After the coronavirus disease 2019 (COVID-19) pandemic, the postacute effects of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection have gradually attracted attention. To precisely evaluate the health status of convalescent patients with COVID-19, we analyzed symptom and proteome data of 442 plasma samples from healthy controls, hospitalized patients, and convalescent patients 6 or 12 months after SARS-CoV-2 infection. Symptoms analysis revealed distinct relationships in convalescent patients. Results of plasma protein expression levels showed that C1QA, C1QB, C2, CFH, CFHR

BACKGROUND: Most epidemiologic studies and clinical testing use single nasal/nasopharyngeal swab (NPS) for respiratory syncytial virus (RSV) detection. Studies document that RSV detection improves if another specimen is added to NPS, but the impact of using multiple specimen types has not been assessed. We quantified RSV detection increase using multiple-specimen testing versus NPS alone.
METHODS: Hospitalized adults aged ≥40 years with acute respiratory illness were prospectively enrolled in 7 US/Canadian hospitals. NPS, saliva, sputa, and acute/convalescent sera were collected and tested.

UNLABELLED: Monoclonal antibodies to SARS-CoV-2 can offer prophylactic and therapeutic protection against severe disease, with particular utility for immunosuppressed and vulnerable populations. With the constant emergence of new variants, understanding the neutralizing potency of monoclonal antibodies to dynamic spike protein epitopes is crucial. We show that a set of VH1-24-derived N-terminal domain (NTD)-directed antibodies, isolated from a convalescent donor early in the pandemic, displayed remarkable neutralization resilience against many Omicron SARS-CoV-2 variants, including BA.2, BA.5,

Post-surgical central nervous system infections (PCNSIs) are a major cause of morbidity, poor functional outcomes and mortality in neurosurgical patients. These infections complicate operations of the CNS or are related to the use of neurosurgical devices or drainage catheters. Gram-negative bacteria, with multiple resistance patterns, are often isolated and these infections are difficult to treat, due to suboptimal antibiotic therapeutic levels in the cerebrospinal fluid (CSF). This is a retrospective study of PCNSIs between 2014 and 2024 in a single center of a tertiary hospital in Thessalon

Hepatitis A virus (HAV) infection in adults is usually self-limited, yet atypical extrahepatic features may complicate the course. We report a 20-year-old male with fever, jaundice, and moderate ascites. Evaluation showed marked hepatocellular injury with direct-predominant hyperbilirubinemia and positive HAV IgM. Imaging demonstrated hepatosplenomegaly, bilateral pleural effusions, and gallbladder wall edema without calculi, consistent with acalculous cholecystitis; there was no encephalopathy. He received conservative management. After initial improvement, a transient mid-course bilirubin re

BACKGROUND/OBJECTIVES: The complex interplay between sarcoidosis and COVID-19 remains an important area of research, since COVID-19 leads to long-term changes in the immune system. However, COVID-19 is often followed by autoimmune diseases, including newly manifesting sarcoidosis. The goal of this study is to characterize CD4+ T cell subsets, playing a pivotal role in the regulation of innate and adaptive immunity, in the peripheral blood of patients with sarcoidosis after COVID-19.
METHODS: The peripheral blood samples from patients with sarcoidosis (n = 61) were studied. We divided patients

Pilonidal sinus is a common pathology of the intergluteal cleft that can develop into abscess or suppuration. This lesion corresponds histologically to a granuloma that organizes around foreign bodies, most often hairs, and fistulizes to the skin through partially epithelialized orifices. If suppuration and abscess develop, treatment is based either on medical treatment combining analgesics, local antiseptics and sometimes antibiotics, or on emergency incision and drainage in the operating room. This is performed in more than 10,000 patients per year in France. Outside of emergencies, elective

Background and objective The righting reaction (RR) from a laterally tilted sitting position is influenced by trunk function and walking ability; however, its relationship with the severity of lower limb motor paralysis in persons with stroke remains unclear. If the characteristics of RR response patterns are clearly identified, they may aid in determining individualized physical therapy approaches. This study aimed to clarify RR patterns according to the severity of lower limb motor paralysis and to explore their potential implications for developing individualized rehabilitation strategies.

YBSW015 is a bispecific antibody that targets severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). A pair of high-affinity parental monoclonal antibodies (B38 and H4) were discovered from the sera of convalescent patients, and YBSW015 was developed based on these two parental antibodies. To support pharmacokinetic research, a simple, specific and reproducible liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the quantifying YBSW015 in human serum. Two signature peptides (SPs) were selected as surrogate analytes for quantification and qual

An 82-year-old woman, who had undergone total gastrectomy for gastric cancer 22 years earlier, fell at home three months prior and underwent surgical repair for a femoral neck fracture. She had been receiving rehabilitation at a convalescent hospital. One week before presentation to our hospital, she experienced another fall, after which she developed decreased oral intake, generalized weakness, and altered mental status. Due to these worsening symptoms, she was transferred from the rehabilitation hospital to our facility. After admission, brain magnetic resonance imaging was performed, which

