Evolution in metazoans of the TRPM channel family involves multiple gains and losses of genes and domains.

Transient receptor potential (TRP) ion channels of the melastatin family (TRPM) have eight members in mammals with a broad spectrum of functions. We investigated the evolution of this complex gene family across metazoans. The characteristic aminoterminal melastatin domain and the carboxyterminal NUDT9 homology domain with similarity to ADP-ribose pyrophosphatase were added to the common ancestor of TRPM and its sister channel TRPS. Gene duplications before the origin of bilaterians resulted in four TRPM genes: α, β, βlike, and γ. The two latter were discovered in this study. All four and TRPS

Safety evaluation of the food enzyme AMP deaminase from the genetically modified Bacillus subtilis strain CCTCC M 2023264.

The food enzyme AMP deaminase (AMP aminohydrolase; EC 3.5.4.6) is produced with the genetically modified Bacillus subtilis strain CCTCC M 2023264 by Angel Yeast Co., Ltd. The production strain of the food enzyme contains multiple copies of three acquired antimicrobial resistance genes, therefore it does not fulfil the requirements of the qualified presumption of safety approach to safety assessment. However, considering the absence of viable cells and DNA from the production organism in the food enzyme, this is not considered to be a risk. As no other concerns arising from the microbial source

VGDS-PointNet++ for organ segmentation and phenotypic trait estimation in greenhouse tomato seedlings.

INTRODUCTION: To accurately segment point clouds and quickly calculate leaf length and stem diameter, thereby enabling phenotypic analysis and variety selection of greenhouse tomato plants, this paper proposes a voxel grid downsampling (VGDS)-PointNet++-based model for point cloud segmentation and trait calculation. METHODS: The point clouds of the tomato canopy were acquired using a depth camera. After labeling, point cloud augmentation was performed, and the tomato point cloud dataset (TPCD) containing 1,552 sets of data was rebuilt. Voxel grid downsampling was applied to replace the origin

Development of a high-grade glioma preclinical surgery model using an inducible KRAS/TP53 Oncopig.

INTRODUCTION: Surgery remains the primary treatment modality for high-grade glioma; yet, Q10 median overall survival has seen only modest improvement over the past few decades. Lack of reliable preclinical models that recapitulate the volumes and infiltrative nature of human tumors is a persistent challenge for development and evaluation of new surgical techniques. In this context, the Oncopig model has been shown to produce realistic tumors in other organs. It is a transgenic model that encodes for the oncogenic P53 and KRAS mutations which can be switched on through delivery of Cre-recombina

Safety evaluation of the food enzyme glutaminase from the genetically modified Bacillus subtilis strain Glu3-3.

The food enzyme glutaminase (l-glutamine amidohydrolase; EC 3.5.1.2) is produced with the genetically modified Bacillus subtilis strain Glu3-3 by Angel Yeast Co., Ltd. The production strain of the food enzyme contains multiple copies of acquired antimicrobial resistance genes. The food enzyme was considered free of viable cells of the production strain, but the absence of DNA was not demonstrated. As no other concerns arising from the microbial source or from the manufacturing process have been identified, the Panel considered that toxicological tests are not needed for the assessment of this

Loss of the ferripyochelin receptor FptA drives reduced cefiderocol susceptibility and impairs fitness in Pseudomonas aeruginosa PA14.

Pseudomonas aeruginosa is an opportunistic human pathogen and a frequent cause of multidrug-resistant infections. This organism continues to evade antimicrobial therapy despite the clinical introduction of new antipseudomonal antibiotics over the past several years. One of these agents is cefiderocol (FDC), a novel siderophore-cephalosporin conjugate antibiotic that was designed to overcome both intrinsic and acquired β-lactam resistance mechanisms in P. aeruginosa. However, studies have demonstrated that inactivation of energy transducer protein (TonB)-dependent receptors, most notably the ca

From intestinal immune niches to systemic inflammation: programming cellular plasticity and immune redistribution across organs.

Immune-mediated inflammatory diseases have traditionally been investigated within organ-specific frameworks. Although increasing evidence has revealed extensive communication along individual gut-organ axes, these interactions are often described through separate mechanisms, including microbial dysbiosis, barrier disruption, soluble mediators, and immune-cell redistribution. A central challenge remains to determine how intestinal immune states are established and how they are translated into organ-specific inflammatory responses. In this review, we propose that the intestine functions as a dyn

Editing Candida: Origins and Advances of CRISPR Tools.

Pathogens causing candidiasis encompass a diverse group of ascomycetous yeasts that have become essential models for studying fungal adaptability, pathogenicity, and host-pathogen interactions. Although many candidiasis-promoting species exist as commensals within host microbiota, several have acquired virulence traits that enable opportunistic infections, positioning them as a leading cause of invasive fungal disease in humans. Deciphering the molecular and genetic determinants that underpin the biology of organisms responsible for candidiasis has long been a central objective in medical and

Interleukins in community-acquired pneumonia: from biomarkers to precision medicine.

Community-acquired pneumonia (CAP) is still a leading cause of death due to infection globally, yet precise severity assessment remains a significant clinical problem. More than any other group of cytokines, interleukins are central to the regulation of inflammation and shed light on this intricate pathology. In the present review we summarize the biological and clinical characteristics of some of the principal interleukins (ILs) in CAP, classified primarily according to their physiological activity as pro-inflammatory (IL-2, IL-6, IL-8 and IL-12), anti-inflammatory (IL-7, IL-10 and IL-37), du

Emergence of Multidrug-Resistant Pathogens in a Tertiary Care Hospital in Northern India: A Comparative Study Between Critical Care Units and Wards.

