Postoperative complications at the palatal donor site following autologous soft tissue grafting: a systematic review.

This systematic review analyzes postoperative complications at the palatal donor site following the harvesting of autologous soft tissue grafts in periodontal surgery. Although these grafts remain the clinical reference standard due to their effectiveness, their harvesting may be associated with donor-site morbidity, including pain, bleeding, partial necrosis, and sensory disturbances. A comprehensive search was conducted in PubMed, Scopus, and Web of Science, identifying 16 clinical studies published between 2019 and April 2025. The included studies comprised randomized controlled trials, pro

A comprehensive proteomic and transcriptomic mapping study found porcine and human kidneys exhibit a profoundly altered biological profile during warm ex vivo perfusion.

INTRODUCTION: Organ shortages remain a major limitation to kidney transplantation. Normothermic machine perfusion (NMP) is a technique to address this challenge by improving quality assessment and ex vivo organ preservation. However, the molecular state of the organ during NMP, and how it differs from in vivo physiology, remains poorly defined. METHODS: To address this, we performed RNA sequencing and LC-MS/MS analyses of porcine and human kidneys during NMP. Cortical kidney tissue was collected from 60 porcine and 24 human donor kidneys at multiple time-points, including an in vivo baseline

Progressive Pseudorheumatoid Dysplasia Presenting with Cauda Equina Compression: A Case Report.

PURPOSE: Fresh vegetables are essential human dietary components but are increasingly implicated in foodborne infections caused by Escherichia coli. Contamination may occur during cultivation, irrigation, handling, and retail display, and the presence of pathogenic or antimicrobial-resistant E. coli poses significant public health risks. Despite widespread vegetable consumption in Ogun State, Nigeria, limited data exist on the resistance and virulence characteristics of E. coli in these foods. This study therefore assessed antimicrobial resistance profiles and virulence genes in E. coli isolat

Transient culture of adult human hepatocytes as organoids improves engineered liver engraftment.

Bioengineered livers could serve as a bridge or alternative to whole organ transplantation for patients suffering from liver disease. However, identifying cellular configurations that promote adult human hepatocyte survival after tissue transplantation remains a critical hurdle. We hypothesized that transient culture of adult hepatocytes in 3D as organoids would enhance engraftment of these cells in bioengineered tissues in vivo. Here, we established an organoid system that supports culture of adult human hepatocytes from eight different human adult donors aged 19-49. These organoids support r

ROMO1 and mitochondrial complex II/SDH are required for spare respiratory capacity and glucose homeostasis in mice.

AIMS/HYPOTHESIS: Reactive oxygen species modulator 1 (ROMO1) is a highly conserved inner mitochondrial membrane protein that senses reactive oxygen species and regulates mitochondrial dynamics. ROMO1 is required for mitochondrial fusion in vitro, and silencing ROMO1 increases sensitivity to cell death stimuli. The physiological role of ROMO1 remains unclear. METHODS: To determine the role of Romo1 in vivo, we used gene targeting in mice to ablate Romo1 in the whole mouse and to conditionally knock out Romo1 in the pancreatic beta cell. Mitochondrial functional analyses were performed on isola

Neonatal and pediatric thymic grafts generate similar human T-cell chimerism in humanized mice.

Human immune system (HIS) humanized mice, particularly those incorporating human thymic tissue and hematopoietic stem/progenitor cell (HSPC) co-transplantation, provide a powerful in vivo platform to study human T-cell development, tolerance, and effector function in the context of a full human immune system. The thymus is a lymphoid organ that is indispensable for generating a diverse and self-tolerant T-cell repertoire, and its inclusion in HIS models is essential for generating human major histocompatibility complex-restricted T cells. The bone marrow-liver-thymus (BLT) HIS model is made us

Systemic mitochondrial DNA kinetics after controlled donation after circulatory determination of death lung transplantation do not reflect donor warm ischemic injury: DACMEDAMPs, a prospective multicenter translational study.

Controlled donation after circulatory determination of death has markedly expanded the lung donor pool; however, the biological consequences of donor warm ischemia on mitochondrial injury and systemic danger signaling remain uncertain. Cell-free mitochondrial DNA (cf-mtDNA) has been implicated in ischemia-reperfusion injury, yet its systemic behavior in ventilated human lungs is incompletely characterized. We conducted a prospective, multicenter observational study in matched adult lung transplant recipients from controlled donation after circulatory determination of death and donation after b

Evaluation of Emergency Department Professionals' Knowledge and Attitudes Toward Approaching Brain Death and Potential Organ Donor Recognition: A Cross-Sectional Survey.

OBJECTIVES: Early identification of potential organ donors is essential for expanding the donor pool. Although brain death is typically diagnosed in intensive care units, the recognition of patients with imminent brain death should begin earlier, particularly in the emergency department. In this study, we aimed to evaluate the knowledge and attitudes of emergency department professionals regarding brain death, with a specific focus on the early recognition of imminent brain death. MATERIALS AND METHODS: This cross -sectional study was conducted among emergency department health care professio

Impact of the 2024 Organ Procurement and Transplantation Network Kidney Donor Profile Index Rescoring on Donor Kidney Utilization and Allocation out of Sequence: A one-Year Analysis.

