Changes in the apoptotic index, along with gene and protein expression levels, in splenomegaly due to portal vein stenosis in rats.

BACKGROUND: The spleen is an important peripheral immune organ, and splenomegaly (SM) is a common complication of portal hypertension. However, the regulatory mechanisms underlying splenic apoptosis remain poorly understood. MATERIALS AND METHODS: A portal vein stenosis-induced SM model was established in Sprague-Dawley rats, using a sham-operated (SO) group as the control. Apoptotic changes in splenic tissue cells were evaluated using the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, immunohistochemistry staining, quantitative real-time polymerase chain reaction

Dimensionality reduction for partial-volume correction in quantitative SPECT.

OBJECTIVE: The aim of this paper is to develop a method for region-based post-reconstruction partial-volume correction (PVC) of SPECT images, which explicitly estimates the mean activity concentration in regions without assuming strict intra-regional uniformity. APPROACH: Partial-volume correction was applied to177Lu-SPECT images obtained from the simulation of eight anthropomorphic phantoms as well as two example patients. Both sets of data were reconstructed using OS-EM with ten subsets and up to 40 iterations, including compensation for attenuation, scatter, and spatial resolution. Reconst

Targeting androgen receptor as a novel radiosensitizing therapy to improve long-term survival and anti-tumor immunity in glioblastoma via TGF-β/Smad3 Axis reprogramming.

Glioblastoma (GBM) remains the most aggressive primary adult brain cancer with limited therapeutic options. Our prior research demonstrated that androgen receptor antagonists (ARAs) enhance survival in GBM mouse models by preferentially suppressing glioma stem cells. This study investigates the potential of ARAs as radiosensitizers in combination with radiotherapy (RT). Combined effects of ARAs and RT were evaluated using an orthotopic GBM mouse model. RNA-Seq and TCGA analyses delineated AR-associated regulatory networks, while integrated cellular and molecular approaches utilizing human and

Hybrid nanovesicle artificial antigen-presenting cells integrate endogenous tumor antigen display with senescence-mimetic erythrocyte membrane-directed splenic T-cell priming.

Artificial antigen-presenting cells (aAPCs) provide a non-genetic approach to T-cell activation, but their in vivo performance is often limited by predefined peptide-major histocompatibility complex class I (pMHC-I) repertoires and inefficient delivery to secondary lymphoid organs. Here, we engineer senescence-derived hybrid nanovesicles that integrate endogenous antigen display and organ-directed trafficking within a single membrane material. Nanovesicles derived from senescent cancer cells retain increased levels of endogenously generated pMHC-I complexes, whereas nanovesicles derived from s

Giant Echinococcal Cyst of the Spleen Successfully Managed with Repeated Catheterization.

We describe a patient with a giant splenic echinococcal cyst. As surgery was contraindicated and systemic benzimidazole treatment would likely be ineffective, the cyst was successfully managed with courses of catheterization and continuous drainage. Our patient had multiple comorbidities and two previous surgeries for abdominal cystic echinococcosis. Eleven years after the first surgical intervention, a computed tomography scan revealed a giant (18.5 × 13.5 cm) echinococcal cyst in stage CE3a (WHO-Informal Working Group on Echinococcosis classification) of the spleen. Percutaneous treatment (P

Heat-Killed Lactiplantibacillus plantarum DRC2509 Isolated from Kimchi Restores Immune Function After Cyclophosphamide-Induced Immunosuppression in Mice.

The host immune system forms its first line of defense. Its innate effectors, including natural killer (NK) cells and macrophages, cooperate with adaptive lymphocytes to preserve immune competence. Aging, chemotherapy, and physiological stress erode this competence, creating a demand for safe dietary immunostimulants. Lactic acid bacteria are leading candidates because their cell-envelope structures engage host pattern recognition receptors. Interest has recently expanded from live probiotics to include postbiotics (inanimate cells that retain immunostimulatory activity without the constraints

Sequence type-associated variation in blood brain barrier invasion capacity among predominant Listeria monocytogenes strains in China.

Listeria monocytogenes (LM) can cause severe central nervous system infections; however, current understanding of its neuroinvasion is largely based on studies using a limited number of reference strains, leaving differences among sequence types (STs) in their capacity to invade across the blood-brain barrier (BBB) poorly defined. In this study, 12 LM isolates from China, representing ST5, ST8, ST9, and ST87, were investigated using whole-genome sequencing, an in vivo oral murine infection model, and an hCMEC/D3-based in vitro BBB model to assess virulence gene profiles, organ colonization, an

Identification of candidate antimicrobial peptides against the mandarin fish ranavirus using peptidomics analysis.

