The Cyclophilin Effector TaCYPE1 From the Tomato Leaf Miner Tuta absoluta Suppresses Plant Immunity by Targeting Host RNA-Binding Proteins.

The leaf-mining insect Tuta absoluta is a highly destructive pest that deploys candidate effector proteins to modulate host defences; however, the molecular mechanisms underlying this manipulation remain poorly understood. In this study, we characterise TaCYPE1, a predicted secreted cyclophilin-like effector, and demonstrate its role in promoting insect virulence. Transient expression of TaCYPE1 in Nicotiana benthamiana suppresses pattern-triggered immunity (PTI), including INF1-induced cell death, reactive oxygen species (ROS) burst, and defence-related gene expression. Notably, this immune s

Tumour-agnostic immunotherapy: aligning biological rationale, biomarker precision, and real-world implementation.

Tumour-agnostic immunotherapy has reshaped precision oncology by showing that treatment benefit can, in selected settings, be predicted by a shared biological state rather than anatomical origin. Microsatellite instability-high or mismatch repair-deficient cancer is the clearest example, with durable responses across diverse malignancies. By contrast, broader biomarkers show that pan-cancer applicability cannot be inferred from a measurable molecular feature alone. Tumour lineage, antigen presentation, immune microenvironment, prior therapy, and health-system context all influence whether a tu

The Genetic and Epigenetic Mechanisms of mtDNA in Regulation of Inflammation and Metabolism.

Mitochondria serve as the primary cellular powerhouses, generating ATP through oxidative phosphorylation (OXPHOS) to sustain essential cellular processes. Beyond energy production, mitochondria function as critical regulators of metabolic homeostasis, intracellular signaling networks, and programmed cell death pathways. The mitochondrial genome comprises 37 genes encoding 13 OXPHOS subunits, 22 transfer RNAs, and 2 ribosomal RNAs, all transcribed and translated within the organelle. Mitochondrial DNA integrity becomes compromised through diverse pathological stimuli, including metabolic dysreg

Galleria mellonella as a novel invertebrate model for studying Ehrlichia ruminantium pathogenesis and host-pathogen interactions.

Ehrlichia ruminantium, the causative agent of heartwater disease, is an obligate intracellular bacterium that poses significant economic threats to livestock production in endemic regions. Current research models present substantial ethical, logistical, and economic constraints, particularly for studying host-pathogen interactions within arthropod vectors. Here we establish Galleria mellonella larvae as a tractable invertebrate infection model for Ehrlichia ruminantium, enabling experimental investigation of pathogen persistence and host-pathogen interactions in an arthropod system. Following

Acetylation enables a dual-targeting fungal effector to inhibit insect immunity and development.

Pathogenic microbes deploy secreted effectors to subvert host immunity and physiology across kingdom boundaries, yet the molecular mechanisms that maintain effector competence during this transition remain poorly defined. Here, we identify MrNIS1, an effector from the insect-pathogenic fungus Metarhizium robertsii whose stability and secretion are governed by K24 acetylation through physical association with the acetyltransferase MrKAT1 and the deacetylase MrSIR2. Once inside the insect, MrNIS1 acts as a dual-targeting effector to disrupt distinct physiological pathways. It binds the serine pr

Immune system reads geometric pattern.

Immune recognition depends on molecular identity, yet the spatial organization of microbial structures also shapes receptor engagement and signaling. How these geometric effects connect pathogen architecture, host-cell attachment and immune recognition remains incompletely understood. Here I advance pathogen-associated geometric patterns (PAGPs) as a framework integrating particle shape and symmetry with ligand valency, spacing, orientation and mechanical presentation. By synthesizing evidence from innate and adaptive immunity with comparative analyses of viral architecture, this study examine

Cyclosporine A revisited: Molecular mechanisms, non-immune physiological pathways, and the architecture of multi-organ toxicities.

Cyclosporine A (CsA) remains a cornerstone immunosuppressant whose discovery transformed transplantation and immune-mediated disease management. Beyond its classical inhibition of the calcineurin-Nuclear Factor of Activated T-cells (NFAT) pathway, recent research has shown that cyclosporine A modulates multiple host physiological pathways, including mitochondrial function, endothelial signaling, and cytoskeletal dynamics. These mechanisms underpin both its therapeutic efficacy and its characteristic toxicities. CsA modulates cyclophilin A (CypA)-dependent processes that influence mitochondrial

Metabolic reprogramming and plasticity in breast Cancer: Mechanisms driving tumor progression, metastasis, and therapeutic resistance.

Breast cancer remains the most frequently diagnosed malignancy and a leading cause of cancer-related mortality among women worldwide. Despite substantial advances in endocrine therapy, human epidermal growth factor receptor 2 (HER2)-targeted therapy, chemotherapy, and immunotherapy, therapeutic resistance and metastatic progression remain major clinical challenges. Accumulating evidence has established metabolic reprogramming as a fundamental hallmark of breast cancer, enabling tumor cells to adapt dynamically to nutrient limitation, hypoxia, oxidative stress, and therapeutic pressure. Glycoalkaloids Affect the Animal Nervous System: A Review.

Glycoalkaloids (GAs) are plant secondary metabolites produced mainly by species of the Solanaceae family, such as the tomato Solanum lycopersicum L. Although traditionally associated with toxicity, recent research highlights their diverse biological activities, including anti-inflammatory, anticancer, antimicrobial, and neuroprotective effects. These properties arise from their ability to interact with cell membranes, modulate key signaling pathways, and influence immune responses. Selected GAs, such as solasonine, solamargine, and α-tomatine, have demonstrated the potential to regulate inflam

Focal therapeutic conditions matter: How energy affects antigen presentation and T cell activation.