BACKGROUND: In October 2024, the Organ Procurement and Transplantation Network (OPTN) removed hepatitis C virus (HCV) positivity and African American/Black (AA/B) donor race from the Kidney Donor Profile Index (KDPI) calculation. This study evaluates the early impact of this policy change on kidney utilization during its first year. METHODS: We performed a retrospective analysis of OPTN data for organ procurements and adult deceased-donor kidney transplants between November 2023 and October 2025. Donors were stratified by HCV status and race. Kidney utilization and allocation out-of-sequence

A Novel 3D Bioprinting Strategy for Bioengineering of Urethra with Clinical Relevance.

Urethral strictures can cause significant discomfort and progressive urinary tract damage if left untreated. Current reconstructive options, including urethral resection and buccal mucosa grafting, are associated with several limitations such as donor-site morbidity, limited tissue availability, and variable long-term outcomes. To address these challenges, we developed a novel multilayered 3D-bioprinted urethral construct designed to closely mimic the native urethral architecture. Using an Integrated Tissue and Organ Printing System (ITOP) equipped with a rotating mandrel, tubular urethral con

Open Recovery Of The Vascularized Composite Bladder Allograft For Bladder Transplantation.

Prior to the first human bladder transplant (NCT#06337942), a standardized method for vascularized composite bladder allograft (VCBA) recovery compatible with routine donation-after-brain-death (DBD) multi-organ recovery workflow was necessary. A reproducible open surgical technique for VCBA recovery was developed and implemented and is described herein. Under an IRB-approved protocol (IRB#4B-21-5), open VCBA recoveries were performed on 10 DBD donors during standard multi-organ recovery. The primary objective was to develop and refine a reproducible technique to recover the bladder maintainin

Models of the human heart for biomedical research: Opportunities and challenges.

Model systems that mimic human cardiac structure and function are essential for the development of novel diagnostics and effective treatments for cardiovascular diseases. While non-human vertebrate models, from zebrafish to pig, remain vital to cardiovascular research, the translatability of findings to human patients is often limited. Therefore, animal experiments should be supplemented with human model systems, including human induced pluripotent stem cell-derived cells, 3D engineered constructs, and last but not least, native tissue preparations and isolated primary cardiomyocytes. However,

Exploring donor-derived cell-free DNA as a rejection biomarker during uterus transplant follow up.

BACKGROUND: Uterus transplantation (UTx) is a treatment option for women with absolute uterine factor infertility who wish to become pregnant and have a healthy live birth. Tissue biopsy is the gold standard for diagnosing allograft rejection. Circulating donor-derived cell-free DNA (dd-cfDNA) has recently emerged as a non-invasive biomarker with strong predictive value for rejection in solid organ transplants. METHODS: dd-cfDNA quantification by next-generation sequencing (NGS) was evaluated in two UTx patients from transplantation to childbirth. The results were compared with cervical biops

End-stage valvular heart disease requiring heart transplantation: risk profile, surgical modifiers, and myocardial pathology.

BACKGROUND: End-stage valvular heart disease (VHD) constitutes a small but high-risk subset of heart transplant (HTx) recipients, and data describing their natural history and determinants of progression to HTx are limited. We aimed to characterize the clinical, genetic, surgical, and pathological substrates of patients undergoing HTx for end-stage VHD and to assess whether timely cardiac valvular surgery (CVS) mitigates progression to end-stage heart failure. METHODS: We retrospectively screened 961 consecutive recipients who underwent HTx for end-stage heart failure between January 2012 and

Ischemic injury and liver graft congestion increase post-transplant interleukin-6 and tumor necrosis factor-alpha in hepatocellular carcinoma.

Liver transplantation (LT) is an essential treatment for patients with end-stage liver disease (ESLD), including hepatocellular carcinoma (HCC). Post-transplant complications such as venous congestion and tumor recurrence remain challenging, and recent studies suggest that ischemia-reperfusion injury (IRI) and the resulting inflammatory response play a crucial role in these complications. This study was undertaken to investigate the relationship between early post-operative cytokine profiles and the development of venous congestion and tumor recurrence in LT recipients with HCC. Forty-two pati

Fatal Donor-Derived Disseminated Acanthamoeba Infection in a Liver Transplant Recipient.

Acanthamoeba spp. was determined to be the cause of illness after an adult liver transplant recipient died from rapidly progressive encephalitis. Upon autopsy, amebae were identified in multiple organs, including the transplanted liver. The donor had history of kidney transplantation and died of encephalitis presumed to be due to toxoplasmosis. Acanthamoeba was identified in donor brain tissue by indirect immunofluorescence assay. Clinicians assessing prospective solid organ donors with encephalitis should consider Acanthamoeba infection.

A Narrative Review of Advances in Skin Tissue Engineering: From Physiological Architecture to 3D Bioprinting.

The skin is a complex, multilayered organ. Damage from burns, trauma, chronic ulcers, or surgical defects often outstrips the regenerative capacity of conventional healing. Traditional skin grafting, while standard, is limited due to donor site morbidity, lack of appendages, and altered biomechanical/sensory properties. This narrative review traces the evolution of skin repair from autografting to bioengineered substitutes and 3D bioprinting, emphasizing physiological design principles, biomaterials, and translational challenges. The literature synthesis mainly focused on skin architecture and
