Closing the Surveillance-to-Stewardship Gap in antimicrobial resistance: a narrative review of global lessons from WHO GLASS, UK ESPAUR, and low- and middle-income countries.

BACKGROUND: Antimicrobial resistance (AMR) surveillance has expanded faster than any other component of the global response, yet resistance continues to rise. In a growing number of settings, the principal constraint is the failure to translate available data into action, a disconnect we describe as the Surveillance-to-Stewardship (S2S) Gap. OBJECTIVES: To define the S2S Gap and its component gaps, examine where it binds across health systems of different maturity, analyse why it persists, and propose an operationalisable, testable framework for closing it. SOURCES: A cross-sectional analysi

In Silico Pipeline for the Development and Primer Design of Microsatellite Markers in Mangifera similis Blume.

Mangifera similis Blume, a wild relative of the mango, is an important genetic resource for improving mango varieties. This chapter outlines an in silico pipeline for the development and primer design of microsatellite (SSR) markers for the chloroplast and the whole genome of M. similis, using bioinformatics and next-generation sequencing. The method enables efficient microsatellite identification, saving laboratory time and costs, and is particularly useful for tree species with limited genomic data. The workflow comprises steps such as DNA extraction, sequencing, library construction, in sil

Application of Single Nucleotide Polymorphisms (SNPs) in Sesame (Sesamum indicum L.) for Genetic Diversity, Mapping, and Marker-Assisted Selection.

Sesamum indicum L. is one of the oldest oilseed, cultivated oleaginous species, which is highly valued due to the excellent quality of the oils and owing to its antioxidative effects. Even though it is a major economic crop, there is a historical gap in breeding of sesame compared to the major staple crops, mainly because of a lack of genomic resources. With the emergence of next-generation sequencing methods, single-nucleotide polymorphisms (SNPs) with their noted benefits, compared to older marker systems, such as dense marker coverage, whole-genome coverage, and high-throughput automation c

SSR Marker-Assisted Selection and Monitoring of High-Metabolite Cell Lines in Plant Tissue Culture: Protocols and Somaclonal Variation Assessment.

Simple Sequence Repeat (SSR) marker-assisted selection has proven to be the most powerful tool for genetic monitoring and selection in in vitro culture systems. It is widely used in detecting somaclonal variation and producing true-to-type in vitro plantlets with superior quality. Genetic fidelity is the critical point in plant tissue culture, and SSR markers act as a supporter to pass this critical point through locus specificity, codominancy and high polymorphism. SSR markers allow the precise detection of genetic changes during callus formation, organogenesis, somatic embryogenesis, and lon

Untargeted UHPLC-HRMS Lipidomic Profiling of Ticks for Alpha-Gal Syndrome Research.

Lipids are a fundamental group of biomolecules that play diverse roles in biological systems, including energy storage, membrane architecture, and intra- and extracellular signaling. They are classified into eight major categories, and more than 50 000 distinct lipid structures are currently reported in the Lipid Maps Structure Database (LMSD). Given their importance in cellular metabolism and physiology, accurate identification and quantification of lipids are essential. In ticks, lipid metabolism is closely associated with physiological processes related to survival, pathogen transmission, a

Development and Application of Restriction Fragment Length Polymorphism (RFLP) for the Improvement of Cotton Crops.

Restriction fragment length polymorphism (RFLP) has been an important molecular tool in advancing cotton (Gossypium spp.) improvement by enabling researchers to detect stable genetic variations across the genome. Using restriction enzymes and labeled probes, RFLP generates highly reproducible, co-dominant markers that help identify differences among cultivars, wild species, and breeding lines. This technique has been widely used to construct early genetic linkage maps, locate quantitative trait loci (QTLS) associated with fiber quality, yield, and disease resistance, and study genome structure

Development and Application of Random Amplified Polymorphic DNA (RAPD) for the Improvement of Cotton Crops.

Random amplified polymorphic DNA (RAPD) markers have played a significant role in accelerating cotton (Gossypium spp.) improvement by enabling rapid genome-wide assessment of genetic variation without the requirement for prior sequence information. RAPD has been widely applied in cotton germplasm characterization, cultivar identification, hybrid confirmation, and the early development of molecular linkage maps. Its simplicity, low cost, and the ability to amplify multiple polymorphic loci in a single reaction made RAPD particularly valuable during the early phases of cotton molecular breeding.

