Highly enriched preparations of monocytes, B and T lymphocytes, and granulocytes from 18 normal donors were serotyped in parallel in a complement-dependent cytotoxicity assay using allogeneic and heterologous antisera defining three independent tissue antigen systems. Eight monkeys received bilateral corneal transplants, one fresh and one cryopreserved, from eight other healthy monkeys of sex opposite to that of the recipients. Three of the animals maintained clear grafts in both eyes, and the sex chromatin patterns of the corneal endothelial cells in these transplants were studied at one, two, and three years after keratoplasty. Definite survival of donor endothelial cells of their progeny was observed in each graft, and the percentage of cells of donor origin was similar for both fresh and cryopreserved grafts at each timer period.

Following the implantation of syngeneic chromosomally marked thymuses under the kidney capsule of normal adult mice, donor T cells were found in the blood and other lymphoid organs. At the present time, two major factors limit further clinical application of islet cell transplantation in treatment of the insulin-deficient diabetic patient. First, the yield of islet tissue obtainable from a single donor pancreas is insufficient for adequate reconstitution of normal beta cell mass. We used the clinical specular microscope to examine the endothelium of 14 clear penetrating corneal grafts that ranged from 4 days to 2 1/2 years posttransplantation. In 32 experiments involving transplantation of human malignant melanomas to the mouse mutant nude, tumourtake was observed in 14 instances. The tumour tissue which took and grew after inoculation was derived from malignant melanoma metastases of the skin (10 takes from 13 transplantations), and from primary malignant melanomas of the eye (4 takes from 6 transplantations). Evidence is presented that there are differences between pig liver and kidney parenchymal cells in the expression of major histocompatibility complex antigens. Analogues of I-region antigens of the mouse are detectable on kidney cells and some peripheral blood leukocytes (PBL) but not on liver, platelets, and other PBL. Such differences between liver and kidney may explain (a) the fate of these organs as allografts and (b) the donor-specific protection of kidney by liver. Other antigens peculiar to skin and kidney have been detected but require further characterization.

Differential survival times of various organ allografts in the rat across the same histocompatiblity barrier were studied by transplanting the kidney, heart, intestine, pancreas, and skin from (BN X Le)F1 hybrid donors to Lewis recipients. Six colostrum-deprived SPF lambs inoculated endobronchially with a second passage broth culture of a Scottish strain of Mycoplasma ovipneumoniae, were killed in batches of two at seven, 14 and 28 days post-inoculation. A network of small to medium-sized communities has been developed to increase the supply of cadaveric kidneys for renal allotransplantation. After a training period, community physicians and the community hospitals were designated as members of the transplant team to retrieve kidneys. A threefold increase in the number of cadaveric kidneys has resulted from this project. This mechanism for organized kidney retrieval on a regional basis could be used to augment the supply of cadaveric organs in other areas of the United States.
