Expression Profiling of a Recombinant Inbred Line Panel Identifies the dod-19 Gene as a Regulator of Ethanol Response Behaviors in C. elegans.

BACKGROUND: Variation in gene expression is hypothesized to be a major source of phenotypic variation underlying the liability for alcohol misuse. Caenorhabditis elegans (C. elegans) has proven to be a valuable model in the identification of genes that modify behavioral responses to alcohol. Genetic variation in wild strains represents allelic differences that are tolerated by natural selection pressures and, therefore, are more likely to model the allelic differences that are present in humans compared with laboratory-generated mutations. METHODS: We examined the influence of natural alleles

Extracorporeal photopheresis for the treatment of steroid-refractory immune related adverse events from immune checkpoint inhibitor therapy: a multi-center phase Ib/II clinical trial.

Severe immune-related adverse events (irAE) in cancer patients receiving immune checkpoint inhibitors (ICI) require treatment interruption and immunosuppression. Based on prior preclinical data, in this prospective, single arm, non-randomized phase-Ib/II clinical trial we test the activity of Extracorporeal Photopheresis (ECP) against steroid-refractory irAEs in 30 patients (EudraCT-No.:2021-002073-26). The primary endpoint is the evaluation of the safety of ECP treatment for patients with ICI-induced irAEs. The study meets its pre-specified primary endpoint of less than 50% of patients develo

Mechanistic pathways of cosmetic ingredient toxicity: a critical review of in vitro and in vivo evidence.

Every day, billions of people apply cosmetic products containing dozens of bioactive compounds whose combined toxicological profile has never been formally evaluated. Human biomonitoring confirms systemic deposition of these compounds in blood, adipose tissue, placenta, and breast milk worldwide. This scoping review, following PRISMA-ScR, synthesizes in vitro and in vivo mechanistic toxicology evidence for six major cosmetic ingredient categories-preservatives, UV filters, engineered nanoparticles, fragrances, surfactants, and colorants-and maps findings onto established adverse outcome pathwa

Lymph node-targeted, mKRAS-specific amphiphile vaccine in pancreatic cancer: the randomized phase 2 AMPLIFY-7P trial.

Camupepimut (ELI-002 7P) drives mutated KRAS (mKRAS)-specific T cell responses that correlated with disease-free survival (DFS) and overall survival (OS) in patients with mKRAS pancreatic adenocarcinoma (PDAC) in the phase 1 AMPLIFY-201 trial. In the phase 2 AMPLIFY-7P trial, patients with stage I-III KRAS-mutated PDAC without recurrence within 6 months of standard therapy were randomized 2:1 to camupepimut (n = 96) or standard-of-care/observation (n = 48). Stratification balanced nodal status. The primary DFS endpoint was not met (median 23.8 versus 21.2 months; HR 0.85, P = 0.5329). However,

Microbiome interactions in renal cell carcinoma: evidence, challenges and opportunities.

Renal cell carcinoma (RCC) is a heterogeneous malignancy with limited predictive biomarkers to guide treatment. Emerging evidence suggests that the microbiome might influence systemic immunity, therapeutic response and clinical outcomes. In metastatic RCC, the gut microbiome has been linked to the efficacy of immune checkpoint inhibitors, whereas the existence of a tumour microbiome remains a topic of debate. Circulating microbial DNA in blood has been proposed as a minimally invasive window into systemic immune status, offering a potential tool for monitoring treatment response, but its role

Transcriptional and phenotypic alterations of regulatory and effector T cells in type 1 diabetes mellitus.

Type 1 diabetes mellitus (T1DM) is characterized by the destruction of pancreatic β-cells, in which the dysfunction of regulatory T cells (Tregs) contributes to impaired immune tolerance. However, Treg heterogeneity remains incompletely understood, which limits the effectiveness of current therapies. Here, we have integrated transcriptome, flow cytometry, glycemic correlation, and cross-model validation to characterize the dysregulation of Tregs and effector T cells (Teffs) in a streptozotocin (STZ)-induced T1DM mouse model. Tregs exhibited a proliferative, effector-like, unstable lineage, and

Ionomics powered single-cell mapping reveals targetable iron dysregulation in endometriosis associated macrophages.

