Role of Heritable and Environmental Contributions to the Development of Severe Intraventricular Hemorrhage in Very Preterm Infants: Results from a Multicenter Twins Cohort Study.

INTRODUCTION: Severe intraventricular hemorrhage (sIVH) remains a significant complication for very preterm infants (VPIs). This study aimed to assess heritable and environmental contributions to sIVH. METHODS: A total of 2,074 twin pairs born at gestational age <32 weeks with known sIVH status were identified. Three statistical methods were applied, including the Pearson χ2 test, intra-class correlation (ICC), and ACE modeling. RESULTS: Both Pearson's χ2 test (p = 0.224) and ICC analysis (p = 0.534) revealed no significant difference after comparing neither, one, or both of the monochorioni

Perinatal arterial ischemic stroke (PAIS) and neonatal cerebral sinovenous thrombosis (CSVT) in the preterm neonate: a systematic review.

Perinatal arterial ischemic stroke (PAIS) and cerebral sinovenous thrombosis (CSVT) cause significant neurological morbidity, including cerebral palsy, in preterm infants. Compared to term infants, the epidemiology, risk factors, and outcomes of PAIS and CSVT in preterm infants are less well understood. This systematic review aimed to summarize the literature on incidence, risk factors, clinical presentation, neuroimaging, and neurodevelopmental outcomes of PAIS and CSVT in infants born before 37 weeks' gestation. A comprehensive search of PubMed, Embase, and Web of Science for studies from 20

Clinical Presentation and Treatment of Hyperemesis Gravidarum in a Lady Carrying Twins in the First Trimester: A Case Reports.

We report a 12-week-old pauci-geste carrying twins who experienced potentially fatal hyperemesis gravidarum. Clinical and paraclinical investigations revealed severe hypovolemic shock/dehydration, ionic disruption, undernutrition, and acute renal failure (GFR: 36.8 mL/min/1.73 m2 according to modified MDRD). The patient spent almost 3 weeks in emergency and critical care at the University Clinics of Bukavu, where her pregnancy was tracked until her twins were born. Hyperemesis gravidarum diagnosis remains clinical, with incomplete etiopathogenesis understanding. Genetic research, including Gro

Mapping the heritability of disease: a nationwide study.

Heritability estimates are essential for understanding genetic and environmental contributions to disease, yet large-scale studies remain scarce. In this study, we leverage the Danish national health registers, including medical records for more than 10 million individuals, to estimate heritability for more than 1000 health outcomes. We estimate heritability using both twins and siblings born in Denmark between 1955-2021, providing insight into the influence of shared sibling environment with estimates that show strong concordance with published twin studies and meta-analyses. We consider the

Borderline personality disorder and other psychiatric, somatic, and behavioral conditions: a nationwide family study.

Borderline personality disorder (BPD) often co-occurs with other health conditions, but the role of genetics vs. environment is unclear. In this multigenerational cohort study using Swedish national registers, the authors quantified the co-aggregation of BPD with other phenotypes in families and estimated the genetic vs. environmental contributions. In a birth cohort of individuals born 1973-2001 and their twins, siblings, cousins, parents, and aunts/uncles, the exposure was BPD in the proband. Odds ratios were estimated for 19 psychiatric, 18 somatic, and seven behavioral/injury outcome pheno

Low birth weight, DNA methylation patterns in cord blood, and inflammation at birth.

BACKGROUND: Low birth weight (LBW) is a significant global health issue associated with an increased risk of long-term chronic diseases, yet its underlying molecular mechanisms remain unclear. DNA methylation serves as a crucial epigenetic mechanism for investigating these associations. Our study employed a unique model of birth weight-discordant monozygotic (MZ) twin neonates and a multi-level validation strategy to elucidate the mechanisms by which LBW affects immune function through DNA methylation at birth. METHODS: We performed genome-wide DNA methylation profiling using methylated DNA i

Parental body mass index and offspring childhood body size and eating behaviour: A structural equation modelling analysis in the Norwegian Mother, Father and Child Cohort Study.

BACKGROUND: The intergenerational transmission of obesity-related traits could propagate an accelerating cycle of obesity, if parental adiposity causally influences offspring adiposity. The extent to which intergenerational obesity associations are due to such causal effects, as opposed to genetic confounding (inheritance), is unclear. We aimed to establish whether associations between parental peri-pregnancy body mass index (BMI) and offspring birth weight (BW), BMI until 8 years of age, and 8-year-old eating behaviour are due to genetic confounding. METHODS AND FINDINGS: Data were from the

Association between ADHD symptoms and device-measured physical activity and sedentary behavior in childhood: A population-based twin study.

BACKGROUND: Attention-deficit/hyperactivity disorder (ADHD) symptoms are associated with adverse health outcomes; this may be partially due to unhealthy lifestyle factors such as low physical activity (PA) and high sedentary behavior (SED). However, this association remains unclear. ADHD and PA have a medium-to-high heritability; therefore, genetically informed studies are warranted. This study aimed to increase the understanding of the association between ADHD symptoms and time spent in PA and SED in children. Additionally, we aimed to determine whether sex, symptom type, and genetics influen

Cardiac morphology and flow dynamics in children born to hypertensive pregnancies.

AIMS: Hypertensive pregnancies affect up to 10% of pregnancies globally and are linked to greater cardiovascular risk in offspring. Subclinical cardiac changes have been reported across developmental stages. This study aimed to assess whether children born to hypertensive pregnancies exhibit altered left ventricular (LV) morphology and cardiac flow characteristics. METHODS AND RESULTS: In this cross-sectional study, cardiovascular magnetic resonance (CMR) was performed on a 3.0-Tesla Siemens PRISMA scanner in children aged 6-9 years: 44 offspring of hypertensive pregnancies (50% female) and 5

Postnatal growth and childhood asthma in preterm and full-term twins: A cohort study.

BACKGROUND: The role of postnatal growth in asthma development in children remains unclear. We examined the association between postnatal growth pattern and childhood asthma and assessed whether this association differed between preterm and full-term children. METHODS: This cohort study included twins from the Child and Adolescent Twin Study in Sweden (born 1992-2003), linked to national registers. Postnatal weight and height to age 5 were obtained from child health records and standardized for age and sex (SDS). Childhood asthma was defined by parent-reported asthma at age 9 or 12. Analyses

Mortality in Multiple versus Singleton Very Low Birth Weight Infants in Germany: A Population-Based Study.

INTRODUCTION: In several large cohorts from the USA and China, multiple as opposed to singleton birth was found to be associated with increased mortality, while studies from European countries have yielded conflicting results. We aimed to assess the association between plurality and survival of very low birth weight infants in relation to gestational age. METHODS: Data of preterm infants <1,500 g birth weight obtained from annually published quality improvement data mandatorily collected from all hospitals in Germany (2010-2017, 2019-2024) were analyzed for mortality of singletons versus mult

Lifestyle behaviours in children born extremely preterm at 2 years: a comparison to a Dutch reference population.

UNLABELLED: Children born extremely preterm (EP, < 28 weeks gestation) are at increased risk of lifestyle-related morbidities. This study characterized parent-reported lifestyle behaviours in EP-born children at 2 years' corrected age (CA) and compared them with a Dutch reference population. In a multicentre prospective cohort study, parents completed the FLY-Kids lifestyle screening questionnaire (2 years' CA), which assessed parental satisfaction and adherence to age-specific recommendations for diet, eating habits, physical activity, screen time and sleep. Outcomes were compared with a Dutc
