Rare subgroup of A phenotype-a case series.

Testing for ABO incompatibility between a potential blood donor and recipient is the cornerstone of pretransfusion testing. Occasionally, weak agglutination reactions occur due to low expression of A and B antigens on the surface of red blood cells. This can lead to inconsistencies in blood group typing, as seen in a case where a patient's forward and reverse typing showed discrepancies. Following a comprehensive analytical assessment, the patient was identified as having a distinctive variant of blood group A, belonging to the less common subgroups-A3, Aend, Ax, Am, Ay, and Ael. Herein, we pr

Determinants of birth preparedness and complication readiness among postpartum women in Bugembe, Jinja City, Eastern Uganda: a community-based cross-sectional study.

BACKGROUND: Birth preparedness and complication readiness (BPCR) is vital for reducing maternal deaths by promoting proactive planning for safe childbirth and medical emergency management. Despite its importance in improving maternal and neonatal health outcomes, the field is still under-explored in Uganda. This study investigates factors associated with BPCR among postpartum women in Eastern Uganda, where maternal mortality remains high. METHODS: A community-based cross-sectional study was conducted in Bugembe town council, Jinja City, Eastern Uganda, from August to September 2022. The study

Standardized reporting of mononuclear cell apheresis collections: a systematic review and expert recommendations.

Circulating cells to be used for cell therapy are typically collected from peripheral blood through apheresis. Objective analysis and comparison of apheresis outcomes are crucial for optimizing procedures. In this study, we reviewed published data on the performance and product characteristics of two widely used apheresis devices, Spectra Optia (Terumo Blood and Cell Technologies) and Amicus (Fresenius Kabi). We systematically searched PubMed on 18 April 2023 to retrieve studies published since 2013 reporting on mononuclear cell (MNC) collection for extracorporeal photopheresis, CD34+/stem cel

Antihuman leukocyte antigen antibodies and risk of fetal growth restriction.

BACKGROUND: Fetal growth restriction is a major cause of stillbirth, neonatal morbidity, and long-term cardiovascular and neurodevelopmental complications. While placental dysfunction is central to many cases, a substantial proportion remains unexplained. Chronic inflammatory placental lesions, such as chronic histiocytic intervillositis or massive perivillous fibrin deposition, are associated with fetal growth restriction and have been interpreted as possible manifestations of maternal antifetal rejection mediated by the human leukocyte antigen system. Maternal antihuman leukocyte antigen ant

Delayed Adverse Reactions Following Whole-Blood and Plateletpheresis Donation: A Prospective Observational Study From a Tertiary-Care Blood Center.

BACKGROUND AND AIM: Delayed adverse reactions occurring after donors leave the blood center may be underrecognized by routine on-site hemovigilance. Comparative data on delayed reactions following whole-blood donation and plateletpheresis within the same institutional setting remain limited. This study aimed to determine the frequency and pattern of delayed adverse reactions and evaluate their associations with donor age, sex, donation status, and donation type. METHODS: This prospective observational study included 17,923 donors (17,633 whole-blood and 290 plateletpheresis) at a single terti

Cephalic Venous Aneurysm and Intravascular Papillary Endothelial Hyperplasia in a Blood Donor.

Venous aneurysms (VAs) and pseudoaneurysms (VPAs) are uncommon vascular lesions with variable etiology and location. We report a 52-year-old male blood donor who developed thrombophlebitis presenting as a right antecubital mass after multiple venipunctures. Initial duplex ultrasound revealed a saccular dilation of the right cephalic vein (15.6 × 7.8 × 25.5 mm) with thrombus, as well as thrombosis of the right brachial vein. The patient was treated with anticoagulation, followed by definitive surgical management. Histopathological examination revealed a VA, as well as intravascular papillary en

The power of collaboration: the untold story of World Blood Donor Day and World Patient Safety Day.

Global health days endorsed by the World Health Assembly (WHA) play a distinctive role in elevating priority issues, mobilizing political commitment, and catalyzing collective action. Among them, World Blood Donor Day (WBDD) and World Patient Safety Day (WPSD) stand out as landmark initiatives that emerged not only from technical necessity but also through sustained collaboration among Member States, WHO, and key partners including international organizations, professional bodies, civil society, and advocates. More than symbolic observances, they serve as governance instruments that confer leg

Impact of blood donor, component and recipient variables on antigen-specific alloantibodies.

A better understanding of factors influencing red blood cell (RBC) alloimmunization is desirable, given the dangers of this non-infectious serious hazard of transfusion. In a case-control retrospective study utilizing the Recipient Epidemiology and Donor Evaluation study (REDS)-III Biological Specimen and Data Repository Information Coordinating Center (BioLINCC) dataset, we investigated blood donor, component and recipient variables associated with RBC alloantibody specificities. 19 697 controls with no new alloantibody detected 2-16 weeks post-transfusion and 861 cases (with antibodies inclu

Transfusion-transmitted Plasmodium vivax malaria in a hemodialysis patient in non-endemic area: A case report.

