Nitro-substituted tert-butyl peresters induce apoptosis and exhibit multi-targeted anticancer activity in breast cancer cells.

STUDY AIMS: The COVID-19 pandemic required healthcare systems to reorganise around patient surges. Previous evidence associated these reorganisations with worse clinical outcomes for non-COVID-19 patients, to varying degrees. This exploratory study aimed to quantify the effects of successive COVID-19 waves on mortality, length of stay and transfers to intermediate and intensive care units among non-COVID-19 patients by comparing COVID-19 and non-COVID-19 periods in a Swiss hospital. METHODS: This retrospective monocentric cohort study extracted electronic health data of patients hospitalised

Video Remote Sign Language Interpreting and Health Communication for Deaf Patients: A Randomized Clinical Trial.

IMPORTANCE: Current interpretation services for Deaf patients who use sign language are often ineffective or unacceptable. In-person interpretation is frequently unavailable, and while video remote interpreting (VRI) remains underused, its scalability may be a solution given interpreter shortages and cost barriers. Existing research focuses on user and interpreter preferences, leaving a critical gap in understanding how interpretation formats affect communication quality. OBJECTIVE: To evaluate the effectiveness of VRI in improving communication outcomes between Deaf patients and physicians c

The landscape of gene mutations in a cohort of 3353 Han Chinese children with nonsyndromic hearing loss.

BACKGROUND: Genetic diagnosis plays a critical role in enhancing the identification and clinical management of pediatric hearing loss, ultimately improving both physiological outcomes and social-emotional development. As one of the most prevalent sensory disorders characterized by substantial genetic heterogeneity, hearing loss requires the implementation of efficient diagnostic approaches to inform targeted clinical interventions and personalized care strategies. METHODS: Here, we recruited 3353 Han Chinese children with nonsyndromic hearing loss, including 598 multiplex cases with family hi

From Usher syndrome to Bardet-Biedl syndrome: Diagnosis after an atypical presentation.

Syndromic associations of kidney and ophthalmological diseases are not typical in the young adult. The most common kidney manifestations are malformations of the genitourinary apparatus, while some syndromes can also present with kidney parenchymal disease. We report on a 51-year-old woman evaluated in the nephrology clinic for 12 months due to abnormal kidney function. She had been diagnosed with Usher syndrome due to the development of retinitis pigmentosa and sensorineural hearing loss in the last 20 years, without genetic testing. She had a family history of parental consanguinity, and dis

Exome sequencing and large-scale analysis of electronic medical record-linked biobank data identify candidate deafness genes.

INTRODUCTION: Rapid advances in whole-exome sequencing (WES) have enabled large-scale detection of pathogenic variants. Although hundreds of genes are implicated in hearing loss, up to half of inherited cases remain unsolved, limiting eligibility for gene therapy trials that require genetic diagnosis. Biobanks and electronic medical records (EMRs) offer opportunities to integrate genomic and clinical data at scale and expand the spectrum of hearing loss genes. Despite clinical value, EMRs often lack key information such as inheritance patterns, posing challenges for accurate interpretation. M

Isolated Congenital Ossicular Chain Anomalies.

Isolated congenital ossicular chain anomalies (COCAs) are an uncommon cause of conductive hearing loss in patients with a normal external auditory canal and tympanic membrane, and familial clustering is rarely described. We report three siblings with non-progressive conductive hearing loss and normal otoscopic findings. Two siblings exhibited bilateral involvement, whereas the third had unilateral disease with a normal contralateral ear. High-resolution temporal bone computed tomography suggested incudostapedial (I-S) discontinuity caused by defects in the long process of the incus, without ex

Liposomal amphotericin B (AmBisome) is a lipid-associated formulation of the broad-spectrum polyene antifungal agent amphotericin B. It is active against clinically relevant yeasts and moulds, including Candida spp., Aspergillus spp. and filamentous moulds such as Zygomycetes, and is approved for the treatment of invasive fungal infections in many countries worldwide. It was developed to improve the tolerability profile of amphotericin B deoxycholate, which was for many decades considered the gold standard of antifungal treatment, despite being associated with infusion-related events and nephr

A Missense Variant in the IKZF2 Gene Identified in a Genetically Undiagnosed Family With Hearing Loss.

