Effective healing of diabetic wounds requires spatiotemporal coordination of immunoregulation and tissue regeneration, which is often overlooked in conventional treatments. Here, we developed an extracellular matrix-mimicking dressing, termed ANM, that modulates arginine utilization within the diabetic wound microenvironment to re-establish the spatiotemporal coordination of healing. ANM was in situ fabricated by hand-held electrospinning using a rationally designed arginine- and norvaline-based poly(ester amide), yielding a conformable and breathable nanofibrous dressing. Its porous and hydro

Rhinitis is a heterogeneous inflammatory condition of the upper airway mucosa. Chronic symptoms are often refractory to first-line intranasal antihistamines and corticosteroids, particularly in mixed and nonallergic phenotypes. Add-on anticholinergic treatment alleviates rhinorrhea but is hampered by short duration of action and multiple daily administrations. We describe here unique heterobifunctional M3/H1 dual antagonists designed for once-daily intranasal treatment. By tuning a novel bifunctional scaffold combining a diarylsulfone-based muscarinic antagonist with various antihistamine chem

Helicobacter pylori (H. pylori) induces gastric precancerous lesions (GPL), but their underlying biomarkers remain poorly characterized. To establish a GPL model, BALB/c mice were inoculated with H. pylori via gavage. Immunofluorescence confirmed successful modeling by detecting H. pylori distribution and aberrant expression of precancerous lesion markers. Serum-based label-free proteomics and bioinformatic analyses were performed to screen key proteins. Data are available via ProteomeXchange (PXD076808). Western blot and immunofluorescence were used to verify key proteins and markers of DNA d

Dense active materials must continuously relieve internal mechanical stress to remain structurally stable as they are driven far from equilibrium. In epithelial tissues, this relief occurs through cell division, where, according to Hertwig's century-old rule, cells divide along their long axis. We show that as epithelia densify, this geometric rule is superseded by a mechanical one: cells divide along the principal axis of anisotropic stress, independently of the isotropic stress state. Using direct force measurements and stress inference, we demonstrate that stress orientation governs divisio

Precise gene dosage control is critical for cellular identity and development, particularly for imprinted genes, where dosage imbalances are linked to neurodevelopmental disorders. The Mest/Copg2 imprinted locus exemplifies this fine-tuned regulation: while Mest is constitutively paternally expressed, Copg2 shifts from biallelic to maternally-biased expression specifically during neural differentiation, a transition proposed to involve transcriptional interference by the long Mest isoform, MestXL, which extends into Copg2. However, the mechanisms underlying this switch and whether factors beyo

Transcytosis-mediated delivery effectively enhances tumor penetration of nanodrugs but is less efficacious in stroma-rich tumors. Cancer associated fibroblasts (CAF) can represent a physical barrier for drug delivery in tumor cells. Herein, by selectively targeting NPC1L1-cancer associated fibroblasts, surface-displayed cholesterol enhances the delivery of a platinum nanodrug (PtD-Chol) in stroma-rich tumors, restoring anti-tumor efficacy. The findings reveal that PtD-Chol, acting through the 'shared entry portal' of NPC1L1 triggers cross-multicellular transport (CMCT) within tumors, shuttling

Colorectal cancer (CRC) progression and therapeutic resistance are strongly associated with the ability of tumor cells to evade apoptosis through the dysregulation of key anti-apoptotic pathways. Among the molecular mechanisms involved in CRC progression, microRNAs (miRNAs) have emerged as key post-transcriptional regulators of pathways controlling tumor growth, invasion, therapeutic response, and cell survival. We previously demonstrated that miR-195-5p acts as a tumor suppressor in CRC by impairing proliferation, migration, invasion, and cell-cycle progression. However, its contribution to t

Lipodystrophies are a group of disorders featuring reduced adipose tissue mass or function, which often leads to significant metabolic disease, reduced lifespan and impaired quality of life. Individuals with congenital generalised lipodystrophy (CGL) have severely reduced adipose tissue mass. The loss of healthy systemic lipid storage typically causes hepatic steatosis and lipoatrophic diabetes. In addition, adipocyte-secreted hormones including leptin and adiponectin are dramatically reduced. Leptin has critical roles regulating appetite and broader effects on lipid and glucose metabolism. Acute pancreatitis (AP) is a pancreatic inflammatory disorder that can progress to fatal systemic inflammation and multiorgan failure. Although tumor necrosis factor (TNF) signaling is a key regulator of inflammation, its contribution to AP progression and its therapeutic potential remain incompletely understood. We induced AP in wild-type and Tnfr1-/- mice using caerulein, pancreatic duct ligation, or L-arginine. Single-cell RNA sequencing (scRNA-seq) and bulk RNA sequencing were used to characterize pancreatic immune-cell profiles, and pomalidomide, infliximab, and necrostatin-1 were evaluat

Pancreatic ductal adenocarcinoma (PDAC) is characterized by a dense desmoplastic stroma and early metastatic dissemination, both of which contribute to its poor prognosis. While forkhead box protein 3 (FOXP3) is best known as a lineage-defining transcription factor in regulatory T cells (Tregs), its aberrant expression in cancer cell-specific FOXP3 (c-FOXP3) may also support tumor-cell-intrinsic progression. Our findings suggest that high c-FOXP3 expression is closely linked to aggressive disease behavior and metastatic progression in PDAC. c-FOXP3 enhanced migration, invasion within three-dim