Energy-based minimally invasive cancer focal therapies (FTs), including thermal ablation (Heat), cryoablation (Cryo), and irreversible electroporation (IRE), can induce immunomodulatory effects and can synergize with immunotherapy to engage the immune system for systemic and long-term antitumor response. However, the immunogenicity of each FT can vary based on treatment specifics, and the mechanisms underlying these differences are not fully understood. We employed a model antigen (ovalbumin, OVA) and an in vitro platform to investigate the upstream processing steps that initiate immune primin

Serum proteomic profiling identifies pathways and protein signatures associated with early versus late recurrence in cholangiocarcinoma.

Cholangiocarcinoma (CCA) is characterized by frequent postoperative recurrence, yet reliable biomarkers for distinguishing early from late recurrence remain limited. This study aimed to identify serum protein biomarkers associated with early versus late recurrence and develop a proteomic recurrence score (PRS) to distinguish these recurrence groups after curative resection. Serum proteomic profiling was performed using LC-MS/MS in patients with resected CCA. Candidate proteins were identified through integrated univariate and multivariate analyses, followed by pathway enrichment and protein-pr

Chimeric antigen receptor-macrophages: A new paradigm for cell therapy.

Chimeric antigen receptor (CAR) technology has propelled CAR-T cells to transformative success in hematologic malignancies, yet translation to solid tumors remains limited, motivating exploration of alternative CAR-engineered immune effectors. Macrophages, sentinels of the innate immune system, are abundantly recruited to solid tumors, making them compelling candidates. Preclinical studies show that CAR-engineered macrophages (CAR-M) exhibit precise tumor homing, potent antigen-directed phagocytosis, and the capacity to remodel immunosuppressive tumor microenvironments (TMEs). Notably, early-p

FAs and their metabolites as antibacterial agents: mechanisms, food applications, and translational opportunities.

Antimicrobial resistance and the demand for effective food-preservation strategies have increased interest in fatty acids (FAs) and their derivatives as potential antibacterial agents. FAs can affect bacterial membranes, cellular energetics, biofilm-associated phenotypes, and, in some organisms, specific metabolic pathways. However, these effects vary considerably with FA structure, concentration, ionization state, pH, formulation, bacterial species, and assay conditions. Additionally, microbiota-derived short-chain FAs may also contribute to host defense by influencing epithelial integrity, i

The intrinsic value of self-organized sport and physical activity: a narrative review.

In many populations, much of people's physical activity takes place outside clubs and organized competition, in self-organized forms that participants run themselves, from walking and strength training to skateboarding, parkour, and climbing. Policy and research tend to treat these forms as a residual category and to value them, when at all, for the same instrumental outcomes used to justify organized sport. This narrative review takes a different starting point and asks what is distinctively valuable about physical activity being self-organized. Its central argument is that the worth of these

Evolving skin cancer therapeutics: molecular and genetic drivers, immunotherapy challenges, and advances in nanogel-based nanomedicine.

Skin cancer is among the most common malignancies worldwide, with a rising global incidence. Cancer develops through complex molecular and genetic changes. Current treatments, such as surgery, chemotherapy, targeted therapy, radiation, and immunotherapy, offer hope. However, challenges like immune evasion, drug resistance, and systemic toxicities prevent long-term success. Nanotechnology-based delivery systems may offer a potential solution to these challenges. Cross-linked polymeric nanogel networks are unique nanocarriers with high drug-loading capacity, biocompatibility, stimuli-responsive

Late effects after novel therapies for childhood cancer: Anticipating the future landscape of health outcomes for survivors.

Five-year survival rates for childhood cancer now exceed 85%, with recent improvements driven by advances in understanding of tumor biology allowing for molecular risk stratification. The introduction of novel therapies, including antibodies, antibody-drug conjugates, immune checkpoint inhibitors, cellular therapy, and targeted small molecule inhibitors into early-phase and pivotal trials has the potential to further improve survival. Although these agents have transformed outcomes for some children with cancer, their long-term toxicity profiles remain poorly defined. Large cohort studies have

A digitally immune-optimized influenza vaccine broadly neutralizes swine and human H1N1 influenza viruses and protects from heterologous challenge.

UNLABELLED: Influenza A virus (IAV) zoonotic transmission and continuous evolution in multiple host species heighten the risk of emerging novel strains at the human-animal interface. Digitally immune-optimized hemagglutinin (HA), neuraminidase (NA), and matrix-2 (M2) vaccine antigens were engineered and combined as a single DNA expression construct (DVX-H1N1) to maximize immune responses to human seasonal, pandemic, and zoonotic H1N1 IAVs. Immunization of mice with DVX antigens induced broad neutralizing antibody responses to human- and swine-origin H1N1 isolates. The DVX-H1N1 candidate was th

Protein secretion by the type IV pilus machinery in Francisella tularensis.

Francisella tularensis is a highly virulent, gram-negative bacterial pathogen that causes the zoonotic disease tularemia. F. tularensis infects a variety of host cells and replicates intracellularly while evading and interfering with host immune responses. The molecular mechanisms that facilitate the intracellular replication and virulence of F. tularensis are poorly understood. The Francisella genome contains a set of pil genes that code for the assembly of surface fibers termed type IV pili (T4P). T4P are major bacterial virulence determinants, but the function of the pil system during F. tu
