Cortisol, cortisone and inflammation in relation to burnout symptoms.

Burnout is highly prevalent in high-demand healthcare professions, with medical students and trainees already at increased risk. The pathophysiological processes associated with burnout remain insufficiently understood as previous research is mostly limited to cross-sectional studies focusing on specific biological correlates. This study aims to investigate a wide range of biological correlates associated with burnout symptoms by exploring cortisol, cortisone, and multiple inflammatory markers in medical students and physicians-in-training. A three-year open cohort study was carried out in med

HLA-A, -B, and -DRB1 allele-group and haplotype profiles of the Azerbaijani population.

The Human Leukocyte Antigen (HLA) system is critical for immune recognition and transplantation compatibility. Despite its strategic location at the crossroads of the Caucasus, Anatolia, and the Middle East, comprehensive immunogenetic data for the Azerbaijani population remain limited. This study aimed to characterize HLA-A, -B, and -DRB1 allele-group and haplotype frequencies and establish a framework for optimized donor matching. We analyzed HLA-A, -B, and -DRB1 allele-group frequencies in 2301 healthy donors from Azerbaijan. Data were analyzed using the expectation-maximization algorithm f

DNASE1L3 maintains homeostatic levels of mitochondrial DNA on the mammalian cell surface.

The extracellular space is a critical environment for discriminating self- versus non-self-nucleic acids and initiating appropriate immune responses through signaling cascades to relay information about extracellular nucleic acids. Here, we provide evidence that mitochondrial DNA is tethered to the surface of select mammalian cells through cell surface proteins and heparan sulfate proteoglycans. We demonstrate that cell surface DNA accumulates in large clusters that partially overlap with domains enriched in RNA binding proteins. Finally, we show that human and murine B cell surfaces contain D

Induced regulatory B cells stably expressing interleukin-10 reverse central nervous system autoimmunity by targeting myeloid cells.

The lack of approaches to produce interleukin (IL)-10-producing B cells at high frequency has hampered mechanistic investigation into the generation, fate, and activity of these regulatory cells. We screened pharmacological compounds for their ability to induce IL-10 expression in B cells. One compound, N6, promoted stable IL-10 expression in >95% of B cells. Transfer of IL-10+ B cells induced remission of experimental autoimmune encephalomyelitis. N6-induced B cells accumulated in the central nervous system (CNS) and differentiated into IL-10+ plasma cells, promoting transcriptional and pheno

Deceptive simplicity and law-governed symbiosis: a critical mini-review of the mechanisms underlying the emergence of multicellular organisms.

The mechanisms underlying the evolutionary emergence of multicellular organisms are examined from a systems perspective. The key parameter of this evolution is complexity, both of individual cells and of the multicellular organism. The main problems of cell integration are examined, such as genome integration and the coordination of the functioning of immune and transport systems. As a result, the emergence of a multicellular organism from any unicellular organism is an unattainable task for random mutations and selection. The concept of regulated symbiosis is proposed as a mechanism for the f

Precision immunisation: reaching zero-dose children in underserved communities in Niger State, Nigeria.

Nigeria has one of the highest burdens of zero-dose children globally, particularly in underserved and hard-to-reach communities where routine immunisation systems face persistent barriers. In Niger state, these challenges are compounded by insecurity, seasonal flooding and dispersed populations, limiting the effectiveness of conventional facility-based approaches. We describe the implementation of the Strengthening Routine Immunization in Consequential Areas of Niger State (STRICAN) initiative, which applied a precision immunisation approach to identify and reach previously unreached children

Dysregulated tissue immunity in human fatal asthma.

Asthma is an immune-mediated lung disease causing airway constriction that is fatal in rare cases, though the immune mechanisms underlying asthma severity are poorly understood. Here, we present a comprehensive immunological profiling of lymphoid organs, lungs, and intestines from human organ donors who died of fatal asthma (FA) compared to donors who died of unrelated causes with or without a history of asthma. Compared to control donors, FA donors exhibit elevated plasma IgE along with enhanced and aberrant immune responses in mucosal-associated lymph nodes (LN) and lungs, respectively. In p

T cell immunomonitoring: a comparative analysis of traditional and novel methods to quantify and characterize human antigen-specific T cells.