Pancreatic ductal adenocarcinoma (PDAC) develops chemoresistance through dynamic cell-state transitions, yet the regulatory network architecture governing drug-tolerant states remains poorly characterised, because static expression snapshots cannot capture how interaction topology evolves along treatment-induced trajectories. We established a 3D PANC-1 tissue model on decellularised porcine intestinal matrix, which reaches patient-like proliferation indices and tissue-niche-specific drug responses, and performed single-cell RNA sequencing across four conditions: control, gemcitabine (GEM), TGF

Imidaclothiz (IMZ), a next-generation neonicotinoid pesticide, has garnered significant attention due to its detection in human samples. In this study, we investigated its toxicokinetics and biotransformation in rats following oral administration. IMZ was rapidly absorbed, reaching peak plasma concentration at 0.75 h, followed by a prolonged elimination phase with minimal decrease between 12 and 48 h. Tissue distribution analysis revealed notable accumulation and slower elimination of IMZ in the kidneys, testes, and muscle. Eleven metabolites were identified, with M227, M231, and M233 being re

Polystyrene nanoplastics (PS-NPs) are emerging neurotoxicants with the potential to access the brain following systemic exposure; however, their region-specific effects and underlying mechanisms remain incompletely understood. In this study, adult male mice (C57BL/6J) were intravenously injected with unlabeled or fluorescently labeled PS-NPs (50-60 nm) at a dose of 400 mg/kg, and brain tissues were collected and analyzed 3 days post-injection to evaluate PS-NPs distribution, oxidative stress, and regulated cell death pathways. Confocal imaging revealed preferential accumulation of fluorescent

Thermoembolization (TE) is a new, minimally invasive approach that could improve outcomes in select patients with hepatocellular carcinoma. However, little is known about the effects of this procedure at the molecular level. To gain greater insight into the molecular effects of TE, we performed a pilot study of transarterial TE with 2-propylpentanoyl chloride in a swine model with 3 animals. Liver tissues were harvested 7 days after the procedure, and formalin-fixed specimens were subjected to serial mass spectrometry imaging (MSI) analyses of metabolites, glycans, and tryptic peptides. Precision-cut liver slices (PCLSs) are an ex vivo liver model that allows isolation of extracellular vesicles (EVs), which are nano-sized particles involved in intercellular communication. However, EV recovery from PCLSs is hindered by adhesion to the extracellular matrix and macromolecules such as extracellular DNA. Enzymatic treatments, such as collagenase (COL) and the endonuclease benzonase (BEN), can aid their release, but potential effects on the tissue are unclear. Here, the effect of these enzymes separately, as well as combined, was analyzed to determine their effects on morphology, p

Ferroptosis is an iron-dependent, non-apoptotic form of regulated cell death that is driven by lipid peroxidation. In the past decade, it has emerged as a core pathogenic mechanism in a wide range of human diseases, such as neurodegenerative disorders, cardiovascular disease, cancer, and notably, reproductive dysfunction. This review provides a comprehensive overview of the molecular mechanisms underlying ferroptosis with a specific focus on the System Xc⁻-GSH-GPX4 axis, iron metabolism, lipid peroxidation, and the regulatory roles of key modulators such as Nrf2, ACSL4, and CISD2. This review

BACKGROUND: High-flow nasal cannula (HFNC) is widely used as an early respiratory support strategy for adults with acute hypoxemic respiratory failure (AHRF). Although HFNC may reduce the risk of endotracheal intubation, it remains uncertain whether the available evidence is sufficiently mature and whether this potential benefit translates into improved survival. We performed a systematic review and meta-analysis with trial sequential analysis (TSA) and GRADE assessment to assess HFNC versus standard oxygen therapy (SOT) in adult AHRF.

BACKGROUND: Faecal microbiota transplantation (FMT) and faecal filtrate transplantation (FFT) have emerged as promising methods to prevent intestinal dysbiosis and diarrhoea in piglets after weaning. The aim of this study was to compare the effects of FMT and FFT on intestinal health and faecal microbiota dynamics in weaned piglets. Thirty-six 28-day old piglets were allocated into three groups (Control, FMT, and FFT, n = 12 each) at an experimental farm. Intact and filtered faeces from healthy sows were lyophilised, encapsulated, and administered upon arrival (day 0) and after 4 days (day 4).

Perfluorooctanoic acid (PFOA) has been widely used in industrial and consumer products and was recently classified as a Group 1 carcinogen by the International Agency for Research on Cancer. However, uncertainties remain regarding transcriptomic bioactivity thresholds associated with liver effects and their variability across clinically relevant populations. In this study, previously published transcriptomic data obtained following 24-h and 10-day exposure of primary human liver spheroids (LS) to PFOA were integrated into a physiologically based toxicokinetic (PBTK), population-specific framew

INTRODUCTION: Autologous costal cartilage grafting is commonly used for pediatric tracheal reconstruction; however, it has several limitations, including restricted availability and surgical invasiveness. In this study, we investigated the feasibility of a novel tracheal reconstruction approach using human embryonic stem cell-derived cartilage tissue (ESCaT).
METHODS: ESCaT was differentiated from human embryonic stem cells (SEES2), and large-scale cartilage constructs were generated by integrating tissues derived from multiple culture dishes within a nylon mesh scaffold. Decellularized ESCaT
