BACKGROUND: Cellular imaging analysis using the traditional retrospective approach is extremely time-consuming and labor-intensive. Although AI-based solutions are available, these approaches rely heavily on supervised learning techniques that require high quality, large labeled datasets from the same microscope to be reliable. In addition, primary patient samples are often heterogeneous cell populations and need to be stained to distinguish the cellular subsets. The resulting imaging data is analyzed and labeled manually by experts. Therefore, a method to distinguish cell populations across i

Breast cancer (BC), as the most common malignant tumor among women globally, faces numerous challenges in treatment. Currently, although the combination of surgical resection with various treatment modalities such as chemotherapy, radiotherapy, and immunotherapy can improve patient survival rates, most BC patients are diagnosed at an advanced stage. Additionally, traditional small-molecule targeted anticancer drugs used during chemotherapy suffer from issues such as low target selectivity and frequent occurrence of acquired resistance, leading to suboptimal therapeutic outcomes. In recent year

Cytomegalovirus (CMV) gastritis is a rare opportunistic infection that often affects immunosuppressed patients. It is a DNA virus belonging to the Herpes family most commonly spread through contaminated bodily fluids such as blood, urine, saliva, tears, breast milk, semen, and vaginal fluids. Here, we present a case of a newly diagnosed HIV-positive patient with acquired immunodeficiency syndrome (AIDS) found to have a gastroesophageal (GE) junction ulceration with endoscopic characteristics suggestive of malignancy due to CMV gastritis in the absence of underlying malignancy. Most immunocompe

BACKGROUND: The World Health Organization (WHO) strongly recommends the use of donor human milk (DHM) for low birth weight infants when mother's own milk is unavailable or insufficient. However, the use of DHM requires the availability of human milk banks (HMBs), the majority of which are in middle and high-income countries. Developing countries offer multiple opportunities and challenges regarding the establishment and operationalization of HMBs. This study describes the experience in setting up the HMB in Uganda at St. Francis Hospital Nsambya.
METHODS: The establishment of the first HMB in

BACKGROUND: Estrogen receptor (ER)-positive (ER+) breast cancer accounts for approximately 75% of all breast cancers. Tamoxifen, a selective estrogen receptor modulator, is the standard adjuvant treatment. Although better tolerated than aromatase inhibitors, tamoxifen increases the risk of venous thromboembolism (VTE) 1.4-fold.
AIM: To assess the hemostatic imbalance induced by tamoxifen in adjuvant treatment of ER+ breast cancer.
METHOD: Twenty-five patients in remission from ER+ breast cancer under tamoxifen were included. One hundred and thirty one age- and BMI-matched healthy controls we

BACKGROUND: Toliparibizumab in combination with nab-paclitaxel (T+N) has excellent efficacy inmetastatic or recurrent triple-negative breast cancer (TNBC), but the optimal choice of sequence of therapy is unclear given the trade-offs between quality of life and cost. Cost-effectiveness analyses can quantify these tradeoffs, leading to more informed decisions. Our objective was to assess the societal cost-effectiveness of the T+N regimen for metastatic or recurrent TNBC.
METHODS: Clinical data were extracted from a multicenter, randomized, double-blind trial, TORCHLIGHT (NCT04085276). Patients

Introduction: Recent advances in sequencing technologies have significantly increased our capability to acquire large amounts of genetic data. However, the clinical relevance of the generated data continues to be challenging particularly with the identification of Variants of Uncertain Significance (VUSs) whose pathogenicity remains unclear. In the current report, we aim to evaluate the clinical relevance and the pathogenicity of VUSs in DNA repair genes among Tunisian breast cancer families. Methods: A total of 67 unsolved breast cancer cases have been investigated. The pathogenicity of VUSs

A hallmark of cancer is the dysregulation of the cell cycle. The CDK4/6 inhibitor palbociclib is approved for treating advanced estrogen-receptor-positive breast cancer, but its success is limited by the development of acquired resistance owing to long-term therapy despite promising clinical outcomes. This situation necessitates the development of potential combination strategies. Here, we report that didox, an inhibitor of ribonucleotide reductase in combination with palbociclib, can overcome palbociclib resistance in ER-positive and ER-negative breast cancers. This study shows didox downregu

INTRODUCTION: SARS-CoV-2 infection increases the risk of worse outcomes in cancer patients, including those with breast cancer. Our previous study reported that the SARS-CoV-2 membrane protein (M-protein) promotes the malignant transformation of triple-negative breast cancer cells (triple-negative BCC).
METHODS: In the present study, the effects of M-protein on the ability of extracellular vesicles (EV) derived from triple-negative BCC to regulate the functions of tissue stem cells facilitating the tumor microenvironment were examined.
RESULTS: Our results showed that EV derived from M-prote

KEY CLINICAL MESSAGE: In a patient with metastatic breast cancer, an acquired BRCA mutation in the BRCA gene was detected, resulting in benefits from olaparib treatment. This underscores the importance of ongoing genetic phenotype testing after paclitaxel chemotherapy.
ABSTRACT: Triple-negative breast cancer (TNBC) is associated with a poor prognosis and elevated mortality risk. BRCA mutations are commonly regarded as prevalent mutations in TNBC patients, strongly associated with congenital familial heredity. Dynamic changes in mutation sites, however, are rarely reported. In this case report

Breast cancer is the most prevalent malignancy among women worldwide. Despite significant advances in treatment, it remains one of the leading causes of female mortality. The inability to effectively treat advanced and/or treatment-resistant breast cancer demonstrates the need to develop novel treatment strategies and targeted therapies. Centrosomes and their associated proteins have been shown to play key roles in the pathogenesis of breast cancer and thus represent promising targets for drug and biomarker development. Centrosomes are fundamental cellular structures in the mammalian cell that

BACKGROUND: In the precision medicine era, identifying predictive factors to select patients most likely to benefit from treatment with immunological agents is a crucial and open challenge in oncology.
METHODS: This paper presents a pan-cancer analysis of Tumor Mutational Burden (TMB). We developed a novel computational pipeline, TMBcalc, to calculate the TMB. Our methodology can identify small and reliable gene signatures to estimate TMB from custom targeted-sequencing panels. For this purpose, our pipeline has been trained on top of 17 cancer types data obtained from TCGA.
RESULTS: Our res

BACKGROUND: Fast adaptation of glycolytic and mitochondrial energy pathways to changes in the tumour microenvironment is a hallmark of cancer. Purely glycolytic ρ0 tumour cells do not form primary tumours unless they acquire healthy mitochondria from their micro-environment. Here we explored the effects of severely compromised respiration on the metastatic capability of 4T1 mouse breast cancer cells.
METHODS: 4T1 cell lines with different levels of respiratory capacity were generated; the Seahorse extracellular flux analyser was used to evaluate oxygen consumption rates, fluorescent confocal

One of the most dangerous conditions in clinical practice is breast cancer because it affects the entire life of women in recent days. Nevertheless, the existing techniques for diagnosing breast cancer are complicated, expensive, and inaccurate. Many trans-disciplinary and computerized systems are recently created to prevent human errors in both quantification and diagnosis. Ultrasonography is a crucial imaging technique for cancer detection. Therefore, it is essential to develop a system that enables the healthcare sector to rapidly and effectively detect breast cancer. Due to its benefits in
