Establishment of In Vivo Mouse Models to Study Salmonella-Induced Chronic Infection and Gastrointestinal Cancer.

Salmonella infection increases the risk of chronic diseases, e.g., inflammatory bowel diseases, colon cancer, and gallbladder cancer. However, the mechanisms leading to the development of these conditions are still not completely understood. Here, we describe the methods to establish in vivo mouse models to study Salmonella-induced chronic infection and colon cancer. These models will allow us to investigate the initiation and progression of diseases.

Functionalization of Flavonoids with Lipases.

Flavonoids, valued for their health benefits, face challenges due to low solubility. Enzymatic acylation offers promise in enhancing properties, with the use of lipase B sourced from Candida antarctica (N435). This study has the aim to improve naringin solubility via esterification using as acyl donors short- and medium-chain fatty acids (vinyl acetate, propionate, and laurate) catalyzed by N435. Lipases play a crucial role as catalysts, facilitating the acylation process. Our study encompasses executing acylation and characterizing products. This process significantly enhances aqueous and org

Effects of Organic Substrate Concentrations on Biological Mn Oxidation and Reduction under Aerobic Conditions.

Biological Mn oxidation mediated by manganese-oxidizing bacteria (MnOB) plays a central role in Mn cycling in natural and engineered systems. Although most MnOB are heterotrophic and frequently associated with oligotrophic environments, the quantitative relationship between organic substrate concentrations and Mn oxidation activity remains unclear. We herein systematically evaluated the effects of organic substrate concentrations on MnOB activity using a down-flow hanging sponge (DHS) reactor (0-1,000 mg COD L-1), batch experiments with an enriched biomass, and the pure MnOB strain Pseudomonas

ER quality control of Crumbs2 and Notch proteins by GXYLT1-dependent xylosyl elongation of O-glucose glycans.

Protein O-glucosyltransferase 1 (POGLUT1/Rumi) transfers an O-linked glucose (O-Glc) monosaccharide from UDP-Glc to a serine residue in epidermal growth factor (EGF)-like repeats with a specific consensus sequence. In mammals, GXYLT1 and GXYLT2 transfer xylose to this O-Glc monosaccharide. Previous studies showed that Poglut1 deletion in mice leads to embryonic lethality with defects in crumbs family member 2 (CRB2) trafficking and NOTCH1 signaling. Both CRB2 and Notch receptors have multiple EGF repeats with the O-Glc modification site. However, the roles of xylosyl elongation of O-Glc in pro

Beyond the Nurse's Office: How School Nurses can Build Trust and Transform Perceptions.

Although nursing is consistently ranked among the most trusted professions, its role is often overlooked or misunderstood. School nurses earn credibility and trust through daily practice: credentials establish expertise, while professional conduct makes it visible. By presenting themselves professionally, school nurses affirm their role and build stronger relationships with staff, students, and families, making them the first point of contact for health questions or crises. Using the three elements of professional comportment-actions, presence, and words-this article offers practical strategie

Lysine butyrylation modulates central carbon metabolism and photosynthesis in cyanobacteria.

Cyanobacteria, evolutionarily ancient photoautotrophs, serve as key model organisms for studying diverse biological processes. Lysine butyrylation (Kbu) is an emerging post-translational modification implicated in diverse biological processes from bacteria to mammals. However, the distribution and functional significance of Kbu in cyanobacteria have yet to be elucidated. In this study, we present the first proteome-wide analysis of Kbu in the model cyanobacterium Synechococcus sp. strain PCC 7002 using peptide pre-fractionation, butyrylation antibody enrichment and high-accuracy mass spectrome

The razABCD regulatory island links azalomycin F biosynthesis in Streptomyces iranensis to orsellinic acid production in the fungus Aspergillus nidulans.

Streptomyces iranensis induces the production of orsellinic acid and a range of its derivatives in the fungus Aspergillus nidulans. The arginoketide azalomycin F was identified as the bacterial signaling molecule that activates otherwise silent biosynthetic gene clusters (BGCs) in diverse fungi. To date, however, little is known about the regulatory mechanisms governing the azalomycin F BGC, beyond the requirement of the regulatory gene bldA. Here, building on the known role of bldA, we report the discovery of a regulatory island, razABCD, encoding four LuxR-family transcriptional regulators.

Optimized Detection of Acidic ADP-Ribosylation by PARP14 in Vitro Using Multi-Modal Mass Spectrometry Reveals Putative Autoregulatory Patches.

ADP-ribosylation is a highly dynamic process that is increasingly linked to a diverse set of cellular functions and pathophysiological outcomes. The modification of target substrates (e.g., proteins, DNA, RNA) involves ligation to an array of different chemical moieties. Recent work has shown that the ester bonds formed between ribose and aspartate (Asp)/glutamate (Glu) residues are particularly labile. A common strategy to overcome this lability is the replacement of the ADP-ribose (ADPr) by hydroxylamine. Attachment of hydroxamic acid (HA) permanently shifts the mass of labeled Asp/Glu resid

The role of pharmacogenomics in effective pain management with opioids.

