A Novel Cross-Kingdom Regulatory Mode of Thermostable miR166 from Taraxacum mongolicum Inhibits HepG2 Cell Proliferation in Association with Downregulation of the Candidate Target STAP2.

Taraxacum mongolicum contains various bioactive components. This study investigated the potential of thermostable microRNAs (miRNAs) derived from this plant as cross-kingdom regulators and evaluated their anti-proliferative effects on HepG2 cells. High-throughput sequencing was used to screen thermostable miRNAs and predict their target genes. Enrichment and target gene expression in gavaged mice were analyzed to assess cross-kingdom regulatory potential. Cellular assays evaluated anti-proliferative activity. miR166 was identified as a thermostable miRNA in T. mongolicum, and bioinformatics in

Blood-transforming coating enables hemostasis and vascularized bone re-generation via bioactive gelation.

Conventional bone repair materials often treat blood as an obstacle through physical isolation or passive hemostasis, overlooking its physiological role in the regenerative cascade. Here, we propose a "Blood-Transforming Coating" concept and engineer a functionalized microsphere system (GM/TCP@PEG). The NHS ester-coated microspheres rapidly crosslink with amino-rich blood components to form a robust hydrogel while covalently anchoring to tissue surfaces, transforming blood into a synergistic component that enhances material function. Structural characterization confirmed the core-shell archite

Silybum marianum peptide-rich fraction alleviates acute liver injury by inhibiting TRAF3/NIK/NF-κB and is associated with changes in gut microbiota composition.

Acute liver injury (ALI) is a severe and prevalent condition that can progress to irreversible liver damage and failure. This study aimed to investigate the prophylactic hepatoprotective potential of Silybum marianum peptides (SMP) in ALI and elucidate its underlying mechanisms. Using a carbon tetrachloride (CCl4)-induced ALI mouse model, we evaluated the dose-dependent effects of SMP. Additionally, we explored the modulatory effects of SMP on the NF-κB pathway, gut microbiota composition, and metabolite profiles. The role of SMP in the TRAF3/NIK/NF-κB signaling axis was further validated in a

Toward harmonized PROMISE PSMA scoring: a simple approach for correcting 18F-PSMA-1007 liver uptake based on 68Ga-PSMA-11 and 18F-DCFPyL reference values.

BACKGROUND: The PROMISE framework standardizes PSMA PET interpretation using reference organs, particularly the liver. For hepatobiliary-excreted tracers (e.g., 18F-PSMA-1007), the spleen was proposed as an alternative reference, though without quantitative validation. We evaluated PSMA-ligand uptake patterns among 18F-PSMA-1007, 68Ga-PSMA-11, and 18F-DCFPyL to assess the validity of spleen-based referencing and develop a simple approach that may harmonize 18F-PSMA-1007 liver uptake with that of renally-excreted tracers. RESULTS: 331 consecutive PSMA PET-CT studies (n = 118/95/118 for 68Ga-PS

Cyanine-Based Near Infrared-II Aggregation-Caused-Quenching Probes for Particokinetic Analysis.

Aggregation-caused quenching (ACQ) probes provide an integrity-reporting strategy for tracking nanocarriers by emitting strong fluorescence when molecularly dispersed within the hydrophobic core of intact carriers and becoming quenched after release into aqueous biological environments. However, previously reported near-infrared-II (NIR-II) ACQ probes suffer from limited effective emission in the NIR-II window and potential fluorescence reillumination after probe release, which may compromise nanoparticle quantification and thereby particokinetic analysis. Here, we developed a series of Flav7-

Lipid nanoparticles for precision nucleic acid delivery: linking cellular engineering to organ-specific targeting.

Lipid nanoparticles (LNPs) represent clinically validated non-viral platforms for nucleic acid delivery, yet expanding their application toward precision gene therapy demands more than efficient particle formulation or tissue accumulation. Functional delivery requires navigating a sequential biological cascade: establishing biological access to target tissues, undergoing cell-specific uptake, escaping endosomal entrapment, and releasing intact cargo into the cytosol. Because each stage presents a distinct rate-limiting barrier, success at one step does not guarantee downstream therapeutic acti

Sophorolipid-mediated interfacial modification alters digestion-driven structural evolution of hexosomes and improves baicalin bioaccessibility.

Interfacial properties play an important role in determining the digestion behavior and structural evolution of lipid-based nanocarriers, yet their influence on the gastrointestinal fate of flavonoid glycoside remains incompletely understood. In this study, sophorolipid (SL)-modified inverse hexagonal (HII) hexosomes were developed to investigate how interfacial modification influences the digestion behavior and bioaccessibility of baicalin (BA), a poorly water-soluble flavonoid glycoside selected as a model compound due to its limited aqueous solubility and bioaccessibility. Severe Plasmodium falciparum infection with multi-organ involvement in an asplenic patient: a case report.