T cells are a key component of the adaptive immune system, playing a central role in the body's defense against pathogens and diseases. Antigen discovery and longitudinal immunomonitoring of antigen-specific T cell responses offer valuable insights into the activity and efficacy of immunotherapies and can guide development of new prophylactic or therapeutic treatments, such as vaccines and cellular therapies. Traditional immunomonitoring methods for T cell analysis, while foundational, offer limited insights because of the complexity and vast heterogeneity of antigen-specific T cells. Here, tr

L-Arginine inhibits NLRP3 inflammasome activation.

Nutritional status and metabolic homeostasis are intricately associated with the inflammatory response. However, the direct mechanistic link between nutrition, metabolism and inflammation remains unclear. Here, we show that the amino acid L-arginine (L-Arg) is a potent endogenous inhibitor of the NLRP3 inflammasome, a central component of the inflammatory signalling pathway. We show that L-Arg binds directly to the Asp31 site of the NLRP3 protein, blocking its interaction with the adaptor ASC and subsequent inflammasome assembly and activation. L-Arg supplementation inhibits the NLRP3 inflamma

Transcriptomic analysis identifies immune-related clusters and PPP2R1A as a candidate gene associated with venous thromboembolism recurrence.

Venous thromboembolic disease (VTE), encompassing deep vein thrombosis (DVT) and pulmonary embolism (PE), is associated with substantial morbidity, mortality, and recurrence risk. However, immune-related transcriptomic heterogeneity and recurrence-associated molecular features in VTE remain incompletely defined. This study aimed to explore immune-related transcriptomic clusters associated with VTE recurrence and to prioritize recurrence-associated candidate genes. Public GEO datasets were analyzed, with GSE19151 used as the discovery cohort and GSE48000 used for external validation. Consensus

3D-Printed Bioadhesive Interfaces as Proliferative Niches for Tissue Repair.

Bioadhesives have traditionally been viewed as materials for wound closure, sealing and haemostasis However, in tissue repair they also create the first engineered interface that influences cell retention, microenvironmental homeostasis and subsequent remodelling Recent advances in 3D printing are shifting bioadhesives from passive, uniform layers to architecturally controlled interfaces with spatially programmable chemistry, mechanics, transport and biological function. In this Review, we propose an interface-centred framework in which qualifying 3D-printed bioadhesive interfaces function as

Nonreciprocal entanglement by dynamically encircling a nexus.

Nonreciprocal entanglement, characterized by inherently robust operation, is a cornerstone for quantum information processing and communications. However, it remains challenging to achieve nonreciprocal entanglement characterized by stability and robustness against environmental fluctuations. Here, we propose a universal nonlinear mechanism to engineer magnetic-free nonreciprocity in dissipative optomechanics by utilizing bistability, a phenomenon ubiquitous across nonlinear physical systems. By dynamically encircling the nexus of bistability, a cusp converged by the bistable surfaces, we obta

Supramolecular Pretargeting Lipid Nanoparticles Based on Host-Guest Recognition Potentiate Cancer Immunotherapy by Reprogramming Tumor Microenvironment.

Lipid nanoparticles (LNPs) have emerged as a promising platform for gene/drug delivery in cancer therapy, however their nonspecific biodistribution and extensive hepatosplenic sequestration result in insufficient intratumoral accumulation, thereby undermining the therapeutic efficacy. Here, we establish a sequential pretargeting strategy exploiting the high-affinity host-guest recognition between adamantane (ADA) and cucurbit[7]uril (CB[7]) to enhance tumor enrichment. Preadministered ADA-conjugated anti-PD-L1 antibody (ADA-αPD-L1) primes tumors for efficient capture of subsequently infused CB

dsRADAR: Imaging and Detecting Cellular dsRNA by Repurposing RNA-Binding Proteins.