Hearing loss is one of the conditions characterized by a high degree of genetic heterogeneity, and whole exome sequencing (WES) serves as a key method for identifying pathogenic variants. To date, 155 genes have been reported to be associated with nonsyndromic hearing loss. Recently, a study by Velde et al. found that the IKZF2 (OMIM#606234) gene is associated with nonsyndromic hearing loss. In our cohort of nearly 300 patients with undiagnosed hearing loss who underwent WES, we discovered a case harboring a variant in the IKZF2 gene, specifically c.485A > C (NM_001387220.1). By investigating

A Novel Missense Variant in LMX1A Leads to Autosomal Dominant Nonsyndromic Hearing Loss.

Hereditary nonsyndromic hearing loss (NSHL) is a prevalent entity associated with over 150 known causative genes, including LMX1A, which has fewer than 10 reported pathogenic variants. Here we present a novel missense variant in LMX1A in a family of European descent with hereditary hearing loss. Clinical and family histories were obtained, and genetic testing was performed on two affected members using the BMGDL Hearing Loss Panel. Allelic frequencies were explored across population databases, and in silico predictors were used to assess pathogenicity. Genetic testing revealed a variant of unc

Do's and Don'ts of Taking Care of Deaf Patients.

AUDIENCE: Emergency medicine residents, fellows, and attending physicians, any practicing provider in a medical setting that may serve Deaf patients. INTRODUCTION: Emergency medicine providers often interact with Deaf and Hard of Hearing (DHH, or just HOH, for only hard of hearing) patients. Various limitations, however, affect their ability to effectively engage with DHH patients such as acuity, lack of time, and/or readily available communication tools (eg. virtual or in-person interpreters), among other challenges. These barriers contribute to numerous DHH healthcare disparities. Estimatin

Interpretation of de novo mutations (DNM) and genetic counseling for sporadic hearing loss based on family trio-based sequencing.

De novo mutations (DNMs) are significant genetic factors contributing to sporadic hearing loss (HL) and complex HL syndromes. To analyze the genetic counseling characteristics and interpretation of pathogenic DNMs for sporadic HL, we retrospectively analyze the clinical information of probands and their parents from 410 sporadic HL core pedigrees enrolled in the "Chinese Deafness Genome Project (CDGP)" between October 2015 and October 2023. We apply family trio-based genome sequencing (targeted gene capture and high throughput sequencing, mitochondrial genome sequencing, and copy number varian

Expanding the Clinical Phenotype Associated with the NIN Gene; Report of a Patient with Short Stature, Microcephaly and Hearing Loss.

To date, there are very few reports regarding patients with bi-allelic variants in the NIN gene. There is one report of two sisters with severe short stature, microcephaly, and developmental delay with compound heterozygote missense variants in the NIN gene and one paper reporting a homozygote variant in the NIN gene with progressive, high-frequency sensorineural hearing loss in four siblings. The only other report is of four members of a consanguineous family with spondyloepimetaphyseal dysplasia with joint laxity-leptodactylic type (SEMDJL2) with a homozygous variant in the NIN gene. A novel TIMM8A mutation in Mohr-Tranebjaerg syndrome without hearing loss and with basal ganglia iron deposition.

Mohr-Tranebjaerg syndrome (MTS) is a rare X-linked recessive neurodegenerative disorder caused by pathogenic variants in the TIMM8A gene. TIMM8A, also known as Deafness-Dystonia Peptide-1 (DDP1) is a mitochondrial intermembrane space protein involved in the import and insertion of hydrophobic membrane proteins from the cytoplasm into the mitochondrial inner membrane. MTS typically presents early-onset progressive hearing loss, dystonia, visual impairment, and cognitive decline. Here, we report a case of a male adolescent with a previously undescribed variant in TIMM8A, associated with progress
