PURPOSE: Advances in automatic speech recognition (ASR) and natural language processing have the potential to meaningfully impact language assessment. Despite the increasing availability of these technologies, little is known about their performance in routine assessment tasks such as language sample analysis (LSA), which is commonly used in the identification of developmental language disorder (DLD). Here, we present an enzyme-free workflow for isolating murine adult neural stem cells (aNSCs) using tissue grinder- and dissociator-based mechanical dissociation (TIGR-MD). We describe dissection of the dentate gyrus and subventricular zone, mechanical tissue dissociation, cell recovery, and establishment of primary neurosphere cultures. We further provide procedures for neurosphere maintenance, passaging, and validation. TIGR-MD avoids enzymatic exposure during initial tissue dissociation and supports standardized establishment of primary murine aNSC cultures.

The new sulfolipid-like natural product tritonicysteinolic acid (TTC) was isolated from an extract of the marine bacterium Tritonibacter and exhibited inhibitory activity against PGAM5, a protein phosphatase that localizes in the inner mitochondrial membrane. The full stereochemistry of TTC was elucidated through total synthesis, thereby enabling the synthesis of various derivatives. Structure-activity relationship (SAR) studies were conducted, which revealed that the derivatives exhibited higher inhibitory activity against PGAM5 phosphatase relative to the parent natural product.

PURPOSE: Teenagers and young adults (TYA) with cancer have distinct developmental, psychosocial, and clinical needs that require age-appropriate care. Healthcare policy in the United Kingdom advocates care in centralised specialist age-appropriate TYA Principal Treatment Centres (PTCs).
