State of the global tuberculosis epidemic: burden, progress towards WHO End TB Strategy targets and systemic challenges.

Mycobacterium tuberculosis remains the leading cause of death from a single infectious pathogen globally despite decades of effective chemotherapy. In 2024, an estimated 10·7 million people developed tuberculosis, including approximately 620 000 people living with HIV (PLHIV), and tuberculosis caused an estimated 1·23 million deaths overall, including approximately 150 000 deaths among PLHIV. Men accounted for more than half of the cases, and children represented a substantial burden, reflecting ongoing transmission and diagnostic gaps. Approximately a quarter of the world's population has bee

Real-world substance use in body dysmorphic disorder: An ecological momentary assessment study of motives, contexts, and suicidal ideation.

Body dysmorphic disorder (BDD) frequently co-occurs with substance use, yet little is known about real-world patterns, motives, and contexts of use. This study characterized baseline substance use and examined day-to-day alcohol and cannabis use across 28 days of ecological momentary assessment (EMA) in adults with moderate to severe BDD with recent substance use or suicidal ideation. Eighty-seven participants (73.6% women; mean age 29.4 years, SD = 9.0, range 18-55) completed diagnostic interviews, self-report measures, and EMA surveys. At baseline, more than half (51.7%) met criteria for a c

Astrocyte molecular rhythm disruption in nucleus accumbens promotes increased binge-like drinking in mice.

Alcohol misuse is a leading cause of preventable death worldwide. Chronic alcohol is associated with disrupted circadian rhythms, yet molecular mechanisms linking circadian rhythm dysregulation and alcohol consumption are poorly understood. Current FDA-approved treatments for alcohol use disorder (AUD) do not target molecular rhythms or sleep-wake cycles. Mammalian circadian rhythms are regulated by transcription-translation feedback loops that regulate expression of 'clock genes' (e.g., Arntl encoding for BMAL1). Both human and rodent studies demonstrate associations between clock gene varian

COMT rs4680 polymorphism modulates openness to experience in women with alcohol use disorder.

PURPOSE: The objective of the study was to examine the association between COMT rs4680 genotypes, alcohol use disorder diagnosis, and differences in Five-Factor personality traits. METHODS: We examined 213 adult women in total, including 101 with a diagnosis of alcohol use disorder (AUD) and 112 non-addicted controls. All participants completed the NEO Five-Factor Inventory (NEO-FFI) and were genotyped for the COMT rs4680 polymorphism using real-time PCR. Group comparisons and genotype × diagnosis interactions were analyzed using ANOVA and post hoc tests. RESULTS: Women with AUD scored signi

Migrant women, alcohol and drinking experiences: a narrative review.

Migrant women's alcohol use has been understudied despite evidence that gender, culture, and migration shape drinking experiences. This review synthesizes qualitative research on how migrant women experience host-country drinking cultures, their gendered drinking experiences and factors influencing their alcohol use. The review was conducted across five databases (ProQuest, Dimensions, Scopus, PsycINFO and PubMed). Inclusion criteria were peer-reviewed qualitative studies published since 2000 examining migrant women's experiences with alcohol and drinking cultures. Twenty-two articles were inc

Subjective alcohol response across drinking contexts: a systematic review with bayesian meta-analysis.

RATIONALE: Alcohol subjective response (SR) predicts drinking behaviour and risk for alcohol use disorder, but there is limited consensus on how SR differs across contexts, research settings, and alcohol problem severity. OBJECTIVES: This systematic review and meta-analysis examined how alcohol SR differs across (1) commercial (e.g., bar/club/restaurant) versus non-commercial (e.g., home) settings and (2) social versus solitary drinking contexts, and if these effects are moderated by (3) laboratory versus naturalistic study setting and (4) non-problem versus problem alcohol use. METHODS: Stu

Survivorship Care Before Completion of Definitive Radiotherapy and Oncologic Outcomes in Head and Neck Cancer.

IMPORTANCE: Across cancer populations, a growing and aging survivor population combined with increasing use of multimodal therapy has expanded the need for survivorship care, particularly in head and neck cancer (HNC). Although survivorship begins at diagnosis, survivorship care is often emphasized after treatment completion, and whether earlier integration during treatment is associated with adherence to treatment and survival remains unclear. OBJECTIVE: To evaluate whether early multidisciplinary survivorship care before definitive radiotherapy completion is associated with adherence and su

Social jetlag and alcohol use in adolescents: A random-intercept cross-lagged panel analysis.

OBJECTIVE: Alcohol use and social jetlag (i.e., the misalignment between weekday and weekend sleep timing) frequently co-occur during adolescence, yet their temporal dynamics across different timescales remain underexamined. This study assessed bidirectional relationships between alcohol consumption and social jetlag at the person, 4-month, and weekly levels. METHOD: U.S. high school students (N = 302, analytic n = 297; Mage = 16.21 years) completed three 21-day assessment bursts, each comprising three consecutive weekly surveys and timeline follow-back assessments. Bursts were separated by 4

Risky sexual behaviors and associated factors among adolescent girls and young women in the context of HIV prevention pre-exposure prophylaxis in Tanzania: Analysis of the Tanzania HIV Impact Survey 2022-2023.

Risky sexual behavior contributes to HIV acquisition among adolescent girls and young women (AGYW) in Sub-Saharan Africa (SSA), including Tanzania. However, nationally representative evidence on its prevalence and associated factors remains limited. This study examines the prevalence and associated factors of risky sexual behavior among AGYW aged 15-24 years in Tanzania, in the context of the expanding Pre-Exposure Prophylaxis (PrEP) provision. A secondary analysis of nationally representative data from the 2022-2023 Tanzania HIV Impact Survey (THIS) was conducted among 3,669 sexually active A

Racial/Ethnic and Sexual Identity Differences in the Associations Between School Bullying and Cyberbullying Victimization and Types of Substance Use.