Viral diseases severely restrict the sustainable development of aquaculture, with Ranavirus exhibiting a broad host range and high pathogenicity but limited control options. Antimicrobial peptides (AMPs) are regarded as ideal antiviral candidates owing to their broad-spectrum bactericidal and antiviral properties. In this study, healthy and MRV-infected mandarin fish were used. Peptidomics analysis revealed 896 peptide fragments across skin (33), gill (289), liver (155), and head kidney (419). Based on APD, CAMPR3, and DBAASP, 58 candidate AMPs were identified and classified into α-helix (9),

Magnetic Particle Imaging Reveals Heterogeneous Retention, Leakage, and Redistribution of Nanoparticles Following Intratumoral Injection.

Intratumoral (i.t.) delivery of nanoparticles (NPs) is widely used to achieve high local NP concentrations. However, the temporal and spatial fate of i.t.-injected NPs remains poorly understood. We present a quantitative approach using whole-body in vivo magnetic particle imaging (MPI) to track magnetic NPs (MNPs) following i.t. injection. Using fiducial-calibrated imaging, we quantified MNP mass over time in subcutaneous 4T1 breast tumors. Longitudinal imaging revealed progressive loss of i.t. MNP content and heterogeneous off-target redistribution across animals despite standardized delivery

Sequence-defined Digital Bottlebrush Polymers for Programmable Oligonucleotide Delivery.

Oligonucleotide therapeutics hold transformative potential, yet their clinical translation is hindered by delivery barriers, including rapid renal/hepatic clearance and poor organ specificity. Bottlebrush polymer conjugates have emerged as a promising vector to address these limitations, but conventional architectures with uniform backbones can only achieve an unmodifiable, rigid biodistribution profile. Here, we report a library of sequence-defined "digital" bottlebrush polymers, precisely engineered with controlled placements of chemical motifs that modify physicochemical properties─includin

Lipid Nanoparticle Surface Engineering with Heparosan Polysaccharides Enhances mRNA Efficacy via Reduced Endosomal Retention.

In this study, we developed a library of mRNA lipid nanoparticles (LNPs) containing clinically used ionizable lipids. We surface-modified these nanoparticles with heparosan polysaccharides and systematically compared their performance with conventional poly(ethylene glycol) (PEG)-modified formulations. Our heparosan (HEP)-coated LNPs increased mRNA expression in dendritic and macrophage cell lines by up to ∼10-fold. Mechanistic studies using live-cell imaging and intracellular nanoparticle trafficking analysis revealed that HEP-modified LNPs exhibited reduced colocalization with late endosomal

Lactarius hatsudake Tanaka Polysaccharides Inhibit Hepatocellular Carcinoma by Modulating the Gut Microbiota and Its Metabolites.

We previously reported that Lactarius hatsudake Tanaka polysaccharide (LHP) can inhibit hepatocellular carcinoma, but the mechanism remains unclear especially gut microbe-mediated antitumor immunity. This research found that LHP and fecal microbiota transplantation were associated with reduced tumor growth. The ratio of CD8+/CD4+ T cells and levels of interferon-γ were increased following LHP intervention in the tumor, spleen, and blood. Additionally, in vivo and in vitro microbiome analyses indicated that LHP treatment significantly increased beneficial bacteria such as Bifidobacterium animal

Enhancing Ion Yields in MALDI-2-MSI Using Dopant-Assisted Post Ionization for High-Resolution MS-Imaging of Lipids and Metabolites.

Matrix-assisted laser desorption ionization mass spectrometry imaging (MALDI-MSI) is the most widely used label-free method for mapping the molecular distribution of lipids and metabolites from tissue sections and cell cultures. To mitigate the problem of partially low MALDI ion yields for several of these analyte classes, different post ionization techniques have recently been developed─for example, MALDI-2, where a second UV-laser pulse intersects the initial particle plume to generate additional charges. Here, we show that the MALDI-2 signal intensities of important analyte classes can be f

Discovery of a Potent, Selective, and Orally Efficacious STAT6 PROTAC for the Treatment of Atopic Dermatitis.

