Vaccine Effectiveness Estimates Against Influenza A(H3N2)-Associated Hospitalized Severe Acute Respiratory Infections in Beijing, China, 2025/26 Influenza Season.

BACKGROUND: Data on influenza vaccine effectiveness (VE) against hospitalized severe acute respiratory infection (SARI), particularly in Asia, remain limited for the 2025/26 Northern Hemisphere influenza season. This study aimed to evaluate real-world VE against A(H3N2)-associated SARI hospitalization and provide timely, locally relevant evidence to inform seasonal influenza vaccination policy. METHODS: A test-negative design was used to estimate VE against influenza A(H3N2)-associated SARI hospitalization in Beijing, China, from 10 November 2025 to 18 January 2026. VE was estimated by compar

Seasonal influenza mRNA vaccine induces stronger innate and comparable or better adaptive responses than licensed inactivated vaccines.

Seasonal vaccination is the most important strategy for mitigating the large disease burden caused by influenza virus. Licensed inactivated influenza virus vaccines rely on slow, egg-based production, underscoring the need for faster, more effective platforms that elicit robust immune responses. Here, we compare the immunogenicity of an unmodified mRNA-lipid nanoparticle vaccine encoding influenza hemagglutinin (HA) from A/Michigan/45/2015 (H1N1)pdm09, A/Singapore/INFIMH-16-0019/2016 (H3N2), B/Phuket/3073/2013 and B/Colorado/06/2017 with two approved, strain-matched, inactivated vaccines (Vaxi

Evaluation of influenza vaccine effectiveness among older adults in Jiaxing, China, 2022-2025: A test negative design-based study.

Influenza imposes a substantial burden on older adults, underscoring the need for annual evaluation of vaccine effectiveness (VE). Using a test-negative case-control design, this study assessed VE among patients aged ≥60 y with acute respiratory symptoms in Jiaxing, China, across three influenza seasons (2022-2025). Among 3,214 participants, vaccination provided moderate overall protection against laboratory-confirmed influenza, with adjusted VE of 42.5% (95% CI: 31.2-51.9). Protection was highest in the 2024-2025 season (47.2%) and against influenza A(H1N1) (42.4%). Notably, subgroup analyses

Mechanistic insights into flavonoid-mediated regulation of human influenza virus replication.

Influenza A virus (IAV) remains a major global health concern due to its rapid mutation rate, antigenic variability, and the emergence of resistance to frontline antivirals such as oseltamivir and zanamivir. Despite available vaccines and therapeutics, their limited efficacy against emerging strains underscores the urgent need for new antiviral approaches. Therefore, plant-derived bioactive compounds, particularly flavonoids, have gained attention for their ability to target both viral and host pathways, yet their mechanisms of action remain incompletely understood. This review synthesises cur

'Veni, Vidi, Vaccini': consensus-based vaccination pathway implementation in a paediatric tertiary hospital in Tuscany, Italy.

Fragile paediatric patients are at higher risk of vaccine-preventable diseases, yet coverage remains suboptimal. Hospitalisation offers an opportunity to review and update immunisation, but fragmented practices create inequities, with access depending more on hospital context than clinical need. This study aimed to design, implement, and evaluate an in-hospital vaccination pathway for these patients. A mixed-methods approach was undertaken at Meyer Children's Hospital IRCCS, Florence, Italy. Baseline assessment included nine focus groups and an online questionnaire with healthcare professional

Research Progress on Avian Influenza Virus and Autophagy: A Review.

Avian influenza virus (AIV), a zoonotic pathogen capable of cross-species transmission, poses a significant global health threat due to its rapid evolutionary adaptation. This review consolidates evidence from the past decade on AIV-autophagy interactions, emphasizing mechanistic insights and therapeutic potential. Research indicates that various AIV strains can trigger autophagosome formation via viral components, although the completeness of autophagic flux is not fully understood. These virus-host interactions are notably influenced by viral genotypes (e.g., H5N1 vs. H9N2) and host species

The role of immunosenescence and chronic medical conditions on the immune response to respiratory syncytial virus in older individuals.

INTRODUCTION: Respiratory syncytial virus (RSV) is an important respiratory pathogen among older adults and those with chronic conditions. METHODS: Two systematic literature reviews identified the impacts of chronic conditions on RSV infection outcomes in adults, and how immunological changes alter RSV infection susceptibility in older individuals. Searches using MEDLINE/Embase identified relevant papers published between 1 January 1990, and 2 August 2023. RESULTS: Of 4,699 records, 212 were selected. Inclusion criteria were based on relevant comorbidities, outcomes, and study methodologies.

Protective effects of influenza B neuraminidase antibodies against symptomatic influenza virus infection.

