Repurposing BH3 mimetics to deplete tumour-infiltrating regulatory T cells and enhance anti-tumour immunity in non-small cell lung cancer.

Tumour-infiltrating FOXP3+ regulatory T cells (Tregs) exert suppression of anti-tumour immunity in non-small cell lung cancer (NSCLC), contributing to poor prognosis and immunotherapy resistance. The BCL-2 family pro-survival protein, MCL-1, is a critical controller of lymphoid Treg viability, yet its role in tumour-infiltrating Tregs remains poorly defined. Here we find that tumour-infiltrating effector Tregs in human NSCLC exhibit an activation-associated shift in BCL-2 family pro-survival protein expression typified by elevated MCL-1 expression. Pharmacological inhibition of MCL-1 with the

A map of intra- and intercellular immune responses across diverse in vitro stimuli and inflammatory disease.

BACKGROUND: In vitro stimulation of healthy human immune cells is widely used to model the immune states observed in disease, both to investigate pathology and to test therapeutic approaches. However, experiments typically focus on individual cell types or stimuli and a comprehensive cellular comparison of common immunomodulators and their relevance to disease is lacking. METHODS: We used single-cell transcriptomics to generate a reference profile of human peripheral blood mononuclear cells treated with 11 different in vitro stimuli, totalling over 150,000 cells across 21 immune cell types. [A study on HNA-1~-5 genotypes and frequencies in the blood donor population of jiangsu province].

Objective To investigate the distribution characteristics of the genotypes and allele frequencies of HNA-1~-5 alleles in a blood donor population from Jiangsu Province. Methods Genomic DNA was extracted from 300 peripheral blood samples collected from healthy donors. All samples were genotyped using an optimized polymerase chain reaction-sequence specific primer (PCR-SSP) protocol. Randomly selected results were verified by direct sequencing, and genotype distributions were tested for conformity to Hardy-Weinberg equilibrium. Samples identified as potential HNA-1 null were confirmed by polymer

Mosaic chromosomal alterations and relation to haematological cancer in two large Danish cohorts.

Mosaic chromosomal alterations (mCAs) are large somatic gains, losses and copy-neutral loss of heterozygosity identified in the peripheral blood, which represent a relatively unexplored spectrum of clonal haematopoiesis. We aimed to describe mCAs in two large Danish cohorts with extensive long-term registry-based follow-up and examine associations with haematological malignancy. We analysed genotype data from 97 752 participants from the Danish Blood Donor Study and 86 301 patients from the Copenhagen Hospital Biobank using the MoChA software. Association to haematological cancers was modelled

Extended red cell antigen frequencies in a Southwestern Colombian donor population: Implications for phenotype-matched transfusion strategies.

BACKGROUND AND AIMS: The complex history of genetic admixture in Latin American populations creates unique erythrocyte antigen profiles. This study aimed to determine the extended red blood cell (RBC) antigen frequencies in a blood donor population from Southwestern Colombia, a region with significant African ancestral heritage, to support phenotype-matched transfusion strategies and optimize donor selection. MATERIALS AND METHODS: A cross-sectional study was conducted on 501 voluntary blood donors at a highly complex hospital blood bank in Cali, Colombia. Extended serological phenotyping for

First Vel-null alloimmunized blood donor identified in Uruguay.

A 52-year-old, first-time blood donor presented with panreactivity during routine antibody screening. Current serologic evaluation showed uniform 2+ reactivity by the gel-indirect antiglobulin test and the conventional tube low-ionic-strength saline-indirect antiglobulin test. The molecular analyses revealed a homozygous 17-base pair deletion spanning nucleotides 64-80 in exon 3 of SMIM1 (VEL*01N.01/VEL*01N.01), confirming the Vel-null phenotype. The antibody was identified as anti-Vel IgG3, reacting in tube testing, exclusively in the anti-human globulin phase. We present the first case of a

Eligibility to donate blood under an individual donor assessment policy: A national cross-sectional study of gay, bisexual and other men who have sex with men in Aotearoa, New Zealand.

