The endocannabinoid system (ECS) is a ubiquitous lipid signaling network that regulates tissue homeostasis. Increasing evidence indicates that the ECS undergoes profound remodeling during pancreatic injury and may influence key pathological processes underlying acute pancreatitis (AP) and chronic pancreatitis (CP). Interest in this field has grown in parallel with increasing cannabis use among individuals with pancreatic disorders and the urgent need for alternatives to opioid-based pain management. Components of the ECS, including cannabinoid receptors, the endogenous ligands, and enzymes tha

Aging is a major risk factor in the development of type 2 diabetes mellitus (T2DM). With age, pancreatic β-cells undergo functional and cell identity changes that can compromise insulin secretion, thereby contributing to impaired glucose tolerance and T2DM. However, glucose homeostasis also depends on pancreatic α-cells. Excessive plasma glucagon levels, along with impaired regulation of α-cell secretion, are involved in the pathophysiology of T2DM. Despite their importance, the effects of aging on α-cells have been overlooked. In the present study, we analyzed pancreatic α-cells in aged mice

Transplantation of allogeneic islets of Langerhans, which include the insulin-producing β cells of the endocrine pancreas, holds curative potential for type 1 diabetes (T1D). However, protecting the allograft from the host immune system has long been a challenge impeding wider use of this therapy. Inducing mixed hematopoietic chimerism via allogeneic hematopoietic stem cell transplantation (HSCT) can achieve long-lasting donor-specific immune tolerance, but the toxicities of conventional HSCT conditioning agents limit the use of this approach. In this issue of the JCI, Bhagchandani et al. have

AIMS/HYPOTHESIS: MicroRNAs are potential predictors and mediators of metabolic disease. Cystic fibrosis-related diabetes (CFRD) is the most common comorbidity in cystic fibrosis (CF). Here we aimed to investigate serum microRNAs in individuals with CF and differing glucose tolerance status. Specifically, we hypothesised that the circulating microRNA profile varies depending on glucose tolerance status and can change rapidly in response to a glucose challenge in individuals with CF.
METHODS: We studied a cohort of 93 adult Danish participants with CF from four glucose tolerance categories: nor

Serine/threonine phosphatase 1 (PP1) and phosphatase 2A (PP2A) play important roles in mediating cellular signaling in different tissues to different stimuli, including in protein synthesis, growth, cell cycle regulation, and secretion. However, their roles in various pancreatic exocrine functions, such as pancreatic acinar fluid/electrolyte secretion, is still unclear. Therefore, in the present study, we examined the ability of vasoactive intestinal peptide (VIP) and secretin, which stimulate cAMP generation in pancreatic acini, to activate serine/threonine phosphatase 1 (PP1) and phosphatase

AIMS/HYPOTHESIS: Regenerating endogenous pancreatic beta cells is a potentially curative yet currently elusive strategy for diabetes therapy. Mimicking the microenvironment of the developing pancreas and leveraging vascular signals that support pancreatic endocrinogenesis may promote beta cell regeneration. We aimed to investigate whether recovery from experimental hypovascularisation of the endocrine pancreas could trigger mouse beta cell proliferation.
METHODS: A doxycycline (DOX)-inducible transgenic mouse model was used to induce conditional intra-islet hypovascularisation. In this model,

Aldosterone, a stress-related hormone, may contribute to pancreatic dysfunction. Its role in pancreatic physiology, inflammation, and fibrosis remains poorly understood. This study aimed to evaluate the effects of aldosterone on pancreatic histomorphology and islet cellularity under chronic stress conditions, and to determine whether eplerenone, a selective aldosterone receptor antagonist, can counteract them. Twenty-four young male Wistar rats were randomly divided into four groups (n = 6): control, chronic unpredictable mild stress (CUMS), control + vehicle (control + Veh), and CUMS + eplere

Immune cells in the pancreas are known to participate in organ development. However, the resident pancreatic immune system has yet to be fully defined. Immune cells also play a role in pathology and are implicated in diseases such as diabetes induced by intrauterine growth restriction (IUGR). We hypothesized that the resident immune system continues to develop during the neonatal period and is disrupted by IUGR. Using single-cell RNAseq and flow cytometry we identified many immune cell populations in the near-term fetus (at embryologic day 22) and neonatal (postnatal day 1, 7, &14) islets, non

Obesity is a chronic metabolic disorder characterized by systemic and hypothalamic inflammation that impairs neuronal signaling and disrupts the fine-tuning of energy balance regulation. Leptin resistance, a hallmark of obesity, arises from this inflammatory state and contributes to increased food intake and reduced energy expenditure (EE). Fibroblast growth factor 19 (FGF19), a gut-derived hormone with beneficial metabolic effects in peripheral tissues, also acts centrally to reduce body weight. However, the mechanisms underlying its hypothalamic action remain unclear. In this study, we inves

