Curcumin Attenuates Escherichia coli-Induced Enteritis and Macrophage Inflammation in Broilers.

Escherichia coli (E. coli)-induced enteritis impairs intestinal barrier function, immune homeostasis, and growth performance in broilers. This study investigated whether dietary curcumin alleviates E. coli-induced intestinal injury by modulating macrophage-mediated inflammatory responses through the p38 MAPK signaling pathway. Broilers were fed either a basal or curcumin-supplemented diet and challenged with E. coli. Growth performance, intestinal inflammatory cytokines, morphology, barrier-related gene expression, goblet cell density, and ileal macrophage responses were evaluated. Lipopolysac

Integrated bioinformatics analyses for GSDMB in carcinogenesis and progression of bladder cancer.

BACKGROUND: Emerging evidence suggests that pyroptosis influences the development of various diseases. Gasdermin B (GSDMB), an intracellular protein that executes pyroptosis, has recently attracted attention for its potential role in tumor biology. However, its specific function in bladder cancer (BLCA) remains unclear. Therefore, this study aimed to investigate the potential role of GSDMB in the carcinogenesis and prognosis of BLCA patients. METHODS: Mendelian randomization (MR) studies were conducted to examine relationships between the expression of GSDMB and BLCA with expression quantitat

Pathological networks and multi-target interventions in sepsis-associated acute lung injury: from pathogen-host interactions to gut-lung axis regulation.

OBJECTIVE: Sepsis is a life-threatening organ dysfunction resulting from a dysregulated host response to infection, accounting for an estimated 11 million deaths annually worldwide. Among its most prevalent and severe complications is acute respiratory distress syndrome (ARDS). Traditional research has focused on isolated pathways such as inflammation and oxidative stress; however, the results of clinical trials for related targeted drugs have been less than ideal. The core reason is that sepsis-associated acute lung injury (SA-ALI) is not driven by a single, independent pathway, but rather by

Genome-Wide DNA Methylation Profiling Reveals Stage- and Severity-Associated Epigenetic Alterations in Children with Posterior Urethral Valve-Associated Chronic Kidney Disease.

BACKGROUND: Chronic kidney disease (CKD) is an important cause of morbidity in children, particularly in those with congenital anomalies of the kidney and urinary tract. Although epigenetic alterations have been implicated in CKD, stage-associated genome-wide DNA methylation changes remain poorly characterized in posterior urethral valves (PUV). This study aimed to characterize DNA methylation patterns across CKD stages in children with PUV and identify potential methylation markers associated with CKD severity. METHODS: Genome-wide DNA methylation profiling was performed on peripheral blood

Single-cell transcriptomics deciphers cancer-associated fibroblast heterogeneity and immune regulatory mechanisms in the bladder cancer tumor microenvironment.

BACKGROUND: Bladder cancer (BLCA) is a common malignancy, with muscle-invasive bladder cancer (MIBC) associated with a 5-year survival rate below 50%. Cancer-associated fibroblasts (CAFs) are heterogeneous stromal components of the tumor microenvironment (TME) that contribute to tumor progression, therapy resistance, and immune evasion. However, the molecular basis of CAF diversity and immune regulation in BLCA remains incompletely understood. METHODS: We analyzed single-cell transcriptomic data from GEO dataset GSE135337 using dimensionality reduction, unsupervised clustering, differential e

From cells to pregnancy: Applications of placental organoids in translational research.

The development of trophoblast organoids (TOs) has revolutionized placental research by providing proliferative, three-dimensional systems recapitulating the key aspects of placental architecture and function. These models have become invaluable tools for studying trophoblast differentiation and early placental development. Importantly, TOs also offer new opportunities for investigating mechanisms underlying pregnancy-related disorders such as preeclampsia or fetal growth restriction, and host-pathogen interactions, advancing our understanding of how infections impact early pregnancy. However,

Non-HLA Antibodies in Kidney Transplantation: Mediators of Allograft Injury or Markers of Dysregulated Humoral Immunity?

Antibody-mediated rejection (AMR) remains a major cause of kidney allograft injury and loss. Although donor-specific antibodies against human leukocyte antigens (HLA-DSAs) are central to humoral alloimmunity, they do not explain all cases of microvascular inflammation or graft dysfunction. This review critically evaluates non-HLA antibodies as potential mediators of allograft injury and markers of dysregulated humoral immunity by integrating functional, experimental, clinicopathological, and clinical evidence. Receptor-targeting antibodies, especially those against the angiotensin II type 1 re

Novel Liposomal Adjuvants Enhance the Immune Response to Recombinant Staphylococcus aureus Antigens in Dairy Heifers.

The development of effective adjuvant systems remains a critical challenge in veterinary vaccine design, particularly for infectious diseases causing significant economic losses in dairy cattle. This study evaluated the immunological profile induced by a mixture of recombinant antigens, formulated with cationic liposomes containing different combinations of gemini lipopeptide AG2-C16, bovine-specific CpG oligodeoxynucleotide, and S. cerevisiae mannans, in dairy heifers. Thirty-five dairy heifers aged 8-10 months were randomly assigned to seven groups and immunized with recombinant Staphylococc

Respiratory Syncytial Virus Vaccination Strongly Induces RSVpreF-specific IgG and Neutralizing Activity in Immunocompromised Individuals.

