INTRODUCTION: Fanconi anemia (FA) is the most prevalent inherited disorder leading to bone marrow failure, resulting from a rare autosomal recessive genetic condition that affects all three types of blood cells. A key characteristic of FA is the body's heightened sensitivity to DNA-damaging agents, particularly those that induce crosslinking, which serves as an important diagnostic marker. Children at higher risk-such as those with unexplained growth delays, congenital defects, or a family history of FA-can significantly benefit from genetic testing.
METHODS: This study involved 140 pediatric

We aimed to systematically review literature investigating the impact of COVID-19 on insulin resistance and beta-cell dysfunction in humans. Ovid MEDLINE and Embase were searched for studies published between December 2019 and May 2024. Observational studies examining adults with no history of type 2 diabetes comparing the development of insulin resistance and beta-cell dysfunction between COVID-19 exposed groups vs. controls were included. Risk of bias was assessed using adapted Newcastle-Ottawa and Joanna Briggs Institute scales. Among 6901 studies screened, 10 met the inclusion criteria. Ac

AIMS: Antibody-negative type 1 diabetes (T1D) is a provisional diagnosis with unclear etiology. This study aimed to determine the prevalence and phenotypic characteristics of monogenic diabetes within a large antibody-negative T1D cohort.
METHODS: A total of 482 antibody-negative T1D patients were included from a clinically diagnosed T1D cohort. Targeted sequencing using a custom gene panel covering 36 genes and the mitochondrial 3243 A > G mutation was performed. Demographic, metabolic, and human leukocyte antigen (HLA) data were analyzed. Beta-cell function was assessed in patients with mon

MODY (Maturity-Onset Diabetes of the Young) is characterized by autosomal dominant mode of inheritance, early onset of diabetes in the absence of autoimmunity directed to pancreatic β-cells, impaired insulin secretory capacity, however, maintained over time, and extra-pancreatic manifestations in some patients. Its prevalence has been estimated 0.6% to 6.5% of all diabetes in Europe and the USA. Pathogenic variants in the genes encoding glucokinase or transcription factors HNF1A or HNF4A are responsible for the majority of cases of monogenic forms of diabetes referred to as MODY. The objective

INTRODUCTION: This study aims to explore the relationships between various obesity-related indicators and gestational diabetes mellitus (GDM), as well as assess their ability to predict GDM development.
MATERIALS AND METHODS: Data from the 2007-2020 National Health and Nutrition Examination Survey included 4,000 women with either a history of live birth or a gestational age of at least 28 weeks. We used weighted multivariable logistic regression to examine the links between nine obesity indicators-such as body roundness index (BRI), relative fat mass (RFM), lipid accumulation product (LAP), v

OBJECTIVE: It has been postulated that pregnancies may contribute to the lifetime risk of type 2 diabetes mellitus (T2DM) in women, although direct evidence is lacking. In this context, we hypothesized that pregnancy may impact the long-term natural history of the pathophysiologic determinants of T2DM: insulin sensitivity and β-cell function. We sought to evaluate 10-year trajectories of these features in parous women with and without an additional pregnancy.
RESEARCH DESIGN AND METHODS: Metabolic characterization was done in 303 women at 1 year postpartum and on three or more occasions over

Osteoporosis (OP), a metabolic bone disease characterized by low bone mineral density (BMD), has an underexplored link with the De Ritis ratio (aspartate aminotransferase/alanine aminotransferase) - a marker of metabolic and inflammatory status. This study investigated the association between the De Ritis ratio and both OP and BMD in middle-aged and older adults. BMD was assessed by dual-energy X-ray absorptiometry, and OP was diagnosed based on a T-score ≤-2.5. First, multivariate logistic regression was employed to estimate the adjusted odds ratio (95% confidence interval) for OP in relation

AIMS: Cardiometabolic pregnancy complications, including gestational diabetes mellitus (GDM) and hypertensive disorders of pregnancy (HDP), share similar risk factors and increase cardiometabolic risk in women. Medical/lifestyle management reduces their risk but requires early identification of high risk individuals. This study aimed to develop a risk prediction model identifying women at high risk of developing GDM and/or HDP.
METHODS: A composite prognostic risk prediction model for developing GDM and/or HDP comprising first trimester sociodemographic and clinical variables was developed us

INTRODUCTION: Pheochromocytoma is a well-established cause of secondary hypertension and can lead to dysglycemia. However, its initial presentation as atypical diabetes, characterized by severe glycemic instability, is uncommon and poses a significant diagnostic challenge.
CASE PRESENTATION: A 49-year-old non-obese Chinese woman presented with a 15-month history of progressive polyuria and polydipsia. Initially diagnosed with type 2 diabetes, she developed significant glycemic variability with recurrent hypoglycemia and profound hyperglycemia, alongside a rapid decline in β-cell function. Dur

BACKGROUND: Road injuries are a leading cause of mortality and morbidity worldwide. Years of international efforts have aimed to strengthen policy engagement, including the 2020 UN General Assembly's proclamation of the Second Decade of Action for Road Safety (2021-30), targeting a 50% reduction in road traffic deaths and serious injuries by 2030. The aim of this study is to provide estimates to monitor progress and identify intervention gaps.
METHODS: As part of the Global Burden of Diseases, Injuries, and Risk Factors Study 2023, we estimated incidence, mortality, and morbidity of road inju

BACKGROUND/OBJECTIVES: Autoimmune Polyendocrinopathy with Candidiasis and Ectodermal Dystrophy is an extremely rare autosomal recessive disorder caused by inborn errors of immunity. It is due to a loss-of-function mutation in the AIRE autoimmune regulator gene. Its manifestations include autoimmunity affecting endocrine glands, in addition to non-endocrine manifestations including dental enamel hypoplasia, alopecia areata, hepatitis, and chronic mucocutaneous candidiasis. Globally, 10 cases per million are affected by this condition, with higher incidence in highly consanguineous populations.

Human menopause and the prolonged post-reproductive lifespan (PRLS) are distinctive features of female aging with profound implications for long-term health. Evolutionary theories, such as the Grandmother Hypothesis and other models (e.g., intergenerational reproductive conflict), suggest menopause evolved under ancestral conditions of high extrinsic mortality in early life, short lifespans, and inclusive-fitness benefits derived from kin investment. In such environments, postmenopausal survival was brief, minimizing selective pressure from estrogen deficiency. However, modern advancements in

BACKGROUND: Despite a general improvement in the HIV burden over the past two decades, gendered social determinants including intimate partner violence (IPV) continue to affect women's vulnerability to HIV, which could be further exacerbated as resources dwindle over the coming years. Our study aimed to quantify recent trends in the HIV burden, the magnitude of the HIV burden associated with IPV, and the potential impact of declining financial support globally.
METHODS: Using the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2023 framework, we estimated annual HIV incidenc

BACKGROUND: Diabetes mellitus, an endocrine system disease, is a common disease involving many patients worldwide. Many studies are performed to evaluate the correlation between micronutrients/macronutrients on diabetes but few of them have a high statistical population and a long follow-up period. We aimed to investigate the relationship between intake of macro/micronutrients and the incidence of type 2 diabetes (T2D) using logistic regression (LR) and a decision tree (DT) algorithm for machine learning.
METHOD: Our research explores supervised machine learning models to identify T2D patient