Opioids are commonly prescribed as standards of care for various types of pain. Multiple pathways can impact pain response, and understanding medication metabolism is imperative to achieving optimal analgesic response. Pharmacogenomics (PGx) can assist with understanding interindividual variability and aid in safe medication prescribing. Genetic variants in pharmacokinetic and pharmacodynamic genes have been evaluated, and literature generally favors use of pharmacokinetic associations in clinical practice. Tramadol and codeine have the most literature to support integration of PGx into routin

Polarity dependent Hippo-Yap regulation controls cortical development.

Epithelial polarity is established by partitioning the apical and basal domains within the cell. Antagonistic inhibitions between the apical and basal polarity complex proteins segregate them into their designated domains. Failure to achieve this causes developmental disorders and cancer. While the biochemical basis of this antagonism has been identified, the complexity of context-dependent cell-cell interactions obscures its cellular underpinnings. Here, through a series of genetic rescues in a murine model of brain malformation, the cellular mechanisms underlying this antagonism are uncovere

Automated Check-Ins to Care Assistant? The Use of Kiosks in the School Health Room.

From airlines and hotels to fast food restaurants, kiosks play a role in many businesses, replacing what was once a human helping to make a reservation, check in, or place an order. Therefore, it is no surprise that kiosks are also being used in healthcare settings. Kiosks in the school health office, with the appropriate school population, can be a valuable school nurse tool. Kiosks can be used for student check-in and tracking as well as a means for data collection and report generation. However, there are potential drawbacks and areas where this technology could be improved and further eval

Advancing chemical reaction prediction in data-scarce drug discovery with active and geometric deep learning.

The success of generative molecular design depends on the synthetic accessibility of the molecules it creates. While computational models can help predict chemical reactions, their current effectiveness is limited by a lack of reference data and the difficulty of accurately predicting reaction regioselectivity. Here, to address this, we present an integrated closed-loop workflow that combines efficient reaction data acquisition with high-accuracy deep learning. We developed an active learning strategy, driven by a tree-based ensemble, to guide the acquisition of the most informative laboratory

Grandparent involvement and children's emotional and behavioral problems: the mediating role of social competence and the moderating role of family functioning.

This study examined whether social competence accounts for part of the association between grandparent involvement and children's emotional and behavioral problems, and whether family functioning modifies the strength of the association between grandparent involvement and social adaptability. Data were collected from 729 parents of children aged 3-7 years in China across three time points. Participants completed measures of grandparent involvement, family functioning, social adaptability, and emotional and behavioral problems. Moderated mediation analysis revealed that grandparent involvement

Consensus recommendations on age-related sarcopenia in South Asia: A nutrition-focused approach.

Age-related sarcopenia, defined as the progressive decline in muscle function, strength, and mass with age, is a growing health concern in South Asian countries (Bangladesh, India, and Sri Lanka) due to their rapidly aging populations. It tends to appear earlier, is frequently overlooked, and worsens after age 60. Despite its prevalence, standardized tools for screening and assessing sarcopenia and dietary status are lacking. Since diet quality is a modifiable risk factor, nutritional strategies play a key role in prevention and management. However, international dietary guidelines are often u

A comprehensive study on cervical cancer diagnosis using deep learning and artificial intelligence techniques.

Cervical cancer is a significant health problem across the globe, especially in nations with low or middle incomes, where access to early screening is limited and contributes to high mortality rates. Traditional diagnostic methods, including Papanicolaou smears (Pap smears) and Human Papillomavirus testing (HPV testing), often suffer from accessibility constraints, lower accuracy, and scalability issues. Recent advancements in artificial intelligence (AI) and Machine Learning (ML) have exhibited significant efficacy by improving detection accuracy and streamlining the diagnostic procedure. Thi

TRPML3 and TRPML1/3 differentially regulate unconventional MIF secretion through TMED10 and secretory autophagy.

The intracellular Ca2+ channel TRPML3 regulates autophagy by multimerization in a phosphoinositide (PIP)-dependent manner; however, its role in other membrane dynamics remains unclear. Here, we report that TRPML3 regulates type III unconventional protein secretion (UcPS) through two distinct pathways: TMED10-channeled UcPS and secretory autophagy. TRPML3 interacted with TMED10 and macrophage migration inhibitory factor (MIF), a signal sequence-lacking pleiotropic cytokine whose secretion mechanism has remained undefined. We found that cytosolic MIF is transported to membrane-enclosed compartme

Hormone Replacement Therapy Is Associated with Increased Risk of Anterior Cruciate Ligament Tear in Midlife Women in the United States.

BACKGROUND: Across the female lifespan, the incidence of anterior cruciate ligament (ACL) tears has demonstrated a bimodal age distribution, and this may be related to fluctuating lifetime estrogen. However, this relationship-and the role of hormone replacement therapy (HRT) and its potentially modifying effect on ACL tear risk-has been underreported. The purpose of this study was to (1) characterize the ACL tear risk pattern in women compared with men in a national database and (2) assess the relationship between HRT and the risk of ACL tear among midlife women. METHODS: A retrospective coho
