Acute E-Cigarette Aerosol Condensate Exposure Disrupts the Transcriptome and Proteome Profiles of Human Bronchial Epithelial BEAS-2B Cells.

The growing popularity of electronic cigarettes (e-cigarettes) necessitates a better understanding of their biological effects. In this study, we aimed to evaluate the effects of e-cigarette aerosol condensates generated from either e-cigarette carrier liquid alone or with e-cigarette liquid with nicotine and flavor on bronchial epithelial cells. BEAS-2B cells were exposed to e-cigarettes for 24 h, and transcriptional and proteomic profiling, including assessment of protein modifications, was performed. Additionally, cell-based assays were used to evaluate mitochondrial function, rate of prote

Secondary Cardiac Rhabdomyosarcoma: A Unique Presentation.

Cardiac tumors are rare and complex, with malignant forms carrying a poor prognosis. These tumors are classified as either primary or secondary, with the latter, including metastases, being more prevalent. Rhabdomyosarcoma, a rare but aggressive soft tissue sarcoma, can occasionally metastasize to the heart. Here, we report the case of a 75-year-old male with metastatic rhabdomyosarcoma originating in the fibular muscles, extending to the left atrium, left ventricle, and bronchi. The patient presented with acute dyspnea, palpitations, and cardiogenic shock. Imaging revealed a large intracardia

3-dimensional bronchial deformation analysis in left upper lobectomy: inferior pulmonary ligament dissection versus preservation.

BACKGROUND: Upper lobectomy was the independent factor that caused a greater reduction in postoperative lung function. This was deducible from the upward movement of residual lobes caused by releasing inferior pulmonary ligament. In case with left upper lobectomy, more dramatic postoperative pulmonary changes were developed compared with after right upper lobectomy, due to larger volume it has. We aim to analyze the bronchial deformation associated with the postoperative lung function in the setting of inferior pulmonary ligament dissection. METHODS: Patients who underwent left upper lobectom

Azygos vein arch injury caused by an ultrasonic scalpel during lung cancer surgery.

Unexpected azygos vein injury during thoracoscopic lung resection is rare. Here, we describe a case of azygos vein arch injury caused by an ultrasonic scalpel. The patient underwent segmentectomy for right upper lobe lung cancer. The ultrasonic scalpel was activated while its blade tip was inadvertently grasping the vascular wall of the azygos vein arch during management of bleeding from the pulmonary artery stump, resulting in injury to the azygos vein arch. Because the injury site was temporarily sealed, it was not initially detected. When the right upper lobe was retracted ventrally to diss

Bronchovascular Tree-Guided Weakly Supervised Learning Method for Pulmonary Segment Segmentation.

Pulmonary segment segmentation is crucial for cancer localization and surgical planning. The voxel-wise annotation of pulmonary segments is laborious, as the boundaries between segments are indistinguishable. To this end, we propose Anatomy-Hierarchy Supervised Learning (AHSL), a novel weakly supervised learning method. It incorporates anatomical priors into pulmonary segment segmentation through hierarchical supervision. Specifically, AHSL adopts a dual-level supervision paradigm at both the lobe and segment levels. At the lobe level, supervision constrains pulmonary segments to remain within

N-nitrosodimethylamine induces cytochrome P450 activation but not epithelial remodeling in well-differentiated human airway epithelial cells.

N-nitrosodimethylamine (NDMA) is a volatile environmental contaminant formed from diverse precursor chemicals widely present in byproducts of industrial processes, personal care products, and pharmaceuticals. Therefore, NDMA is frequently detected in both indoor and outdoor air. However, its potential risk from inhalation exposure has been largely underestimated and the mechanisms underlying its toxicity remain insufficiently studied. To address this knowledge gap, we investigated the effects of NDMA on primary human bronchial epithelial (HBE) cells differentiated in air-liquid interface cultu

U2AF regulates the translation and localization of nuclear-encoded mitochondrial mRNAs.

The mechanisms underlying molecular targeting to mitochondria remain enigmatic, yet this process is crucial for normal cellular function. The RNA-binding proteins U2AF1 and U2AF2 form a heterodimer (U2AF) that shuttles between the nucleus and cytoplasm, regulating splicing in the nucleus and translation in the cytoplasm. Our study in human bronchial epithelial cells (HBECs) identifies an unexpected role for U2AF in mitochondrial function. We demonstrate that U2AF interacts with nuclear-encoded mitochondrial (NE-mt) mRNAs and proteins, inhibits translation, localizes to the mitochondria, and re

Simultaneous Ligation of Vessels During Robotic Right Upper Lobectomy.

BACKGROUND: Herein, we reported a new technique as the simultaneous ligation of vessels after bronchus section that could facilitate the achievement of Robotic-Assisted Thoracic Surgery (RATS) Right Upper Lobectomy (RUL). METHODS: The clinical data of all consecutive patients undergoing RATS RUL for management of early-stage Non-Small Cell Lung Cancer (NSCLC) were included in the study. Patients were divided into 2 groups based on whether RUL was performed by simultaneous ligation of vessels (Simultaneous Ligation Group) or by traditional single isolation-ligation technique (Control group). Deposition Patterns of Carbon Black Nanoparticles in Pulmonary Lobes and Airway Generations: Implications for Lung Remodeling.