BACKGROUND: Protein-based respiratory syncytial virus (RSV) prefusion F (RSVpreF) vaccines show promising immunogenicity in immunocompromised individuals. We have recently shown that RSV-specific CD4 T-cell responses and IgG antibodies were significantly induced, but data on functional humoral responses as well as associations between cellular and humoral immunological endpoints remain limited. We therefore extended our previous studies and performed a head-to-head assessment of vaccine-specific RSVpreF IgG and neutralizing antibody activity across different immunocompromised populations, incl

Preface to the Special Issue: "Microbiota and Gut-Brain Axis".

The microbiota-gut-brain axis has emerged as a central framework for understanding how microbial communities influence nervous system development, function, and disease. This Special Issue highlights three interconnected themes: microbial metabolites and neurochemistry, developmental and neuroimmune mechanisms, and translation to neurological and psychiatric disease. The articles in the collection examine how microbial metabolites modulate neuroinflammatory and neurochemical pathways, how stress, development, immune signaling, and gut health shape gut-brain communication across the lifespan, a

Prenatal NO₂ exposure and cardiac developmental dysfunction: CD4⁺ T cell imbalance drives cardiac fibrosis through placenta-heart axis immune transmission.

Epidemiological studies show maternal nitrogen dioxide (NO2) exposure associates with congenital heart disease (CHD) risk, but subtypes and mechanisms unclear. Here, we first conducted a systematic review and meta-analysis, which confirmed that prenatal NO₂ exposure significantly increased the risk of coarctation of the aorta (COA) (OR = 1.15, 95% CI: 1.04-1.26). Using a mouse model of prenatal NO₂ exposure, we demonstrated that prenatal NO2 exposure adversely affected fetal cardiac structure and function at embryonic day 18.5, characterized by ventricular dilation, myocardial wall thinning, s

An engineered bacteria-chitosan hybrid robot for lactate depletion and synergistic STING activation to potentiate bladder cancer immunotherapy.

The treatment of bladder cancer faces multiple challenges, including poor tumor tissue penetration, low drug delivery efficiency, and an immunosuppressive microenvironment. To overcome these bottlenecks, this study constructed a novel bacteria-chitosan hybrid robot. The system utilizes electrostatic interactions to combine engineered Escherichia coli expressing lactate oxidase with positively charged chitosan nanoparticles, with the latter co-loaded with a chemotherapeutic drug Doxorubicin (DOX) and the DNA repair inhibitor olaparib. Following intravesical instillation, the engineered bacteria

Ancestry-Dependent Immunologic and Prognostic Effects Characterize the Prostate Cancer Urinary Proteome.

Urine is an attractive biomarker analyte for non-invasive longitudinal monitoring of health and disease, particularly for diseases of the genitourinary tract, like prostate and bladder cancer. The composition of an individual's urine reflects both genetic and lifestyle characteristics that differ across geographies and populations, like diet, hydration and other socio-economic factors. While men of African ancestry have elevated prostate cancer risk, it is unclear to what extent this influences urinary biomarkers. We therefore quantified the urinary proteomes of 329 localized prostate cancer p

Prenatal PM2.5 exposure drives epigenetic reprogramming of fetal macrophages linked to atopic dermatitis.

Prenatal environmental exposures are increasingly recognized as contributors to atopic dermatitis (AD), yet the underlying mechanisms remain unclear. Fine particulate matter (PM2.5), a complex mixture of airborne pollutants, has been associated with elevated risk of allergic diseases, particularly during early development. Here we show that first-trimester PM2.5 exposure is associated with an increased risk of AD in early childhood and induces epigenetic alteration in the placenta. Integrative multi-omics analyses, including single-cell approaches, reveal hypomethylation of FCER1G in fetal mac

Metabolomic Profiling of Medicinal Plant-Based Soft Porridge and Evaluation of Cytotoxicity.

Background/Objectives: Traditional medicinal plants play a crucial role in infants' healthcare in underdeveloped countries and communities where access to modern medicine is limited. In Limpopo, the Vhavenda tribe has a custom of feeding babies as young as a day old with tshiunza, a very light and warm, soft porridge prepared in medicinal plant extracts, believed to help treat various ailments and boost the immune and digestive systems. However, reports of negative side effects, including vomiting, haematuria, renal inflammation and hospitalisations, highlight safety concerns of these traditio

Exosomes-mediated molecular mechanisms underlying the transformation of ovarian tissue into a functional placental analogue in black rockfish ( Sebastes schlegelii).

Viviparity has independently evolved multiple times in teleosts, leading to diverse modes of maternal nutrient provisioning. In black rockfish ( Sebastes schlegelii), embryos gain dry weight during gestation, supported by a placental connection that facilitates maternal-fetal nutrient transfer. Although prior studies have outlined the morphology and evolutionary convergence of the black rockfish placental analogue, its full architecture and underlying molecular mechanisms remain unresolved. Here, we show that the maternal component of the placental analogue, derived from ovarian follicular tis

Extracellular vesicle-based therapeutic strategies for spinal tumors and associated nerve damage: advances, challenges, and future directions.

The clinical management of spinal tumors presenting with acute spinal cord compression with consequent neural injury poses two concurrent challenges: achieving oncological control and preserving or restoring neurological function. Surgical decompression and chemoradiotherapy can relieve mechanical compression and reduce tumor burden, but they do not directly restore the damaged neural microenvironment. In recent years, extracellular vesicles (EVs) have emerged as promising cell-free nanotherapeutic candidates. Preclinical evidence suggests that they can cross the blood-spinal cord barrier and