BACKGROUND: To protect the blood supply, many countries defer higher-risk donors such as men who have had sex with men (MSM) in the last 3 months. However, this may be unnecessarily restrictive and increase non-compliance risks. New Zealand (NZ) has shifted from this approach to individual donor assessment (IDA), but the potential impact is poorly understood. We aimed to estimate the increase in blood donor eligibility and examine the characteristics of newly eligible MSM. METHODS: We used data from SPOTS, a large national cross-sectional online survey of MSM. Items included socio-demographic

HTLV screening and confirmatory testing among blood donors in São Paulo: A cross-sectional study.

INTRODUCTION: Human T-cell lymphotropic virus types 1 and 2 (HTLV-1/2) are transfusion-transmissible retroviruses associated with severe diseases, and blood donor screening is mandatory in Brazil. Increasing HTLV-related discard rates at our institution prompted this study to determine the prevalence and incidence of HTLV-1/2 and to investigate the presence of HTLV-3 among donations reactive in the ARCHITECT rHTLV-I/II CMIA assay. METHODS: A retrospective cross-sectional study was conducted among 220 340 volunteer blood donors at Fundação Pró-Sangue/Hemocentro de São Paulo, Brazil, between Se

Cross-Model Explainability Consistency in Hepatitis C Stage Classification: A SHAP, LIME, and Counterfactual Analysis Across Five Machine Learning Architectures.

Background: Hepatitis C virus (HCV) affects approximately 50 million people worldwide and progresses silently through distinct hepatic stages, yet most machine learning staging approaches offer no clinician-facing explanation and rarely evaluate whether explanations hold across architectures. Methods: We trained five models-Logistic Regression, Random Forest, XGBoost, LightGBM, and SVM-on the UCI HCV dataset (615 patients, four classes after merging the seven-instance suspect blood-donor group into a single Donor/Control class) and applied SHAP, LIME, and DiCE counterfactuals. Anti-PEG Immunogenicity of mRNA-LNP Vaccines in Humans: Evidence for Population-Level Changes in the Anti-PEG Antibody Repertoire.

Background/Objectives: Polyethylene glycol (PEG) is widely used to enhance the stability and pharmacokinetics of nanomedicines, including lipid nanoparticle (LNP)-based mRNA vaccines. However, both pre-existing and vaccine-induced anti-PEG antibodies may compromise the efficacy and safety of PEGylated therapeutics. Methods: In this study, we analyzed the specificity and avidity of anti-PEG antibodies in human blood donor samples from Unvaccinated individuals and recipients of PEGylated mRNA-LNP vaccines (Comirnaty and Spikevax), polysorbate-containing vaccines, or vaccines lacking both PEG and

Seroprevalence of hepatitis B and C in asymptomatic blood donors in Saudi Arabia: A systematic review and meta-analysis.

Viral hepatitis remains a serious risk to transfusion safety, despite being considerably reduced by donor screening. Objectives: The aim of this study was to evaluate the seroprevalence of HBV and HCV among asymptomatic blood donors in Saudi Arabia in the past 15 years (2010-2025). A literature search has been conducted on seven databases. Prevalence rates with 95% confidence intervals (CIs) were pooled via a random effects model. Twelve studies, examining 597 820 blood donors, were included in this study. The overall pooled prevalence rate of HBV and HCV among blood donors was 0.455% (95% CI:

Self-determination and plasma donation: The role of autonomous motivation.

The retention of plasma donors is a major challenge for blood collection establishments. The literature shows that self-determination theory (SDT) presents a relevant conceptual framework for analysing the determinants of blood donation. This research aims to improve the understanding of the motivational mechanisms among donors. 277 French plasma donors (Mage = 43.21 years; SD = 13.83) responded to a French adaptation of the Blood Donor Identity Survey. Regressions were performed to predict intention to donate again, as well as subsequent donation behaviours. Controlling for age, gender and do