Carbon black nanoparticles (CBNPs) pose respiratory risks due to occupational and environmental exposure, yet their spatially heterogeneous effects on lung remodeling remain unclear. We quantified remodeling patterns using high-resolution computerized tomography (HRCT) in 69 carbon black packers and 107 controls, with carbon content in airway macrophages (CCAM) serving as a bioeffective dosimeter. CBNP-exposed workers exhibited elevated mean lung density in the left (β = 7.75 HU) and right upper lobe (β = 6.22 HU, all p values ≤ 0.047), showing clear dose-dependent trends with CBNP internal do

Effects of Airway Generation Depth on Particle Deposition Behaviour in the Main Respiratory Airways-A Computational Modelling Study.

PURPOSE: Effective inhaled drug delivery depends on formulation, device, and a clear understanding of aerosol transport in the respiratory tract. This study explores how lower airway anatomical complexity affects airflow and particle deposition in the main respiratory airways. METHODS: Seven 3D bronchial models with varying airway generational depths were developed. Numerical simulations were conducted using the discrete phase model (DPM) to simulate aerosol transport and deposition under transient inspiratory flows at inhalation peak flow rates of 60 and 120 L/min. RESULTS: Our results show

Host transcriptomic analysis reveals a defective intracellular environment that limits SARS-CoV-2 replication in CFTR-deficient airway epithelium.

Cystic fibrosis (CF) is characterized by chronic airway inflammation, yet clinical observations have revealed more favorable COVID-19 outcomes than originally predicted. Several studies demonstrated a significant decrease of SARS-CoV-2 replication in CF-mutated bronchial cells suggesting that CFTR dysfunction may interfere with viral replication, though the underlying mechanisms remain unclear. To elucidate these mechanisms we performed transcriptomic profiling of SARS-CoV-2-infected bronchial epithelial cells with wild-type (WT) or mutated CFTR, using both immortalized and primary airway mode

Discovery and management of accessory hemidiaphragm during video-assisted thoracoscopic segmentectomy for pulmonary nodule: a case report.

An accessory hemidiaphragm is a rare congenital intrathoracic malformation characterized by a fibromuscular diaphragm located superior to the normal diaphragm, typically featuring a central hiatus through which pulmonary vessels and bronchi traverse. This anomaly is often associated with scimitar syndrome. We present a unique case of an isolated accessory hemidiaphragm-devoid of the characteristic vascular anomalies typical of scimitar syndrome-discovered incidentally during thoracoscopic surgery performed for a pulmonary nodule. This report elaborates the rationale for performing a concurrent

Bronchopulmonary Segmentation in Humans and Dogs: A Comparative Narrative Review Highlighting Anatomical Gaps and Surgical Implications.

Bronchopulmonary segmentation (BS) is an anatomical concept, poorly described in veterinary medicine, and involve a specific segmental bronchus, a corresponding segmental artery, segmental veins draining into intersegmental veins, and intersegmental septa. While pioneer studies were comparative in humans and domestic animals, this research path diverged decades ago, and the field has since lacked a proper, functional categorization of canine lungs into bronchial segments. Veterinary anatomical nomenclature does recognize this segmentation, but the classification is traditionally based on terti

Protocol for visualization and quantitative analysis of murine lung immunological architecture using iBALT-UNMAP following bacterial challenge.

The spatial distribution of adaptive immune cells is a critical determinant of the quality and magnitude of mucosal immunity. Here, we present a protocol for tyramide signal amplification (TSA) immunofluorescence that preserves the lung architecture and quantifies inducible bronchus-associated lymphoid tissue (iBALT) following murine Bordetella bronchiseptica infection. Starting from paraffin-embedded lungs, we describe steps for staining, imaging, and morphometric analysis. This protocol can be optimized for other pathogens and experimental settings to allow mapping of spatial immune response

Pulmonary Sequestration in the Guise of Pneumonia: Clues to a Hidden Entity.

Pulmonary sequestration is an important yet often overlooked congenital lung malformation in adolescent medicine characterized by dysplastic, non-functioning pulmonary tissue that lacks communication with the tracheobronchial tree and derives its blood supply from an anomalous systemic artery. We report the case of a 12-year-old female patient referred to our institute with a six-month history of intermittent fever, cough, and chest pain, initially managed as a case of right-sided pneumonia elsewhere. Contrast-enhanced computed tomography of the thorax revealed a cavitary soft tissue lesion in

World's Largest Single-Surgeon Experience in Robotic Resection of Pulmonary and Bronchial Carcinoid Tumors.

Carcinoid tumors of the lung and bronchus are relatively rare. This is a consecutive (non-selected) series from a validated prospective database of patients from one surgeon (RJC). From July 1996 to March 2026 one thoracic surgeon (RJC) performed 19,425 operations and 243 patients (1.3%) had a carcinoid tumor. In February 2009 we started performing robotic surgery and have performed 3,100 robotic operations as of March 2026, of which 134 (4.3%) patients had a carcinoid tumor resected robotically. Eighty-one patients (60%) underwent lobectomy, 17 (13%) had a sleeve resection of the airway, and
