Stage-stratified prognostic impact of comorbidities on breast cancer-specific survival: A population-based flexible parametric modelling study.

BACKGROUND: Comorbidities are common among women with breast cancer, yet their independent contribution to prognosis remains poorly characterised, particularly across disease stages. Most prognostic models prioritise tumour-related factors and when comorbidities are included, they are often summarised using aggregate indices, limiting evaluation of specific conditions. We investigated whether individual comorbidities exert effects on breast cancer-specific survival stratified by stage. METHODS: We analysed 3323 women aged 20 - 79 years diagnosed with invasive breast cancer in Queensland, Aust

[Systemic pruritus: focus on hematological, nephrogenic, and hepatobiliary etiologies].

Systemic pruritus refers to a chronic, generalized sensation of itching in the absence of primary cutaneous lesions. It may represent an important clinical manifestation of underlying internal disease and is particularly associated with hematologic, nephrologic, and hepatobiliary disorders, often displaying characteristic clinical features. In some cases, pruritus precedes the diagnosis of the underlying condition by months or even years and may therefore serve as an early indicator of a hematologic malignancy or cholestatic liver disease. More commonly, however, it occurs concurrently with th

High Prevalence of Non-BRCA Pathogenic Variants and a Recurrent PALB2 Variant in an Argentine Hereditary Breast and Ovarian Cancer Cohort.

Hereditary breast and ovarian cancer (HBOC) syndromes are strongly associated with germline pathogenic variants. While genetic testing historically focused on BRCA1/2, multigene panels have revealed clinically relevant variants in numerous non-BRCA genes. However, data on the spectrum and prevalence of these variants remain limited in Argentina, particularly in regions with distinctive admixture patterns. This study aimed to determine the frequency and spectrum of pathogenic and likely pathogenic variants (P/LPVs) in non-BRCA genes among patients with a personal and/or family history of breast

A clinical pilot study for personalized risk-based breast cancer screening utilizing a polygenic risk score.

BACKGROUND: Population-based mammographic screening is primarily age-based. However, breast cancer risk is multifactorial, and women may benefit from personalized risk-based screening. This pilot study aimed to explore the use of polygenic risk score (PRS) as a tool for risk stratification in personalized screening. METHODS: We included 80 women aged 40-49 years referred for clinical mammography. Exclusion criteria were prior breast cancer or premalignant breast disease, and previous genetic testing. After DNA collection, PRS was calculated from 2805 Single Nucleotide Polymorphisms (SNPs). Sc

Current overview of breast cancer risk assessment models - practical application in outpatient gynecological practice.

The rising incidence of breast cancer represents a serious medical problem. Women diagnosed with breast cancer before the age of 45 - that is, those who were not included in mammography screening programs - constitute a specific patient group. Identification of risk factors for the pathogenesis of breast cancer has led to the development of triage models based on both medical history data and genetic testing, as well as imaging methods; their updated versions are based on a combination of these modalities. The oldest empirical Gail model relies solely on basic medical history. The IBIS model,

Elevated platelet-to-lymphocyte ratio is associated with increased breast cancer risk: A cross-sectional analysis of the NHANES 2009 to 2020.

Inflammation is a recognized driver of tumorigenesis, particularly in breast cancer. The platelet-to-lymphocyte ratio (PLR) has emerged as a readily available inflammatory indicator; however, its independent association with breast cancer risk in a nationally representative civilian cohort has not been quantified. We aimed to evaluate the cross-sectional relationship between PLR and breast cancer prevalence among U.S. adults and to examine whether this association is modified by sociodemographic or clinical factors. We analyzed 14,152 women aged ≥20 years from the continuous National Health an