Development of Genome-Wide Simple Sequence Repeat Markers from Whole-Genome Sequence of Mungbean (Vigna radiata).

Mungbean (Vigna radiata L.) is a major pulse crop cultivated extensively across Asia and is considered an important model species among legumes because of its diploid genome and relatively small genome size. The availability of reliable, economical, and highly polymorphic molecular markers is critical for assessing genetic diversity, constructing linkage maps, and implementing functional genomics-based breeding programs. Among the available marker systems, simple sequence repeat (SSR) markers are highly polymorphic, abundant, co-dominant, and extensively used for allelic characterization acros

ISSR Marker-Based Authentication of In Vitro-Derived Medicinal Plant Lines for Enhanced Phytochemical Production.

Medicinal plants have been used to heal people with thousands of diseases. Phytochemicals are the main reason for a plant being a medicinal plant. Compared to field-grown plants, in vitro-derived plantlets have high phytochemical production. In vitro-derived plantlets are also the most reliable method for producing healthy, disease-free medicinal plants of superior quality, with enhanced phytochemical properties. Due to the presence of phytochemical compounds such as phenolic compounds, flavonoids, tannins, and terpenoids, medicinal plants possess therapeutic properties. These therapeutic prop

Genotyping Protocol for Identifying Genetic Risk Markers Associated with Alpha-Gal Syndrome.

Alpha-gal syndrome (AGS) is an IgE-mediated allergy to galactose-α-1,3-galactose (α-Gal), typically triggered by tick bites. Genetic factors, including human leucocyte antigen (HLA) class II polymorphisms and selected single nucleotide polymorphisms (SNPs) in immune and glycan-related genes, are hypothesized to modulate susceptibility and clinical expression. Here we provide a step-by-step protocol to genotype risk markers from ethylenediaminetetetraacetic acid (EDTA) whole blood sample, combining (i) manual DNA extraction (Qiagen), (ii) SNP genotyping by a real-time/quantitative PCR (qPCR) me

Modeling Antigen Allergenicity with Bioinformatics Approaches.

Allergenicity is a multifactorial and context-dependent property that emerges from the interaction between intrinsic protein features and host immune mechanisms. Although regulatory frameworks rely primarily on sequence homology to known allergens, increasing numbers of novel and engineered proteins require more comprehensive computational strategies for preclinical assessment. This chapter describes a modular bioinformatic pipeline for the prediction and characterization of potential protein allergens. The workflow integrates FAO/WHO sequence-based criteria for regulatory compliance, structur

Pre-driven and post-gated memristive Hopfield network: Dynamics, FPGA realization and image encryption.

Many existing memristive synaptic models merge presynaptic and postsynaptic activities into a single aggregated stimulus, making their distinct functional roles difficult to interpret. To address this issue, this paper proposes a memristive synapse with a pre-driven and post-gated state-update mechanism, in which presynaptic activity drives synaptic-state evolution while postsynaptic activity nonlinearly modulates the direction and strength of the update. The proposed synapse is embedded into a compact two-neuron memristive Hopfield neural network to investigate how this functional decompositi

Stress Tolerance Assays for Salmonella enterica: Methods for Evaluating Heat, Acidity, and Osmotic Resistance.

Salmonella remains a leading cause of foodborne illness worldwide and imposes substantial public health and economic burdens, particularly due to its persistence in food production systems. This persistence is driven not only by antimicrobial resistance but also by adaptive survival strategies, including stress tolerance that allows the cells to endure sublethal and lethal stresses encountered across the farm-to-fork continuum. Quantitative characterization of these phenotypes is crucial for understanding pathogen survival dynamics and strain-specific variability, thereby improving food safety

Ewing Sarcoma-Specific (Re)Expression Models.

Gene expression and knockdown systems are powerful tools for studying the function of single genes and their interactions within pathways. Plasmid transfection and viral transduction have revolutionized the field of molecular biology and paved the way for various gene-editing strategies such as TALEN, Zink finger nucleases, and ultimately CRISPR. In Ewing sarcoma (EwS), almost as many genes are repressed by the expression of EWSR1-FLI1 as are upregulated by the fusion oncogene. Here, we present a useful point-by-point protocol for generating transgene expression systems in EwS that allow (cond