BACKGROUND: Endometriosis affects approximately 10% of women of reproductive age worldwide. Although elemental components of refluxed menstrual blood have been implicated in disease progression, comprehensive ionomic profiling of the endometriotic microenvironment remains limited. Characterizing the localized elemental landscape may generate new hypotheses regarding disease mechanisms. METHODS: We employed inductively coupled plasma mass spectrometry (ICP-MS) to profile 63 elements in endometriotic cyst fluid. Single-cell RNA sequencing (scRNA-seq) was performed to explore cellular subpopulat

Endothelial cell-specific TGM2 deficiency promotes progression of temporal lobe epilepsy by disrupting S1P metabolism and intercellular communication.

BACKGROUND: Temporal lobe epilepsy (TLE) is a common and medically refractory neurological disorder with a complex pathogenesis. Blood-brain barrier (BBB) dysfunction is a key pathogenic factor. As a multifunctional protein, TGM2 has been shown to participate in vascular homeostasis, metabolic pathway regulation, and immune-inflammatory responses. This study aimed to systematically investigate the expression pattern, core functions, and molecular regulatory mechanisms of TGM2 in TLE, and to clarify its clinical value. METHODS: Temporal cortex tissue and plasma samples from drug-resistant TLE

First-trimester aggregate index of systemic inflammation and adverse pregnancy outcomes in hospitalized women with threatened miscarriage: a single-center retrospective cohort study.

BACKGROUND: Complete blood count-derived inflammatory indices have been associated with miscarriage, but the incremental value of the aggregate index of systemic inflammation (AISI) beyond established indices such as the systemic immune-inflammation index (SII) remains uncertain. We examined the association and prognostic performance of first-trimester AISI in hospitalized women with threatened miscarriage. METHODS: This single-center retrospective cohort study included 312 women hospitalized with threatened miscarriage between January 2020 and December 2022. The primary outcome was a composi

Precise immunomodulation by mesenchymal stem cells mitigates hepatitis B virus-specific CD8⁺ T cell-mediated acute liver injury without compromising viral clearance.

Hepatitis B virus-related acute-on-chronic liver failure (HBV-ACLF) is associated with a high mortality rate. Although mesenchymal stem cell (MSC) therapy markedly improves liver function and survival in patients with HBV-ACLF, it remains unclear whether the immunomodulatory effects of MSCs suppress antiviral CD8⁺ T cell function and trigger HBV reactivation. Here, we developed a novel in vitro HBV-specific CD8⁺ T cell-mediated acute liver injury organoid model, which was co-cultured with either menstrual blood-derived MSCs (MenSCs) or umbilical cord-derived MSCs (UC-MSCs). We found that both

Engineering human endogenous retrovirus-K for improving immunogenicity and broad cancer targeting.

BACKGROUND: HERV-K (HML-2) is the most studied transcriptionally active endogenous retrovirus in human cancers, yet immune tolerance and incomplete mechanistic understanding have impeded its clinical use as an immunotherapy target. We hypothesized that rational engineering of the HERV-K envelope putative immunosuppressive domain (ISD) could improve immunogenicity. METHODS: We performed a mutational screen across the ISD region, evaluated NF-κB activation and then analyzed antigen expression in human monocyte-derived dendritic cells (moDCs) using flow cytometry, RNA-seq, and TEM. Human PBMCs w

A 28-color panel for classical and non-classical T lymphocytes in decidua and PBMC in rhesus macaques.

This 28-color panel was developed to identify classical and non-classical T lymphocytes in decidual leukocytes and peripheral blood mononuclear cells (PBMC) of pregnant rhesus macaques ( Table 1 ). By profiling these T lymphocytes, we can investigate how maternal immunity balances tolerance to fetal antigens with protection against vertically transmitted pathogens. The selected markers define memory populations and characterize tissue residency, activation, proliferation, cytotoxicity, trafficking, and exhaustion status. This panel also delineates B lymphocytes and NK cells to confirm expected

Immunometabolic synaptopathy: linking metabolic syndrome to epileptogenesis through trained immunity and parvalbumin interneuron vulnerability.