Plasmodium falciparum is the leading cause of severe malaria. The spleen plays a central role in the clearance of the infection in conjunction with antimalarial treatment. Splenectomized patients lack sufficient clearance functions. We report a case of severe P. falciparum infection with multi-organ involvement acquired in Uganda in a splenectomized patient with a persistently high parasitemia level despite appropriate antimalarial treatment. The case emphasizes the challenges associated with severe malaria in asplenic patients.

Selective Systemic Responses During Aeromonas salmonicida Subsp. Salmonicida Infection in Northern Snakehead (Channa argus).

Northern snakehead (Channa argus) is a bimodal air-breathing fish in which erythrocyte-associated parameters remain relatively stable during physiological challenges such as aerial exposure. However, whether this characteristic is maintained during systemic infection remains unclear. In this study, temporal host responses were characterized following experimental Aeromonas salmonicida subsp. salmonicida infection by integrating haematological, leukocyte morphological, biochemical, and multi-tissue transcriptional analyses. Most erythrocyte-associated indices, including RBC, HGB, HCT, MCV, MCH,

Co-Infection of Aeromonas veronii and Citrobacter braakii in Yellow Catfish (Tachysurus fulvidraco).

Recently, a disease outbreak was observed in farmed yellow catfish (Tachysurus fulvidraco) in Tianjin, China. Naturally infected fish exhibited surface haemorrhages, abdominal distension, anal redness, and hemorrhagic ascites. Histopathological examination revealed systemic lesions in the stomach, intestine, liver, and spleen. Two Gram-negative bacterial isolates, designated PFL and PFS, were isolated from the liver and spleen of diseased fish, respectively. The isolates were identified as Aeromonas veronii and Citrobacter braakii by morphological and biochemical characterisation combined with

New Drugs in Myelofibrosis: Incremental Progress in Symptom Management Without Disease Modification.

Janus kinase inhibitors are the cornerstone of treatment for myelofibrosis, but have limited disease-modifying activity. Combinations of ruxolitinib with pelabresib, navitoclax, or selinexor improve spleen responses, although benefits for symptoms and anemia are less consistent, and toxicity may increase. Such approaches may optimize splenomegaly before transplantation. Emerging CALR-directed therapies, particularly INCA033989, offer potential for selective suppression of the malignant clone while improving spleen, anemia, and symptom outcomes. Anemia-directed agents targeting TGF-β/BMP-SMAD s

Molecular characterizing and expression analysis of MCSF and its receptor in largemouth bass (Micropterus salmoides) and spotted sea bass (Lateolabrax maculatus).

Macrophage colony-stimulating factor (MCSF) is a key hematopoietic cytokine that regulates the proliferation, differentiation, and survival of mononuclear phagocytes through its receptor, MCSFR. In this study, we cloned and characterized the full-length cDNAs encoding MCSF and MCSFR from two commercially important perciform fish, largemouth bass (Micropterus salmoides) and spotted sea bass (Lateolabrax maculatus). The largemouth bass MCSF (MsMCSF) cDNA contains a 792-bp open reading frame (ORF) encoding 263 amino acids, whereas the spotted sea bass MCSF (LmMCSF) cDNA has an 813-bp ORF encoding

Inhibitory effects of transfer factor against avian reovirus infection in chicks.

Avian reovirus (ARV) infection causes viral arthritis, growth retardation, and immunosuppression in chickens. Current immunoprophylaxis, which relies mainly on vaccination, offers limited protection. This study evaluated the efficacy of transfer factor (TF) against ARV infection. One-day-old specific-pathogen-free (SPF) chicks were randomly divided into 4 groups (n = 35 each): a prophylaxis group (TF administered at 3 to 5 days of age), a treatment group (TF administered at 9 to 11 days of age), an infected group, and a blank control group. All chicks except the blank controls were infected wi

FLASH effect on the liver and spleen of mice-a transcriptional perspective.

BACKGROUND: FLASH radiotherapy (FLASH-RT), delivered at an ultra-high dose rate (typically ≥ 40 Gy/s), has been shown to reduce normal tissue toxicity compared with conventional dose-rate radiotherapy (CONV-RT). However, the transcriptomic mechanisms underlying FLASH-mediated protection remain incompletely understood. PURPOSE: This study aimed to compare the biological effects of FLASH and CONV irradiation on mouse liver and spleen tissues and to identify molecular pathways associated with the FLASH effect from a transcriptional perspective. METHODS: C57BL/6J male mice received a single 12 G