Bullying victimization is found to be associated with substance use, and this association is relevant to racial/ethnic minority and sexual minority (lesbian, gay, and bisexual) adolescents. However, few studies have focused on the role of intersectionality in the linkage between bullying victimization and substance use. The current research explores the relationship between school- and cyberbullying victimization and various types of substance use among a racially/ethnically diverse sample of heterosexual, lesbian, gay, and bisexual adolescents. The 2023 Youth Risk Behavior Surveillance System

Estimated incidence and number of new cases with prenatal alcohol exposure and fetal alcohol spectrum disorders in Finland 1990-2025.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) arising from prenatal alcohol exposure (PAE) are the leading preventable cause of neurobehavioral disorders. Early pregnancy is particularly vulnerable to ethanol toxicity, yet alcohol use often continues until pregnancy recognition. In Finland, national incidence estimates of PAE and FASD remain limited. OBJECTIVE: To estimate the incidence of any PAE and heavy PAE in Finland between 1990 and 2025, and to model the annual number of children with FASD born in Finland. METHODS: We developed a mathematical modelling framework integrating stud

Dorsomedial striatum calcium permeable AMPA receptors in the development of aversion-resistant alcohol drinking.

RATIONALE: Alcohol use disorder is defined by drinking alcohol despite knowledge of negative consequences, often referred to as aversion-resistant drinking (The dorsomedial (DMS) and dorsolateral striatum (DLS) are necessary for goal-directed and habitual action selection, respectively. Leading hypotheses posit that once drug use becomes compulsive, DMS dependence degrades while DLS dependence increases. This shift may be mediated by changes in synaptic weights from glutamatergic inputs. OBJECTIVES: Using a combination of western-blot, micro-injections, and ex-vivo electrophysiology in male a

Developing and Validating a Patient-Centered Measure of Well-Being in Bipolar Disorder: The WELL-B.

OBJECTIVES: Traditional measures of well-being in bipolar disorder (BD) often equate well-being with the absence of mood symptoms, overlooking strengths-based indicators like acceptance, agency, and positive emotion that individuals with BD identify as central to their lived experience of wellness. To address this misalignment, we developed and psychometrically validated a new well-being measure, the Well-being Evaluation for Living with Bipolar (WELL-B), grounded in the concept of thriving. METHODS: The WELL-B was co-designed with individuals with BD and caregivers through a collaborative le

Neighborhood violence, visible drug activity, and substance use among urban youth: Mental health and community factors.

BACKGROUND: Adolescent substance use and exposure to neighborhood violence frequently co-occur in U.S. urban settings, particularly among racially minoritized and low-income youth. Despite shared exposure to environmental risk, some adolescents report low to no substance use. Guided by the adverse community experiences and resilience framework, this study identified classes of neighborhood risk and substance use among urban adolescents and examined whether emotional, behavioral, community, and demographic factors differentiated profile membership. METHODS: Cross-sectional survey data were col

Sports-team participation and risk behaviors among US high school students: behavior-specific and sex-heterogeneous associations in the national YRBS, 2011-2023.

Sports-team participation is often regarded as a favorable adolescent context, but whether its associations are consistent across distinct risk behaviors and across sex remains unresolved. We conducted a repeated cross-sectional analysis of seven U. S. National Youth Risk Behavior Survey cycles (2011-2023; 110,409 records) using survey-weighted regression and marginal standardization. Participation in one or more sports teams was associated with higher adjusted prevalence of alcohol use (PD, +4.46 percentage points; 95% CI, +3.38 to +5.55) and physical fighting (+2.53; 95% CI, +1.62 to +3.43),

HPV18 DNA detection and pathological laryngopharyngeal reflux in a young woman with glottic squamous cell carcinoma in situ: a brief report.

Laryngeal cancer predominantly affects elderly males with long-term smoking and alcohol histories, and its occurrence in young females is extremely rare. We report a 26-year-old woman with glottic squamous cell carcinoma in situ who presented with persistent hoarseness. She had no history of smoking, alcohol use, or radiation exposure. Upon evaluation, high-risk HPV18 DNA was detected in FFPE tumor tissue by Lcn PCR, and 24-hour MII-pH monitoring indicated pathological laryngopharyngeal reflux. Targeted sequencing of 535 genes showed no driver mutations, tumor mutation burden elevation, or mic

The metabolic transition of liver cancer: a 34-year analysis of global burden across the 10 most populous countries, 1990-2023.

BACKGROUND: The global burden of liver cancer continues to rise, yet comprehensive assessments of trends in behavioral and metabolic risk factor-attributable mortality and disability-adjusted life years (DALYs) remain limited. We aimed to quantify these trends across the world's 10 most populous countries from 1990 to 2023. METHODS: Using data from the Global Burden of Disease Study 2023, we extracted age-standardized mortality rates (ASMRs) and disability-adjusted life years (DALYs) for primary liver cancer attributable to seven risk factors-two composite exposures (behavioral risks and meta

Expression Profiling of a Recombinant Inbred Line Panel Identifies the dod-19 Gene as a Regulator of Ethanol Response Behaviors in C. elegans.

BACKGROUND: Variation in gene expression is hypothesized to be a major source of phenotypic variation underlying the liability for alcohol misuse. Caenorhabditis elegans (C. elegans) has proven to be a valuable model in the identification of genes that modify behavioral responses to alcohol. Genetic variation in wild strains represents allelic differences that are tolerated by natural selection pressures and, therefore, are more likely to model the allelic differences that are present in humans compared with laboratory-generated mutations. METHODS: We examined the influence of natural alleles