BACKGROUND: Metabolic syndrome (MetS) and epilepsy are clinically associated, but how chronic peripheral metabolic stress might influence seizure susceptibility remains unclear. The potential links among CNS-interface dysfunction, glial responses, and inhibitory-circuit vulnerability require further clarification. PROPOSED FRAMEWORK: We propose immunometabolic synaptopathy as a hypothesis-driven framework linking chronic metabolic dysfunction to impaired inhibitory-circuit resilience. MetS-associated metabolic and inflammatory signals may alter blood-brain barrier function, CNS-border immune

IFN-γ, TNF-α, IL-10, IL-17 and IL-22 secretion capacities and activation profile of T cells in psoriatic arthritis patients under disease-modifying antirheumatic drug therapy.

INTRODUCTION: While biologic disease-modifying antirheumatic drugs (bDMARDs) have transformed management, achieving complete remission remains a challenge due to complex immune networks in Psoriatic arthritis (PsA). This study aimed to characterize the functional modulation of circulating T-helper (Th) and cytotoxic T (Tc) cells in PsA patients following conventional synthetic (csDMARD) and biologic therapy. METHODS: Peripheral blood mononuclear cells were collected from bDMARD-naïve PsA patients at baseline and six months after initiating csDMARD (primarily methotrexate, n=8) or bDMARD (prim

TCR clonotyping of paired blood and tonsillar T-follicular cells reveals the mixed origin of CD38high Tfrs.

Circulating T follicular regulatory (cTfr) cells are measured as blood biomarkers after vaccination, in autoimmune disease (e.g., lupus, rheumatoid arthritis) and cancer immunotherapy, yet whether they originate from natural Tregs (yielding nTfr) or germinal center T follicular helper (GC Tfh) cells (yielding iTfr) remains unknown. This distinction is critical for interpreting cTfr expansion as Treg-derived regulation versus Tfh-derived negative feedback. To characterize tonsillar follicular T cell heterogeneity and resolve the origins of cTfr cells we applied single-cell multiomics and bulk T

Genomic insights into multidrug and extensively drug-resistant Klebsiella pneumoniae from bloodstream infections at three Zambian tertiary hospitals.

BACKGROUND: Klebsiella pneumoniae is a leading cause of hospital-associated infections and a major contributor to antimicrobial resistance (AMR) worldwide. Despite its importance and prioritisation in Zambia's National Action Plan on AMR, genomic surveillance information in sub-Saharan Africa remains limited. This study investigated the population structure, determinants of AMR, and plasmid diversity of K. pneumoniae isolates from three tertiary hospitals in Zambia. METHODS: Thirty archived K. pneumoniae blood isolates collected between December 2023 and May 2025 from University Teaching Hosp

Autochthonous Plasmodium falciparum malaria in a high-altitude, non-endemic region of Ethiopia: a case report.

BACKGROUND: Malaria transmission in Ethiopia is traditionally limited to altitudes below 2000 m. Debark Town, situated at 2850 m, is a high-altitude highland fringe area characterized by a cold climate and has historically been considered a non-endemic zone. This report describes a rare, locally acquired case of Plasmodium falciparum in this extreme environment. CASE PRESENTATION: A 15-year-old Ethiopian female of Amhara ethnicity resident in Debark Town presented with a 5-day history of high-grade fever (39.5 °C), headache, and vomiting. Significantly, she reported no history of travel to kn

Knowledge of cervical cancer and uptake of HPV vaccine among in-school adolescent girls in Kwara State, Nigeria.

BACKGROUND: Cervical cancer ranks as the second most common malignancy among Nigerian women, contributing heavily to preventable mortality, particularly in low-resource settings. HPV infection is the primary cause, and vaccination provides an effective prevention strategy. However, coverage in Nigeria remains critically low. Understanding knowledge and barriers among adolescent girls is imperative for designing interventions. This study explored these determinants among in-school girls in Kwara State. METHODS: This cross-sectional study surveyed 628 adolescent girls aged 9-14 years across rur

Infection prevention and control procedures in nursing homes in the piedmont region of Italy: results from two regional surveys (2021-2024).

BACKGROUND: Infection prevention and control (IPC) in long-term care facilities (LTCFs) is essential to reduce the burden of healthcare-associated infections (HAI) and the spread of antimicrobial resistance (AMR). This paper describes the development of standardized IPC procedures through a stakeholder-informed approach, and integrates results from two regional surveys conducted in Piedmont (first survey 2021-2022; second survey 2024) which assessed the reported availability of written procedure documents in nursing homes and, subsequently, the uptake and perceived utility of the developed sta