Transfusion-transmitted malaria (TTM) is a rare but potentially serious problem particularly in non-endemic areas. Detecting asymptomatic parasitemia is a challenge for current screening strategies, especially in countries with heterogeneous malaria endemicity such as Indonesia. We report a case of probable TTM due to Plasmodium vivax in a 75-year-old woman with end-stage renal disease (ESRD) undergoing regular hemodialysis in a non-endemic city, presented with a one-week history of intermittent fever, chills, myalgia, and generalized weakness, without any recent travel to malaria-endemic area

Increased blood levels of neutrophil- and platelet-derived markers in patients with radiographic axial spondyloarthritis: a pilot study.

A challenge in diagnosing and monitoring radiographic axial spondyloarthritis (r-axSpA) is the absence of reliable biomarkers. This cross-sectional pilot study aimed to evaluate platelet- and neutrophil-derived substances as potential biomarkers in r-axSpA, given the involvement of these cells in disease pathology. Serum and plasma were collected from 13 male, HLA-B27-positive r-axSpA patients without disease-modifying anti-rheumatic drugs and 13 age- and sex-matched blood donor controls. Concentrations of platelet-derived soluble CD40 ligand (sCD40L) and soluble P-selectin (sP-selectin) and n

The significance of vascular endothelial growth factor and systemic inflammatory reaction in bladder cancer patients.

Bladder cancer (BCa) is the most frequently diagnosed malignancy of the urinary system and is characterized by a multifactorial etiology. Multiple growth factors have been implicated in BCa progression. This cross-sectional study investigated whether the pro-angiogenic factors angiopoietin-like 4 (ANGPTL4), insulin-like growth factor 1 (IGF-1), fibroblast growth factor (FGF), and vascular endothelial growth factor (VEGF) could differentiate BCa patients according to the degree of systemic inflammation. Based on histopathological examination, 101 participants were enrolled and divided into a ca

Aiolos and Eos drive distinct human TH17 functional states.

CD4+ T helper (TH)-17 cells play a pivotal role in mucosal immune defense and are implicated in autoimmune diseases and cancer. Although Th17 cell plasticity is well-studied in mice, the factors driving their transition between pro-inflammatory and immunomodulatory states in humans remain less understood. Our study explored the transcriptional and epigenetic landscapes of single-cell cultures of human memory TH17 cells, focusing on clones that produce either immunomodulatory IL-10 or pro-inflammatory IFNγ and IL-22. We found that IL-10+ TH17 cells exhibit a T cell exhaustion-like profile with

IDH3 regulates citrate homeostasis to control metabolic fitness and venetoclax resistance of AML stem cells.

Targeting metabolic dependencies of leukemic stem cells (LSC) may open avenues to improve outcomes of patients suffering from acute myeloid leukemia (AML). LSCs rely heavily on an active tricarboxylic acid (TCA) cycle and mitochondrial oxidative phosphorylation whereas healthy hematopoietic stem and progenitor cells (HSPCs) possess more metabolic flexibility. Here, we identify the TCA cycle enzyme isocitrate dehydrogenase 3 (IDH3) as a critical and selective regulator of LSC maintenance. IDH3 is more abundant in LSCs compared to healthy HSPCs, and TCA cycle activity correlates with inferior cl

Associations of blood donor and product characteristics with platelet transfusion outcomes.

To understand the impact of blood donor and product characteristics on platelet transfusion outcomes, we linked these variables with recipients of single-unit apheresis-derived platelet products between 1 June 2020 and 31 March 2022. We used multivariable logistic regression to examine associations between donor and product characteristics on posttransfusion platelet counts and 24-hour red blood cell (RBC) transfusion events in 2808 transfusion recipients who received ≥1 platelet transfusions (N = 8207 units). Posttransfusion platelet increments >20 × 109/L and RBC transfusion within 24 hours

Updated Burden and Long-Term Projections of Chronic Hepatitis B and C in Japan: A Nationwide Markov Modeling Study.

AIM: Chronic hepatitis B virus (HBV) and hepatitis C virus (HCV) infections remain major causes of liver-related morbidity and mortality worldwide. Although Japan has implemented nationwide screening and treatment programs for decades, updated nationwide estimates and long-term projections are limited. We aimed to estimate the number of carriers and patients in Japan in 2020 and to project through 2050. METHODS: The carriers were classified into six categories: undiagnosed carriers, patients in care, diagnosed but not linked to care, new infections, cured, and deceased. Multiple nationwide da

Blood donor hepatitis B data: An under-recognized surveillance resource in Canada.

BACKGROUND: Hepatitis B virus (HBV) infection surveillance is essential to gauge progress toward elimination, assess success identifying/treating cases, and evaluate prevention strategies. Canadian surveillance relies primarily on case detection. Data on undiagnosed HBV infection prevalence are limited. We explore the insight blood donors can bring to surveillance. METHODS: Drawing upon the authors' expertise, operational knowledge, and published literature, we identified key topics to evaluate blood donor data. We then synthesized the relevant data and publications to propose unique insight

Field evaluation of two commercial feline immunodeficiency virus antibody detection point-of-care kits in domestic cats.

BACKGROUND: Timely diagnosis of feline immunodeficiency virus (FIV) is essential for informed clinical decision-making, particularly when screening cats before admission into multi-cat environments or assessing blood donor candidates. Point-of-care (PoC) antibody detection kits play a central role in these settings, but their clinical utility relies on independent evaluations under field conditions. This study aimed to assess the diagnostic performance of two commercially available PoC kits for which independent evaluation data are lacking. METHODS: A total of 116 archived serum samples from a
