Avian influenza viruses (AIV) represent a major zoonotic threat to global public health and agriculture due to their high mutation rates, antigenic drift, and potential for interspecies transmission. This study addresses the urgent need for innovative, broad-spectrum vaccines that can overcome the limitations of traditional approaches, such as slow production and strain-specific protection, by designing and evaluating a novel multi-epitope protein-based vaccine targeting conserved regions of the AIV hemagglutinin (HA) protein through advanced immunoinformatics tools. Using an in silico approac

Influenza A virus (IAV) in swine is a significant economic concern, and there is a critical need to improve vaccine efficacy. Commercial and experimental vaccine platforms are effective against homologous infection but may not reliably provide protection against drifted or heterologous viruses. Live attenuated influenza A virus (LAIV) vaccines induce mucosal antibody and localized cellular immune responses that may provide partial protection from drifted IAV. However, limited data exist on the induction of mucosal antibody and cellular immune responses and heterologous protection induced by RN

The heat-labile enterotoxins (LTs) of Escherichia coli structurally belong to the AB5 toxin, which consists of an A subunit and a pentameric B subunit. LTs have been extensively reported for their potent mucosal adjuvant activities. The B subunit of type II LT (LT-IIb-B5) recognizes the GD1a ganglioside, which is associated with the internalization of the toxic A subunit and meanwhile involved in the recognition of the Toll-like receptor (TLR-2/1) and downstream NF-κB activation. In developing LT-IIb-B5 as a safe adjuvant, we intend to prevent concerns derived from the intrinsic toxicity of LT

The influenza virus poses a significant global health challenge, causing approximately 500,000 deaths annually. Its ability to evade antiviral treatments and vaccine-induced immunity underscores the need for novel therapeutic approaches. Our study identifies cis-aconitate (cis-aco), a mitochondria-derived metabolite, as a potent dual-action agent against influenza, independently of its metabolic derivative, itaconate. Cis-aco impairs viral polymerase activity, resulting in decreased viral mRNA expression and protein synthesis, as observed for the influenza A/Scotland/20/74 (H3N2) strain. This

Avian influenza virus (AIV) poses a persistent threat to global poultry production and public health. Long-term immunization programs have established a foundational immune barrier, significantly mitigating the risk of highly pathogenic avian influenza outbreaks. However, the high mutability of AIV, complex biosafety requirements, and the accelerating scale of poultry production underscore the need for enhanced mucosal protection. Mucosal immunity represents a critical defense against respiratory virus invasion in poultry, rapidly mobilizing local antibodies, cellular immune responses, and inn

BackgroundDementia is a leading cause of mortality worldwide, with its prevalence projected to rise sharply in the coming decades. Established risk factors include advanced age, hypertension, diabetes, obesity, smoking, excessive alcohol consumption, physical inactivity, social isolation, and depression. Increasing evidence suggests that vaccination-by preventing infections that may trigger or accelerate neurodegeneration-could play a protective role against dementia, though findings remain inconsistent.ObjectiveTo systematically review observational evidence on the association between influen

Avian influenza (AI), particularly highly pathogenic avian influenza (HPAI), represents a serious and growing threat to global poultry production, international trade, and human health security. Control of AI is complicated by the high evolutionary rate of influenza A viruses, which drives antigenic diversity and ongoing emergence of novel strains. Effective surveillance and disease management therefore depend on timely and accurate diagnostics. While conventional methods-including virus isolation, reverse transcription-quantitative polymerase chain reaction (RT-qPCR), and enzyme-linked immuno

An H3N2 variant, named subclade K, continues to circulate widely during the 2025-2026 influenza season. This virus possesses a hemagglutinin (HA) protein that has eleven substitutions relative to the HA of the Northern Hemisphere 2025-2026 H3N2 vaccine strain. Many of these substitutions are in epitopes in well-characterized HA antigenic sites. Despite this, interim vaccine effectiveness studies indicate that the 2025-2026 influenza vaccine provides moderate protection against H3N2 subclade K infection. We previously reported that many individuals who received the 2025-2026 influenza vaccine p

In China, low pathogenic avian influenza (LPAI) H3N8 virus is widespread among chickens and has recently caused three zoonotic infections, with the last one in 2023 being fatal. Here we evaluated the relative pandemic risk of this 2023 zoonotic H3N8 influenza virus, utilizing our previously published decision tree. Serological analysis indicated that a large proportion of the human population does not have any cross-neutralizing antibodies against this H3N8 strain. LPAI H3N8 displayed a dual affinity for α2-3 and α2-6 sialic acids and replicated efficiently in human bronchial epithelial cells.

