Prenatal Detection of HSPG2-Related Dyssegmental Dysplasia, Silverman-Handmaker Type: A Rare Case Report.

INTRODUCTION: HSPG2-related dyssegmental dysplasia (heparan sulfate proteoglycan 2) is an extremely rare and severe autosomal recessive skeletal dysplasia. The low incidence and the overlapping features with other types of lethal skeletal dysplasia represent the main challenges in diagnosis. Early diagnosis is essential and can be achieved by ultrasound examination and genetic analysis. This report describes a case of HSPG2-related dyssegmental dysplasia, Silverman-Handmaker type (DDSH) diagnosed prenatally through a combination of detailed ultrasound imaging and subsequent genetic analysis.

Prenatal Diagnosis of Compound Heterozygous Beta-Thalassemia: A Report of Two Cases.

Beta (β)-thalassemia and related hemoglobinopathies remain a major genetic health burden in India. Prenatal diagnosis plays a crucial role in preventing recurrence of severe disease in high-risk couples, particularly those with previously affected children. We report two cases of prenatal diagnosis performed by mid-trimester amniocentesis in couples with prior children affected by severe β-globin disorders. In Case 1, molecular analysis of fetal DNA revealed compound heterozygosity for HBB c.92G>C (β⁺) and HBB c.126_129del (β⁰), consistent with severe β-thalassemia. In Case 2, fetal testing de

[Prenatal ultrasound and genetic characteristics of 60 fetuses with 16p11.2 microdeletion].

OBJECTIVES: The postnatal phenotypes of fetuses with 16p11.2 microdeletion mainly include autism spectrum disorder, schizophrenia, and intellectual disability, often accompanied by obesity and congenital malformations. However, studies on the prenatal phenotypes of fetuses with 16p11.2 microdeletion remain limited. This study aims to investigate the correlation between prenatal ultrasound findings and genetic characteristics of fetuses with 16p11.2 microdeletion, thereby providing evidence for clinical genetic counseling. METHODS: Clinical data of 60 fetuses prenatally diagnosed with 16p11.2

[Expression of Concern] Anti‑metastatic and differential effects on protein expression of epigallocatechin‑3‑gallate in HCCLM6 hepatocellular carcinoma cells.

INTRODUCTION: Available work from North America indicates that same-sex attracted (SSA) individuals enjoy aspects of rural life but nonetheless report encountering homophobia and experiencing isolation from SSA networks. The experience of prejudice and social isolation are often associated with psycho-social distress among the general population of same-sex attracted individuals. Little is known of how SSA women experience life in rural areas of Australia and how this influences their psycho-social wellbeing. METHODS: This was a small-scale qualitative study using guided interviews to explore

Intra-amniotic categories of preterm prelabor rupture of membranes: gestational age-specific distribution, amniotic fluid microbial profiles, and neonatal outcomes.

BACKGROUND: Preterm prelabor rupture of membranes comprises 4 intra-amniotic categories defined by the presence or absence of intra-amniotic inflammation and microbial invasion of the amniotic cavity. Their gestational age distribution, amniotic fluid microbial profiles, and associated perinatal and neonatal outcomes have not been comprehensively characterized. OBJECTIVE: The primary aim was to determine the gestational age-dependent prevalence, amniotic fluid microbial profiles, and short-term perinatal and neonatal outcomes of the intra-amniotic categories. The secondary aim was to evaluate

Preterm prelabor rupture of membranes: the use of amniocentesis to detect intraamniotic infection reduces maternal and neonatal duration of antibiotic exposure.

BACKGROUND: Current management guidelines for patients with preterm prelabor rupture of membranes at <32+0 weeks of gestation recommend administering antibiotics, although evidence on their benefits for short- and long-term neonatal outcomes is poor. OBJECTIVE: This study aimed to evaluate whether the use of amniocentesis to detect intraamniotic infection reduces maternal and neonatal duration of antibiotic exposure. STUDY DESIGN: This was a retrospective observational cohort study (2014-2023) including patients diagnosed with preterm prelabor rupture of membranes at <32+0 weeks of gestation

Countrywide Variables Associated With the Reduction of Fetuses With Down Syndrome.

In this study, we sought to analyze the associations of global macroeconomic policy decisions on live births with Down syndrome (DS). As countries made prenatal screening more available and financially covered by their governments, we modeled the impact on the "reduction percentage" for DS-that is, the percentage of fetuses with DS that were not born as a consequence of selective terminations. The log-odds of the reduction in live births of children with DS (due to selective abortion) were estimated using maternal age-based prevalence models and actual birth data. These log-odds were then pred

Clinical study of single nucleotide polymorphism-based chromosome microarray analysis in the etiological diagnosis of fetal congenital heart disease.

This study investigates the clinical application of single-nucleotide polymorphisms (SNP)-based chromosome microarray analysis (CMA) in the etiological diagnosis of fetal congenital heart disease (CHD). We 5,116 amniotic fluid samples collected through amniocentesis from January 2022 to December 2024 in Urumqi, Xinjiang, China. Based on fetal ultrasound findings, structural abnormalities, and specific types, CHD was categorized into four groups: isolated CHD, non-isolated CHD, non-CHD, and a normal group. The incidence of aneuploidies were highest in non-isolated CHD cases (16.91%), approximat

[Current role of the Kleihauer-Betke test in monitoring pregnant women].

The Kleihauer test is a widely prescribed examination today. However, we propose revisiting certain indications beyond the prevention of maternal-fetal alloimmunization, particularly concerning the adjustment of anti-D immunoglobulin dosage. Indeed, some prescriptions seem irrelevant based on the literature and the experience of the reference center. These include chorionic villus sampling, systematic prescription in cases of reduced fetal movements or metrorrhagia. Other indications could be limited to specific situations, such as direct abdominal trauma, external cephalic version (ECV) in Rh

Social vulnerability and prenatal diagnosis.

OBJECTIVES: There are limited data on how neighborhood-level risk factors affect the likelihood of having prenatal diagnosis. Neighborhood social vulnerability can be quantified and ranked using the social vulnerability index (SVI), a tool that measures the cumulative effect of external stressors in the local environment that may affect health outcomes. The objective of the study was to determine the relationship between SVI and prenatal diagnosis among pregnant patients who received genetic counseling. METHODS: Retrospective cohort study of all pregnant patients who had genetic counseling at

Preimplantation Genetic Testing Inhibits the Transmission of Pathogenic Variants Associated With Cerebral White Matter Disease.

Hereditary white matter disease is a series of progressive genetic diseases that mainly affect the white matter of the central nervous system. The development of molecular genetics enables the clinical diagnosis, carrier detection, and prenatal diagnosis of hereditary white matter disease. Here, we block the transmission of pathogenic variants in ABCD1 and NOTCH3 in a family with cerebral white matter disease via preimplantation genetic testing (PGT). Pathogenic genes were identified based on clinical manifestations, genetic background, and the results of targeted gene capture sequencing. A bl

Associations between fetal biometric para-meters and maternal characteristics in the prenatal screening phase for aneuploidies.

BACKGROUND AND AIM: Aneuploidies are rare diseases with great impact on an individual's life, as well as on their families, the reason why prenatal screening is performed, allowing families to take an informed decision. Initial prenatal screening includes the double test, triple test, non-invasive prenatal testing and ultrasonography. Among these, ultrasonography plays an important role giving information regarding an elevated risk for aneuploidies. METHODS: Eighty-four pregnant women who underwent prenatal screening were included in this study, of whom 9 cases were diagnosed with an aneuploi