Double-stranded RNA (dsRNA) is recognized by cellular receptors as a sign of viral infection, triggering the innate immune response. Cellular dysregulation can also lead to accumulation of endogenously produced dsRNA that stimulates a viral-like immune response. Additionally, dsRNA contamination in RNA therapeutics can lead to harmful side effects via a similar pathway. Despite the clinical relevance of dsRNA, reliable tools for its detection remain limited. Currently, dsRNA detection relies almost exclusively on the monoclonal antibodies J2 and K1, which suffer from sequence bias and low sens

MeCP2 functions as a DNA methylation reader in immune homeostasis and cancer.

DNA methylation is a key epigenetic mechanism that mediates gene expression and informs cellular identity. Methyl-CpG binding protein 2 (MeCP2) is a protein capable of recognizing and interpreting the methylated DNA code to bring about phenotypic outcomes through co-factor interactions. Altered DNA methylation patterns and MeCP2 function, therefore, disrupt cellular epigenetic homeostasis and contribute to human diseases. MeCP2 loss-of-function or amplification events are responsible for Rett syndrome and MECP2 duplication syndrome, respectively, where patients present with multisystem pathoph

Hyaluronic acid-based tissue-engineered biomaterials for infected diabetic foot ulcers: mechanisms, evidence, and translational challenges.

Diabetic foot ulcers (DFUs) have difficulty healing clinically when infection and mature biofilms coexist with hyperglycemic injury, immune dysregulation, oxidative stress, ischemia-hypoxia, repetitive mechanical loading, and extracellular matrix (ECM) degradation. Debridement, infection management, offloading, vascular assessment and revascularization, when indicated, constitute the foundation of care; biomaterials can be evaluated only as adjuncts to this pathway. Hyaluronic acid (HA) is an attractive hydrated ECM-associated and chemically adaptable matrix that can support local retention, c

Systems-level analysis uncovers eight hub genes and a resveratrol-based therapeutic strategy for ischemic stroke.

Ischemic stroke is a leading cause of mortality and long-term disability worldwide, yet its underlying molecular mechanisms remain incompletely understood. The pathology involves complex interactions between vascular injury, immune dysregulation, metabolic imbalance, and mitochondrial dysfunction. Traditional single-gene or single-dataset approaches have limitations in capturing the multifactorial nature of the disease. We developed a comprehensive systems-level bioinformatics framework integrating five publicly available transcriptomic datasets (GSE119121, GSE146882, GSE22255, GSE37587, GSE58

Immune checkpoint inhibitor-related myotoxicity in lung cancer: a decade-long real-world pharmacovigilance analysis of the FAERS database.

BACKGROUND: Immune checkpoint inhibitor therapy has revolutionized the treatment of several malignancies, with improved survival, yet the accompanying myotoxicity has often been underestimated. We aimed to comprehensively and comparatively analyze the risk of myotoxicity associated with various ICIs using the FDA Adverse Event Reporting System database. METHODS: Data spanning from the first quarter of 2015 to the fourth quarter of 2024 were extracted from the U.S. Food and Drug Administration Adverse Event Reporting System database. Disproportionality analysis was conducted using the reportin

A method for distinguishing and separating retinal microglia from infiltrating macrophages during an ocular inflammatory event.

A significant challenge in the study of neuroimmune responses is the identification and separation of resident microglia from infiltrating bone marrow-derived macrophages. Therefore, we investigated whether dual labelling of Cx3cr1 and Ccr2 is sufficient to discriminate retinal microglia from infiltrating macrophages in mice that develop spontaneous autoimmune uveoretinitis-a condition dependent on infiltration of lymphocytes and antigen presenting cells from the circulation into the retina. Mice expressing red fluorescent protein (RFP) under control of the Ccr2 promoter and green fluorescent
