PURPOSE: Women with BRCA1/2 pathogenic variants (PVs) face high breast cancer risks and complex preventive decisions, including whether to undergo risk-reducing mastectomy, which can generate decisional conflict. Polygenic risk scores (PRSs) offer individualized risk estimates that may support informed preventive decisions. We examined whether PRS disclosure reduces decisional conflict about risk-reducing mastectomy in women with BRCA1/2 PVs and whether PRS magnitude interacts with individual characteristics.
METHODS: Unaffected women with BRCA1/2 PVs (N = 354) were assessed before PRS disclo

BACKGROUND: Breast cancer is the most common cancer among women in Taiwan, yet most epidemiological studies treat Taiwanese Han women as a homogeneous population. Whether breast cancer occurrence differs across major Han subgroups, such as Hakka and Min-nan, remains insufficiently explored.
METHODS: We conducted a nationwide, population-based study by linking the Taiwan Biobank (TWB) with the National Cancer Registry (NCR) and the National Health Insurance Research Database (NHIRD). A total of 78,530 women aged 30-70 years were included, comprising 879 breast cancer cases and 77,651 controls.

BACKGROUND: Inavolisib, a recently approved phosphatidylinositol 3-kinase α (PI3Kα) inhibitor for advanced breast cancer, carries a notable risk of hyperglycemia. Despite this established, on-target adverse effect, clinical endocrinologists often lack sufficient awareness regarding both the risk profile and the specific management strategies required for Inavolisib-induced hyperglycemia. The objective of this report is to describe a patient with fulminant-like diabetes accompanied by Hyperosmolar Hyperglycemic State (HHS) following Inavolisib administration.

A 52-year-old man with no personal or family history of bleeding presented with extensive hematomas and skin bruising following prolonged antibiotic use. Initial laboratory evaluation revealed coagulopathy, with an elevated activated partial thromboplastin time and mild anemia. Mixing studies and factor assays confirmed the diagnosis of acquired hemophilia A, characterized by low factor VIII levels and high-titer factor VIII inhibitors. The patient was initiated on corticosteroids and cyclophosphamide, leading to gradual improvement in both bleeding symptoms and coagulation parameters. Compreh

In cat sensitized children, decision to eliminate exposure to cats depends on the clinical outcome. In this study, we aimed to examine the relationship between molecular cat allergen sensitization and exhaled breath condensate (EBC) IL-4, IL-5, and IL-13 as markers of allergic airway inflammation. We enrolled subjects with skin prick test diagnosis of cat allergen sensitization aged 4-17 years. Age, sex, diagnosis, and clinical symptoms as well as family history of allergic diseases were recorded at enrollment along with the association of symptoms and cat exposure. Asthma severity and allergi

A key concern with menopausal hormone therapy is the potential duration-dependent increase in breast cancer risk. Despite using progestogens (micronized progesterone and dydrogesterone), associated with the lowest risk, a slight increase in risk is still observed over 6 years of treatment. This increase in breast cancer concerns estradiol receptor-positive breast cancers and is consistent with a promoter effect on preexisting lesions. It is thus recommended to select patients according to their level of breast cancer risk before indicating menopause hormone therapy or preferring nonhormonal al

INTRODUCTION: Infantile hemangiomas (IHs) are the most prevalent noncancerous childhood tumors, affecting up to 10% of infants. IHs develop de novo and following excessive proliferation in the first 4-9 months; they typically degenerate spontaneously in 4-7 years. While treatment is not usually required, 10-15% of cases necessitate clinical or surgical intervention due to functional impairments. Here, we present a complicated capillary IH case, from a clinical and genetic perspective.
MATERIAL AND METHODS: A 13-month-old patient, with free family history, presented with a cutaneous capillary-

BACKGROUND: Women with a family history of breast cancer have an elevated risk of developing the disease. In clinical practice, the probability of developing breast cancer over a specified timeframe is frequently estimated using breast cancer risk prediction models. It is currently unclear which of the available models performs best in women with a breast cancer family history.
OBJECTIVES: To identify, describe, and appraise breast cancer risk prediction models developed or validated in women with a family history of breast cancer, and to meta-analyse their performance in predicting breast ca