Clade 2.3.4.4b H5N1 highly pathogenic avian influenza (HPAI) virus is circulating widely in US dairy cattle with significant implications for milk production and cow health. Due to the critical need for intervention strategies for continuing outbreaks of bovine HPAI, a baculovirus vectored inactivated HPAI vaccine expressing the 2.3.4.4b H5 protein (Avian Influenza-H5) was developed utilizing a licensed prescription platform which has been tested and proven safe in multiple species. The immunogenicity of recombinant Avian Influenza-H5 vaccine was tested in 5-month-old calves. The subcutaneousl

Monoclonal antibodies (mAbs) are an emerging class of therapeutics for the prevention and treatment of viral infections. Recent advances in mRNA/lipid nanoparticle (LNP) technology provide a promising new modality for the production of mAbs in vivo, potentially bypassing the need for recombinant manufacturing of mAb proteins. In this study, we compared traditional infusion of protein-based neutralizing mAbs targeting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) or influenza viruses to mRNA/LNP-based production of mAbs in treated mice. High serum concentrations of mAbs were achi

BACKGROUND: Influenza remains a significant public health burden. Although vaccination is the most effective preventive strategy, evidence on influenza vaccine effectiveness (VE) in Korea remains fragmented.
METHODS: A systematic review and meta-analysis were conducted on real-world Korean influenza VE studies published from January 2016 to October 2025. Literature research was performed in PubMed, Embase, and Web of Science. Eligible studies evaluated influenza VE using real-world data sources. Pooled VE estimates were calculated using a random-effects model and stratified by age group and v

Influenza A virus (IAV) remains a major global threat, highlighting the need for host-targeted antiviral strategies. While some probiotics offer prophylactic protection, their therapeutic potential post-infection is poorly understood. Here, we investigated human-derived Lactobacillus brevis ZG2488 for its antiviral potential against IAV. Strikingly, a more pronounced reduction in viral titer was observed when live bacteria were administered therapeutically post-infection, compared to preventive pretreatment. Transcriptomic analysis suggested that the therapeutic effect of viable bacteria was a

Immune defenses decline with age, increasing susceptibility to influenza. Vaccination remains the most effective strategy to prevent severe disease and death, but its efficacy is reduced in older adults, particularly against influenza A(H3N2). The mechanisms underlying this age-related decline in vaccine-specific antibody responses remain unclear. We investigated the magnitude and quality of influenza-specific T-cell responses following quadrivalent inactivated influenza vaccination in adults aged under (n = 100) or over (n = 120) 65 years. Frequencies of T cells specific to influenza A (H1N1,

The absence of an approved influenza vaccine for young infants, combined with the annual requirement for updates, underscores the urgent need for a universal vaccine that is effective in newborns. The H1ssF vaccine, an influenza hemagglutinin stem-bearing nanoparticle, generates broadly protective responses in adult animal models and humans. Here, we use a nonhuman primate model to explore the ability of H1ssF, adjuvanted with R848 and AddaVax, to induce a protective response in the altered immune setting of newborns. We find that the vaccine induces strong stem-specific IgG responses that are

Vaccination is commonly used to minimize disease impacts on birds and promote sustainable poultry production. Vaccination against highly pathogenic avian influenza (HPAI) differs because HPAI is classified as a foreign animal disease in the US and disease control falls under federal jurisdiction. Vaccination for HPAI could negatively affect the nation's ability to export poultry and poultry products, representing the largest barrier to implementation. However, the continued burden of global HPAI cases requires a reevaluation of the cost of ongoing outbreaks and the need for new mitigation stra

Despite existing vaccines, influenza remains a significant public health burden, highlighting the need for vaccine strategies with improved safety and efficacy. Here we develop an autophagy-targeting influenza. A virus as a live attenuated vaccine by harnessing the host cell's autophagy machinery to manipulate viral protein degradation. The virus is generated by introducing a conditionally removable autophagosome-targeting motif in the viral genome, which enables viral attenuation through autophagy-mediated degradation of the tagged viral protein in conventional cells while permitting efficien

RNA viruses evolve rapidly, enabling them to evade host immunity and antiviral therapies and complicating durable diagnostics strategies. There is a pressing need for approaches that provide broad-spectrum viral suppression and detection. We designed four Cas13d crRNAs targeting a conserved 26-nucleotide sequence in coronavirus (CoV) nsp12. Their antiviral efficacy was evaluated in vitro against the target sequences of all seven human CoVs, showing potent activity across the tested samples. The same crRNAs were adapted for a Cas13d-based specific high-sensitivity enzymatic reporter unlocking (

Respiratory viruses, such as influenza A virus (IAV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), remain major global health threats, and the emergence of drug-resistant variants underscores the urgent need for host-targeted antiviral strategies. Prohibitins (PHBs), mitochondrial scaffold proteins involved in diverse cellular processes, are host factors exploited by multiple viruses. Here, we investigated the antiviral potential of two PHB-binding triazine melanogenin derivatives, Mel56 and Mel6, against IAV and SARS-CoV-2. Mel56, but not Mel6, exhibited potent anti-IAV ac

Background/Objectives: Seasonal influenza vaccination is pivotal for protecting high-risk populations, including those with chronic health conditions (CHCs), from severe complications and outcomes. This study aims to describe the sociodemographic characteristics of unvaccinated children (6 months-17 years old) with CHCs, the reasons why vaccine hesitant parents chose not to vaccinate their children, and the factors associated with seasonal influenza non-vaccination among these children. Methods: This cross-sectional analysis used a sub-sample from the 2023 Childhood COVID-19 Immunization Cover
