Mid- and Long-Term Risk of Venous Thromboembolism Among Female Breast Cancer Survivors: A Nationwide Cohort Study.

Venous thromboembolism (VTE) is a serious complication for breast cancer (BC) survivors. We identified records of female BC survivors from the Korean Health Insurance Service database between January 1, 2010, and December 31, 2016. After applying exclusion criteria (age < 18 years, prior cancers or VTE, absence of BC surgery, or missing data), 112,522 BC survivors were included; for the comparison group, BC survivors were matched (1:3) to women without cancer history. VTE risk was evaluated with a Fine and Gray model, accounting for death as a competing risk; multiple time periods were analyze

Priorities for breast cancer risk assessment in UK women under age 50: a survey and discrete choice experiment.

BACKGROUND: National guidelines recommend English women between 30 and 49 years with moderate/high risk of breast cancer (BC) be offered screening and prevention interventions. Identification  largely relies on family history-based criteria amongst women self-presenting in primary care. We aimed to understand the preference for different potential risk assessment strategies in this population. METHODS: A cross-sectional survey including a discrete choice experiment. Risk assessments were defined using six attributes, including initiation strategy, eligibility for a full/detailed risk assessme

Comparison of NICE criteria with the BOADICEA multifactorial risk model to guide breast cancer risk assessment and referral amongst women under age 50 within primary care.

BACKGROUND: The National Institute for Health and Care Excellence (NICE) recommends that women at above-population-level risk of breast cancer (BC) are offered enhanced screening and risk-reducing interventions. Identification of these women largely relies on referral through primary care based on family history criteria. METHODS: We compared the performance of the NICE criteria with versions of the BOADICEA multifactorial risk model in identifying women who developed BC within 10 years. Analyses included 1258 women aged under 50 years with family history, reproductive, lifestyle and polygeni

No genome-wide correlations and little shared genetic architecture between reproductive life-history traits and estrogen receptor-positive breast cancer risk.

Estrogen receptor-positive breast cancer (ER + BC) is one of the most prevalent cancers, but the evolutionary processes shaping genetic variation in ER + BC risk are poorly understood. Both evolutionary life-history theory and evidence from studies of individual ER + BC risk variants suggest that increased genetic ER + BC risk is associated with faster maturation, earlier reproduction, and/or increased reproductive success (i.e. there is a trade-off), but it is unclear how well this pattern is replicated when considering the polygenic architecture of these traits after controlling for potentia

Trastuzumab Deruxtecan Resistance via Loss of HER2 Expression and Binding.

UNLABELLED: Trastuzumab Deruxtecan (T-DXd) is clinically beneficial in HER2-positive and HER2-low metastatic breast cancer. However, therapeutic resistance emerges over time in most patients, with poorly defined resistance mechanisms. Through a molecular characterization of paired patient specimens before and after T-DXd treatment, we found that 49% cases had major decreases in HER2 expression at progression, and among them, 52% exhibited complete HER2 loss. Using isogenic model systems, we demonstrated that decreases in HER2 expression corresponded to reductions in T-DXd internalization and m

Cold Urticaria With Negative Standard Cold Stimulation Testing: Results From the COLD-CE Study.

BACKGROUND: Cold urticaria (ColdU) is defined by recurrent wheals and/or angioedema after cold exposure. Typical ColdU is confirmed by positive standard cold stimulation testing (SCST), defined as whealing within 10 minutes over the stimulated area on the volar forearm after provocation with an ice cube and/or TempTest. SCST-negative ColdU remains insufficiently characterized. OBJECTIVE: To characterize SCST-negative ColdU and identify factors independently associated with it. METHODS: We analyzed 364 patients with ColdU from the international, cross-sectional Comprehensive Evaluation of Col

Early Signs of Success for Missouri's New Notification of Pregnancy.

BACKGROUND: Epithelial ovarian cancer is among the leading causes of death in women and is driven by angiogenesis. Microvascular density (MVD) can be used to evaluate angiogenesis in carcinomas and thus it can be used as a potential biomarker for ovarian cancer. This study is aimed to establish the association between endocan-MVD with clinicopathological factors in primary epithelial ovarian cancer. METHODS: The clinicopathological characteristics were acquired from the medical records filed between January 2008 and December 2018 of 89 epithelial ovarian cancer cases in Hospital Universiti Sa

Validation of a Clinical and Polygenic Risk Prediction Model for Ovarian Cancer in the Nurses' Health Study.

UNLABELLED: Ovarian cancer is the leading cause of death among gynecologic cancers because of its late diagnosis, but risk stratification for targeted prevention can improve outcomes. To identify women at risk of ovarian cancer, pathogenic variant testing is used, especially for those with a family history of the disease. However, high-risk genetic variants are rare, accounting for <20% of ovarian cancer cases overall. More recently, clinical models that help identify ovarian cancer cases incorporate polygenic risk for improved risk stratification. Herein, we cross-validate a polygenic risk-in

Cervical cancer screening coverage and care cascade in Tamil Nadu, India, based on a community-based cross-sectional survey.

Low-and middle-income countries, including India, contribute to over 80% of cervical cancer deaths in the world. Maintaining the continuum of care during screening and diagnosis is important for early detection of cervical cancer. We assessed the screening coverage and care cascade for cervical cancer in Tamil Nadu, a southern state in India with a population of over 78 million. We conducted a state-wide STEPwise approach to NCD risk factor surveillance (STEPS) survey in Tamil Nadu in 2024. We embedded a cervical cancer screening module and interviewed 4184 women aged 30–69 years with the modu

Barriers and facilitators of cervical cancer screening among female healthcare professionals: a systematic review.

BACKGROUND: Cervical cancer remains as one of the leading causes of cancer-related deaths among women, due to the significant disparities with regard to the implementation of cervical cancer screening programs between the high and low—and middle-income countries. Studies have shown that cervical cancer screening uptake among female healthcare professionals (HCPs) has also been remarkably low. This systematic review aimed to identify the barriers and facilitators that affect the uptake of cervical cancer screening among female HCPs. METHOD: In this systematic review, we selected a total of 456
