, we compared the effects of high-dose versus squalene-adjuvanted vaccines on humoral immunity, protection against viral challenge, and glucose metabolism. High-dose vaccinati

Factors influencing influenza vaccine hesitancy among cancer patients: a multicenter cross-sectional study based on the Health Belief Model.

BACKGROUND: Cancer patients often become high-risk groups for influenza due to their immunosuppressed state, and the risk of severe illness after infection significantly increases. However, this group generally has influenza vaccine hesitancy, resulting in an unsatisfactory vaccination rate. This study aimed to investigate the influenza vaccination rate of cancer patients in China, systematically assessed the current situation of vaccine hesitancy, and explored its key influencing factors. METHODS: From April to June 2025, a cross-sectional study design was adopted to conduct a questionnaire s

A Naked Lyophilized mRNA Vaccine Against Seasonal Influenza, Administered by Jet Injection, Provides a Robust Response in Immunized Mice.

Background: Seasonal influenza remains a significant public health problem, and the constant antigenic drift of viruses requires regular vaccine updates. mRNA vaccines offer a promising platform for the development of new, effective influenza vaccines. Administration of the naked mRNA vaccine using a needle-free jet injection system further enhances its safety, reduces cost, and eliminates the need for lipid nanoparticles, which are traditionally used for mRNA delivery. Lyophilization of naked mRNA allows for long-term storage at +4 °C. Methods: We designed and produced an mRNA vaccine against

Broadly neutralizing monoclonal antibodies against influenza A viruses: current insights and future directions.

Monoclonal antibodies (mAbs) have become attractive tools for both the treatment and prevention of influenza A viruses due to their ability to target several viral components, which confers broad therapeutic potential. Advances in biotechnology, such as hybridoma technology, phage display technology, B cell immortalization, and artificial intelligence (Al)-driven antibody design, have significantly accelerated the development of effective mAbs. Clinical trials have shown that mAbs can improve clinical outcomes particularly in high-risk and immunocompromised populations by lowering viral loads

A review of targeting microRNAs as potential therapeutic strategies against respiratory viruses: Current insights and future directions.

Respiratory viruses such as influenza viruses, respiratory syncytial virus (RSV), and coronaviruses continue to impose a global health burden due to their high transmissibility and limited antiviral options. MicroRNAs (miRNAs) have emerged as critical regulators of host pathogen interactions by modulating innate immunity, inflammatory signaling, and viral replication. This review focuses on respiratory RNA and DNA viruses that primarily infect the airways, including influenza viruses, RSV, human metapneumovirus, rhinoviruses, adenoviruses, and SARS-CoV-2. Several miRNAs, including miR-155 and

Natural Alkaloids as Antiviral Agents Against RNA Viruses: A Comprehensive and Mechanistic Review.

RNA viruses pose a persistent global threat due to their high mutation rates, zoonotic potential, and rapid adaptability. Emergence events have risen steadily, as demonstrated by major outbreaks caused by Influenza A, Ebola, Zika, and Chikungunya viruses, followed by the coronavirus epidemics of Severe Acute Respiratory Syndrome coronavirus (SARS-CoV-1) and Middle East Respiratory Syndrome Coronavirus (MERS-CoV) and culminating in the COVID-19 pandemic. These characteristics frequently compromise the durability of existing vaccines and antiviral therapies, highlighting the urgent need for new