Vogt-Koyanagi-Harada disease is an autoimmune disease affecting melanocytes. We report a case of Vogt-Koyanagi-Harada disease where the diagnosis was based on the presence of vertigo, vitiligo, and sunset glow fundus without ocular symptoms. The patient was a 68-year-old woman with a history of type 1 diabetes mellitus who developed vertigo three months prior and vitiligo on the face and arms two months prior. An ophthalmic examination revealed sunset glow fundus with numerous depigmented chorioretinal scars. Keratic precipitates were observed; however, no cells were observed in the anterior c

Certain strains of Influenza A virus (IAV), a primary cause of influenza, can lead to pneumonia. Patients recovering from influenza pneumonia may experience physical discomfort akin to post-acute sequelae of COVID-19 (PASC). Despite extensive clinical research on viral pneumonia during convalescence, animal model studies are scarce, highlighting the need for a reliable model for pharmaceutical research. In this study, BALB/c mice were divided into three groups: NC (control), MC (infected with IAV), and Model (treated with oseltamivir post-infection for five days). A fatigue model was then indu

BACKGROUND: Although guidelines for COVID-19 additional vaccination strategies generally prioritise people with advanced HIV infection, recommendations vary globally, with some countries recommending an annual vaccination for all people with HIV (PWH), while others restrict this to PWH with a CD4+ T-cell count < 200 cells per µL.
METHODS: We conducted a prospective cohort study in 448 adult PWH. The primary outcome was the SARS-CoV-2 spike (S1)-specific IgG antibody level at 1, 6, 12, 18, and 24 months after completing a primary COVID-19 vaccination series (two doses of BNT162b2, mRNA-1273, o

Dengue illness follows three phases: febrile, critical, and convalescent. Coinfection and hyperinflammatory response are common causes of the persistence of fever beyond the febrile phase of dengue. However, finding both these causes together in severe dengue cases is rarely reported in the literature. We present three cases of severe dengue who presented with persistent fever. The first case is of a 14-year-old girl who presented with dengue shock and culture-proven Acinetobacter sepsis. Later she was diagnosed with a hyperinflammatory response to infection and improved after intravenous immu

The 2024 Clinical Practice Guidelines for Influenza of the World Health Organization (WHO) are an updated version of the 2022 Guidelines for the Clinical Management of Severe Illness from Influenza Virus Infections. The 2024 guidelines redefine risk factors for severe influenza, which is crucial for understanding treatment recommendations. The new guidelines not only focus on the clinical management of severe influenza and individuals at high risk of developing severe illness but also cover recommendations for managing non-severe influenza cases. Additionally, they provide guidance on using an

BACKGROUND: Humoral immune response against the pre-fusion (pre-F) conformation of respiratory syncytial virus (RSV) F protein has been proposed to play a protective role against infection. An RSV pre-F maternal vaccine has been recently approved in several countries to protect young infants against RSV. We aimed to assess serum IgG titers against the pre-F and post-F conformations of RSV F protein and their association with life-threatening RSV disease (LTD) in previously healthy infants.
METHODS: A prospective cohort study including hospitalized infants <12 months with a first RSV infection

SARS-CoV-2 ancestral strain-induced immune imprinting poses great challenges to updating vaccines for new variants. Studies showed that repeated Omicron exposures could override immune imprinting induced by inactivated vaccines but not mRNA vaccines, a disparity yet to be understood. Here, we analyzed the immune imprinting alleviation in inactivated vaccine (CoronaVac) cohorts after a long-term period following breakthrough infections (BTI). We observed in CoronaVac-vaccinated individuals who experienced BA.5/BF.7 BTI, the proportion of Omicron-specific memory B cells (MBCs) substantially incr

OBJECTIVES: The respiratory tract is the portal of entry for the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Although a variety of respiratory pathogens other than SARS-CoV-2 have been associated with severe cases of COVID-19 disease, the dynamics of the upper respiratory microbiota during disease the course of disease, and how they impact disease manifestation, remain uncertain.
METHODS: We collected 349 longitudinal upper respiratory samples from a cohort of 65 COVID-19 patients (cohort 1), 28 samples from 28 recovered COVID-19 patients (cohort 2), and 59 samples from 59 h

Direct acting antivirals (DAAs) represent critical tools for combating severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants of concern (VOCs) that have escaped vaccine-elicited spike-based immunity and future coronaviruses with pandemic potential. Here, we used bioluminescence imaging to evaluate therapeutic efficacy of DAAs that target SARS-CoV-2 RNA-dependent RNA polymerase (favipiravir, molnupiravir) or main protease (nirmatrelvir) against Delta or Omicron VOCs in K18-hACE2 mice. Nirmatrelvir displayed the best efficacy followed by molnupiravir and favipiravir in suppressin

Neutralizing antibodies (NAbs) are elicited after infection and vaccination and have been well studied. However, their antibody-dependent cellular cytotoxicity (ADCC) functionality is still poorly characterized. Here, we investigated ADCC activity in convalescent sera from infected patients with wild-type (WT) severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) or omicron variant compared with three coronavirus disease 2019 (COVID-19) vaccine platforms and postvaccination breakthrough infection (BTI). We analyzed ADCC activity targeting SARS-CoV-2 spike (S) and nucleocapsid (N) protei