BACKGROUND: Influenza vaccines are designed to induce hemagglutinin-inhibiting (HAI) antibodies rather than neuraminidase-inhibiting (NAI) antibodies; therefore, NAI antibodies' protective effects need further investigation. NAI antibodies' protective effects, independent of HAI antibodies, have been demonstrated for influenza A viruses, but not influenza B viruses (IBV). METHODS: Employing H6NB reassortant virus targets, we evaluated NAI antibodies' protective effects against IBV in sera from a prospective influenza vaccine effectiveness study among healthcare personnel (HCP) in Israel. Emerging Strategies for Antitumor Immunotherapy and Antiviral Defense Through the cGAS-STING Pathway.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a central regulator of innate immunity and plays a critical role in inducing pro-inflammatory cytokines and type I interferons (IFN-I). This pathway has emerged as a promising target for cancer immunotherapy and antiviral treatments. Despite its promise, the clinical translation of STING agonists is hindered by several challenges, including structural instability, high production costs, and inefficient delivery systems. These barriers underscore the urgent need for further research and innovation to optimize STI

A nanobody against haemagglutinin of H9N2 avian influenza virus shows efficacy in vitro and in vivo.

H9N2 subtype avian influenza virus (AIV) facilitates viral adaptative evolution and cross-species transmission, result in inflicting severe economic losses on the poultry industry and posing significant public health threats. Currently, conventional vaccine-based prophylactic strategies exhibit certain limitations, highlighting an urgent need for the development of novel prevention and control measures. In this study, using phage display technology, we screened 37 potential binding sequences from the PBMCs of an alpaca immunized with the H9N2 virus and its HA protein. AlphaFold3 further predic

RELMα establishes a permissive environment for influenza virus infection through direct effects on lung epithelial cells.

Beyond a physical barrier, the lung epithelium provides an essential line of defense against infectious pathogens, such as influenza A viruses (IAVs), by producing antimicrobial factors and triggering innate and adaptive immunity. On the other hand, IAV infects lung epithelial cells and manipulates their function to establish a non-protective, permissive environment. Identification of epithelial factors and pathways that can be targeted to restore optimal immunity would provide new therapeutic options against IAV. Here, we demonstrate an unexpected non-immune function for the cytokine RELMɑ in

Arming Inactivated Enveloped Virus Vaccines with the GGTA1 Gene: A Potent Method for Amplification of Viral Vaccines Effectiveness and Protection Against Variants.

This review describes a novel method for increasing the effectiveness of inactivated enveloped whole-virus vaccines by targeting them for extensive uptake by antigen-presenting cells (APCs). Several inactivated whole-virus vaccines with dense glycan shields display suboptimal effectiveness because the multiple carbohydrate chains (glycans) on the virus mask immunogenic peptides and surround the virus with a negative electrostatic charge that decreases uptake by APCs. It is postulated that engineering such vaccinating viruses to present the carbohydrate antigen "α-gal epitope" on the glycan shi

Advances in the Development of a Universal Influenza Vaccine.

Influenza remains a persistent global health threat, accounting for an estimated one billion infections and 350,000-600,000 deaths worldwide annually. Beyond substantial mortality and morbidity, influenza imposes major socio-economic costs through healthcare utilisation and productivity losses, amounting to tens of billions of dollars each year. Current seasonal vaccination strategies are constrained by rapid antigenic evolution, which necessitates frequent vaccine reformulation and contributes to suboptimal vaccine efficacy across seasons. These drawbacks highlight the need for a universal in

Haemophilus influenzae Epiglottitis: A Rare Disease Not to Be Forgotten.

Although pediatric epiglottitis has become rare following the widespread use of the Haemophilus influenzae type b (Hib) vaccine, breakthrough infections still occur, whilst other H. influenzae serotypes continue to emerge. We present the case of a 10-year-old child, fully immunized against Hib, who developed epiglottitis. Blood cultures confirmed H. influenzae infection, and further microbiological characterization identified a non-typable strain. This case highlights the growing clinical relevance of H. influenzae strains not covered by the Hib vaccine and reinforces the need to maintain a hi

Maternal Immunization.

Vaccines administered to women during pregnancy can provide protection against serious infectious diseases for the mother, the child, or both. Maternal immunization boosts the concentration of maternal antibodies that can be transferred across the placenta to directly protect children too young to be immunized. In addition, indirect protection through prevention of maternal infection and breast-milk antibodies can be achieved through maternal immunization. In general, inactivated vaccines are considered safe for pregnant women and fetuses, whereas live attenuated vaccines are avoided due to th

Integrating prior knowledge inference with computational multi-omics analysis to reveal host antiviral networks of natural compounds against influenza A virus.

INTRODUCTION: Influenza A virus (IAV) remains one of the major global health threats, and a rapid emerging resistance to direct acting antivirals underscores the need for host directed therapies. METHODS: Here, we integrated prior knowledge inference with a multilevel phenotypic screening and computational multi-omics analysis to identify natural candidate chemical compounds from Traditional Chinese Medicine (TCM)-derived libraries. Twenty compounds enriched for links to antiviral signaling were tested via a GFP-IAV reporter assay, followed by experimental validation using replication kinetic