Signal transducer and activator of transcription 6 (STAT6), a key mediator of IL-4/IL-13 signaling, regulates Th2 differentiation, making it an attractive target for atopic dermatitis (AD). The reported phosphopeptide-based STAT6 proteolysis-targeting chimera (PROTAC) established proof of concept for targeted STAT6 degradation, although its oral bioavailability and degradation potency remain suboptimal. Here, we report the discovery of orally active non-phosphopeptide STAT6 degraders. Among them, WW-210 exhibited picomolar DC50 values for STAT6 degradation and a more than 1000-fold degradation

Evolutionary Characteristics and Immune Response of Il-17 Ligand and Receptor Families in Megalobrama amblycephala Following Aeromonas hydrophila Infection.

The Il-17 ligand family members (Il-17s) and their receptors (Il-17rs) are core immune regulatory gene families against bacterial infection in vertebrates, while their evolutionary characteristics and immune functions remain poorly understood in Megalobrama amblycephala. This study systematically identified Il-17s and Il-17rs in M. amblycephala, analyzing their evolutionary characteristics and immune response mechanisms. Sequence alignment and domain analysis showed that all Il-17s contained the conserved Il-17 domain, while most Il-17rs possessed the SEFIR domain. Furthermore, these genes sho

Tissue-specific inflammatory responses to systemic LPS in mice with LysM-lineage Hmgb1 deletion.

BACKGROUND: Systemic lipopolysaccharide (LPS) exposure induces peripheral inflammation that can engage neuroimmune pathways. High-mobility group box 1 (HMGB1) may contribute to peripheral-central communication, but the role of lysozyme M (LysM)-expressing cells in circulating HMGB1 and compartment-specific inflammatory responses remains unclear. METHODS: Male 7-8-week-old HMGB1 fl/fl LysM-Cre +  mice and HMGB1 fl/fl LysM-Cre - littermate controls received LPS (5 mg/kg, i.p.) or saline. Plasma HMGB1, midbrain inflammatory gene expression, cervical lymph node (CLN) transcriptional responses, an

Dengue virus disease phenotypes in C57BL/6N Ifnar1-/-Ifngr1-/- mice.

Dengue virus (DENV) remains a major global health problem, and experimentally tractable animal models are essential for investigating severe dengue pathogenesis and antiviral or vaccine strategies. However, whereas interferon receptor-deficient mouse models are used, the disease phenotype in C57BL/6N Ifnar1-/- Ifngr1-/- double-knockout (DKO) mice has not been sufficiently characterized. Here, we generated and characterized C57BL/6N-based DKO mice deficient in both type I and type II interferon receptors (Ifnar1-/- Ifngr1-/-). We first assessed susceptibility to DENV-2 NGC infection in wild-typ

High-dose capsaicin exposure is associated with more severe inflammation, intestinal dysregulation, and IL-17 elevation in experimental autoimmune uveitis.

PURPOSE: To investigate whether capsaicin exposure increases the severity of experimental autoimmune uveitis (EAU) and to further characterize the associated alterations in intestinal homeostasis and immune regulation. METHODS: Mice were orally administered different doses of capsaicin (CAP) from the day of EAU induction through the peak of disease on day 21. EAU severity was evaluated by fundus imaging, clinical scoring, and retinal histopathological scoring. Intestinal morphological changes were assessed by hematoxylin and eosin staining, while intestinal barrier integrity was evaluated by

Isoalantolactone exhibits activity against Echinococcus multilocularis associated with mitochondrial perturbation and changes in the lesion microenvironment.

BACKGROUND: Alveolar echinococcosis (AE) is a severe parasitic disease caused by Echinococcus multilocularis. Current therapy relies mainly on benzimidazole drugs and is largely parasitostatic, highlighting the urgent need for new candidate compounds with improved efficacy. We hypothesized that isoalantolactone (ISO) exerts direct anti-Echinococcus multilocularis activity and evaluated whether ISO treatment was associated with changes in the lesion microenvironment and host immune responses during AE progression in vivo. METHODS: In vitro experiments were conducted using isolated Echinococcus

Impact of splenic red pulp macrophages on hematopoiesis and their therapeutic effects against sepsis.

Patients with sepsis frequently experience dysregulation of hematopoietic function in the bone marrow-spleen axis, resulting in hematologic abnormalities, including anemia and leukopenia. We hypothesized that red pulp macrophages (RPMs) in the spleen contribute to hematopoiesis through the bone marrow-spleen axis in septic patients. We established the sepsis model by cecal ligation and puncture (CLP) surgery, and performed bone marrow transplantation (BMT) and splenic RPMs transplantation (RPMT) after CLP. We evaluated splenic RPMs, hematopoietic stem and progenitor cells (HSPCs; LSK cells), a