Background and aim Multidrug-resistant organisms (MDROs) represent a major global public health concern, largely driven by inappropriate antibiotic use, over-the-counter availability of antimicrobials, and the absence of effective prescription audit systems. MDROs are microorganisms, most commonly bacteria, that exhibit resistance to one or more classes of antimicrobial agents. The burden of multidrug-resistant infections has increased worldwide, with marked rises in both healthcare-associated and community-acquired infections, particularly in regions with high levels of antibiotic misuse. Impact of COVID-19 on sputum isolates and hospital outcomes among patients with pneumonia in Sheffield, United Kingdom: a retrospective cohort study.

BACKGROUND AND AIM: The COVID-19 pandemic caused by SARS-CoV-2 severely affected healthcare globally. Strong links have been suggested between the pandemic and changes in the epidemiology of organisms responsible for non-COVID-19 pneumonia. In this study, we examined temporal changes in organisms recovered from sputum culture, integrating laboratory data from all submitted sputum specimens with clinical data from a subset of patients diagnosed with community-acquired pneumonia (CAP). METHOD: A retrospective study was conducted from August 2017 to May 2024 using data derived from samples from

A hybrid combination of in vitro cultured buccal mucosal cells using two different methodologies, complementing each other in successfully repairing a stricture-inflicted human male urethral epithelium.

BACKGROUND: Autologous buccal mucosa cell transplantation has emerged as a promising treatment strategy for urethral stricture disease. However, ambiguity has persisted regarding the optimal cell type and culture conditions that aid successful urethral repair. Clinical study of our previously reported cell-based endoscopic approach, the buccal epithelium expanded and encapsulated in scaffold-hybrid approach to urethral stricture (BEES-HAUS), demonstrated durable epithelial regeneration and long-term urethral patency. The present work provides mechanistic insights supporting the BEES-HAUS appro

Reliability of EEG in brain death diagnosis: an analysis of clinical and technical factors.

INTRODUCTION: In brain death determination (BDD), electroencephalography (EEG) is a commonly used diagnostic method in Germany for proving irreversibility of previously clinically proven loss of brain function. Despite the comprehensive availability of EEG in intensive care units, the reliability of EEG readings is often limited by biological or technical artefacts. This study aimed to determine the detection rate of electrocerebral inactivity (ECI) with a special focus on reliability and challenges of using EEG in BDD. METHODS: A retrospective monocentric analysis of all BDD EEG data acquire

Exploring the Effectiveness of Imipenem/Relebactam in Patients with Antimicrobial-Resistant Hospital-Acquired Infections: Findings from Systematic Literature Reviews.

Introduction: Infections attributed to multidrug-resistant organisms have resulted in a significant clinical burden, high mortality, and excessive costs. Identifying the most appropriate and efficacious treatments will aid in reducing these burdens. Imipenem/cilastatin + relebactam (I/R) is used against multidrug-resistant infections providing an alternative option which may support patients where traditional treatments are no longer effective. Objective: The objective was to evaluate the efficacy of I/R for complicated urinary tract infections, complicated intra-abdominal infections, hospital

Severe pneumonia in children with influenza A-Staphylococcus aureus coinfection: case report.

Severe community-acquired pneumonia caused by coinfection with influenza A virus and Staphylococcus aureus (S. aureus) is rare but may lead to rapid clinical deterioration and high mortality. We report two pediatric cases of severe pneumonia associated with influenza A virus-S. aureus coinfection. Both patients initially presented with nonspecific influenza-like symptoms, followed by rapid progression to acute respiratory failure. One case was characterized by necrotizing pneumonia complicated by pneumothorax and empyema, with S. aureus isolated from respiratory specimens and pleural fluid. Th

Gastrointestinal Cryptococcosis in immunosuppressed patients.

Cryptococcus neoformans is a yeast-like fungus found in soil contaminated with avian droppings. It most often produces disseminated disease in immunocompromised hosts, particularly individuals with impaired T-cell mediated immunity such as those with HIV infection, patients receiving corticosteroids or other immunosuppressive therapy, and solid organ transplant recipients. The usual clinical manifestations are meningoencephalitis and pulmonary involvement, and it remains the most common systemic mycosis in patients with acquired immunodeficiency syndrome. Although the organism can involve near

Influenza A-Associated Thrombotic Microangiopathy With Normal a Disintegrin and Metalloproteinase With Thrombospondin Type 1 Motif 13 (ADAMTS13) in a Young Male Patient: A Case Report.

Thrombotic microangiopathy (TMA) is a life-threatening syndrome characterized by microangiopathic hemolytic anemia, thrombocytopenia, and end-organ injury resulting from widespread microvascular thrombosis. It encompasses several entities, including thrombotic thrombocytopenic purpura (TTP), atypical hemolytic uremic syndrome (aHUS), and secondary forms triggered by infections, medications, or systemic disease. Viral infections, including influenza A, have been increasingly recognized as rare triggers of secondary TMA, through endothelial injury and immune dysregulation, even in the absence of

Tinea Corporis and Tinea Pedis Can Masquerade As Other Cutaneous Conditions in Decedents: Forensic Dermatology of Dermatophyte Infections.

Decedents can have lesions of their skin, mucosa, hair, or nails that may be related to their cause of death; alternatively, these lesions may be coincidentally present and not have associated forensic implications. Superficial fungal infection of the skin, such as tinea corporis and tinea pedis, can mimic other cutaneous conditions, including diseases, neoplasms, or infection. Two decedents had superficial fungal infections that mimicked other conditions, and the cutaneous presentation of their dermatophyte infection was selected to be reported based on their illustrative value.