BACKGROUND: Hyperglucagonemia is a hallmark of diabetes mellitus, resulting from the dysregulation of glucagon secretion by pancreatic alpha-cells. Although glucose sensing and insulin signaling are well-established regulatory processes, the pathways that govern glucagon secretion remain unclear. Recent evidences suggest that insulin-degrading enzyme (IDE) regulates glucagon secretion via an unknown pathway. METHODS: Using IDE-immunoprecipitation proteomic data as a basis, we aimed to ascertain the molecular mechanisms downstream of IDE in the alpha-TC cell model. Knock-down studies of relevan

Type 1 diabetes (T1D) is a progressive autoimmune condition that culminates in loss of insulin-producing beta cells. Pancreatic histopathology provides essential insights into disease initiation/progression yet an integrated perspective onto in situ pathogenic processes is lacking. Here, we combined multiplexed immunostaining, high-magnification whole-slide imaging, digital pathology, and semi-automated image analyses to interrogate pancreatic tail and head sections across T1D stages, including at-risk and at-onset cases. Deconvolution of architectural features, endocrine cell composition, imm

The equine pancreas remains understudied, particularly in the context of endocrine disease. This study aimed to characterize regional islet distribution and composition in the normal equine pancreas and investigate how hyperinsulinemia (HI) and pituitary pars intermedia dysfunction (PPID) influence pancreatic islet morphology and hormone expression. In the first experiment, pancreas samples from eight healthy horses were collected and analyzed across three anatomical locations: left lobe, body, and right lobe. The left lobe exhibited a greater relative islet area and perimeter compared to the

Background: Three-dimensional (3D) cell cultures are increasingly recognized as effective models for studying diseases and developing cell therapies. In the endocrine pancreas field, organoids/spheroids derived from human islet cells enable advances in diabetes research, drug screening, and tissue engineering. While various 3D culture methods exist, approaches such as magnetic bead-assisted aggregation remain underexplored for endocrine pancreatic cells. Additionally, the use of biological scaffolds, especially those derived from decellularized pancreatic extracellular matrix, provides a biomi

Pancreatic endocrine-exocrine crosstalk plays a key role in normal physiology and disease and can be altered by host metabolic states, such as obesity. Classically, endocrine islet beta (β) cell secretion of insulin is thought to promote the development of obesity-associated pancreatic adenocarcinoma (PDAC), an exocrine cell-derived tumor. Here, we show that β cell expression of the peptide hormone cholecystokinin (CCK) is necessary and sufficient for obesity-associated PDAC progression in mice and that CCK expression - rather than insulin - correlates strongly with enhanced tumorigenesis. PURPOSE: Pancreatic neuroendocrine tumors (PNET) are rare malignancies of the endocrine pancreas with diverse clinical outcomes. Whereas some PNETs are indolent, others are aggressive and metastasize quickly. However, clinically relevant molecular stratification for PNET to predict outcomes and guide therapeutic decision-making is limited. Thus, there is an urgent need to understand the molecular heterogeneity of PNETs to refine prognostication and discover novel therapeutic vulnerabilities.
EXPERIMENTAL DESIGN: We performed single-nucleus RNA sequencing on untreated, resected primary, and me

OBJECTIVES: Pancreatic neuroendocrine tumors (pNETs) are uncommon neoplasms, and contemporary US population-level mortality evaluations for malignant endocrine neoplasms of the pancreas are limited. This study assessed long-term mortality trends and forecast future trajectories for malignant neoplasms of the endocrine pancreas using US death-certificate data.
METHODS: In this study, CDC WONDER Multiple Cause of Death public-use death-certificate data from the USA were analyzed for 1999 to 2023. Deaths were identified using ICD-10 code C25.4 when listed anywhere on the death certificate. Age-a

Epidemiological studies consistently report associations between circulating concentrations of persistent organic pollutants and increased type 2 diabetes risk. Measures of pollutant concentrations in pancreas are limited; given the role of the endocrine pancreas in diabetes pathogenesis, this is an important gap in the literature. Additionally, no studies have correlated pollutant concentrations with direct measures of human beta cell function. We measure concentrations from 3 pollutant classes-dioxins/furans, polychlorinated biphenyls, and organochlorine pesticides-in pancreas and peripancre

AIMS: Ageing leads to a gradual decline in the structure and function of many bodily organs, including the endocrine pancreas. The current study examines the impact of ageing on pancreatic islet morphology, and especially the role of islet cell plasticity in this process.
METHODS: Transgenic Ins1Cre/+;Rosa26-eYFP mice, with islet beta-cell tracing capabilities, were employed at 12 and 52 weeks of age, and islet morphology, islet cell turnover as well as changes in beta-cell identity and plasticity assessed.
RESULTS: Ageing was associated with elevated body weight and circulating glucose leve