PURPOSE: Cumulative lifetime endogenous estrogen exposure (CLEEE) is an established breast cancer (BC) risk factor, yet Nigeria faces increasing BC incidence and mortality despite reproductive patterns typically associated with lower estrogen exposure. We evaluated the association of endogenous estrogen exposure and BC risk in this population.
METHODS: We analyzed 609 Nigerian BC cases and 609 controls from the MEND case-control study to examine associations between CLEEE and BC risk by molecular subtype. CLEEE was defined as time from menarche to menopause (or enrollment), minus pregnancy du

This paper presents a time-stratified breast cancer survival analysis that incorporates tumor characteristics, disease stage, and patient features, using machine learning (ML) algorithms to support personalized care planning. Considering nonlinear relationships between input data and the risk of death, as well as the dynamics of patient characteristics over time, we utilize 6 ML algorithms, including XGBoost, Logistic Regression, Random Forest, Decision Tree, AdaBoost, and Multilayer Perceptron, for less than 6 months, 6 months, 1-, 2-, 3-, 5-, and 10-year survival rates prediction. We utilize

Precise breast cancer risk assessment (BCR) is essential for personalised prevention in women with a family history of hereditary breast and ovarian cancer (HBOC). The CanRisk model integrates monogenic variants with reproductive, lifestyle and familial factors and can be extended by Polygenic Risk Scores (PRS). We evaluated the impact of PRS-BC313 in three groups: (1) healthy carriers of (likely) pathogenic variants (LP/P), (2) affected LP/P carriers and (3) healthy female relatives from variant-negative HBOC families. In healthy LP/P carriers, median 10-year breast cancer (BC) risk remained s

BACKGROUND: Black women are more likely to develop and die from early-onset breast cancer. Hereditary breast cancer (HBC) is strongly associated with early age of onset. Accordingly, HBC characteristics in young Black women were evaluated.
METHODS: Black women diagnosed with invasive breast cancer at age 50 years or younger between 2005 and 2018 were recruited through the state cancer registries in Florida and Tennessee. Participants were asked to complete a questionnaire, sign medical records releases, and donate a saliva sample for free genetic testing to identify HBC germline pathogenic or

We present two illustrative cases highlighting diagnostic, surveillance and management complexities of TP53 pathogenic variants (PVs). Case 1 describes a 24-year-old female with early-onset breast cancer and a somatic mosaic TP53 PV with a variant allele frequency of 19% in blood, initially missed by panel sequencing. Case 2 concerns a 59-year-old female with multiple primary tumours and two identical TP53 variants detected in two different tissues which initially suggested somatic mosaicism but were consistent with a myelodysplastic syndrome-related clone secondary to homozygous germline ERCC

BACKGROUND: Breast implant-associated anaplastic large cell lymphoma (BIA-ALCL) is a rare T-cell lymphoma increasingly linked to textured breast implants. It typically presents as a late-onset periprosthetic seroma and carries a generally favorable prognosis when diagnosed early. However, its pathogenesis remains incompletely understood, particularly in patients with genetic cancer predispositions.
CASE PRESENTATION: We report a unique case of BIA-ALCL in a 41-year-old woman who underwent bilateral prophylactic mastectomy and subsequent implant-based reconstruction due to a strong family hist

BACKGROUND: Breast cancer rates are increasing globally amongst premenopausal women, who often face aggressive subtypes with poorer outcomes than postmenopausal women. In the United Kingdom, current guidelines for risk assessment amongst women under age 50 years focus on family history or genetic predisposition, a strategy that fails to identify most women that go on to develop breast cancer. The Breast CANcer Risk Assessment in Younger women (BCAN-RAY) study is evaluating the feasibility and acceptability of a novel risk assessment strategy for women aged 30-39 years, without a strong family
