Current Status and Future Prospects of Immune Checkpoint Inhibitors Research in the Treatment of Multiple Myeloma.

Multiple myeloma (MM) is an aggressive hematological tumor characterized by an abnormal proliferation of bone marrow plasma cells accompanied by an overproduction of monoclonal immunoglobulins. The clinical manifestations of MM are varied, with the main ones being anemia, bone pain, renal insufficiency, infections, hemorrhage, neurological symptoms, and hypercalcemia. Chemotherapy, targeted therapy, and stem cell transplantation can provide some relief to patients. However, with the immunodeficiency and immune tolerance that occurs with the progression of the disease, MM is still not completel

Association of Race/Ethnicity on Resolving Inactive Kidney Transplant Status Prior to COVID-19.

INTRODUCTION: Minority waitlisted kidney patients are less likely to resolve an inactive status change. We investigate race/ethnicity-based differences in the reasons for and resolution of waitlist inactivation. METHODS: The Organ Procurement and Transplantation Network database was queried (December 2014-January 2020). Cause-specific Cox regression and competing risks analysis were conducted to determine the association of inactive status and race/ethnicity, stratified by specific reasons a patient was made inactive. RESULTS: Of the 94,035 patients, 52.5% experienced inactivity. 47.0% were

Imaging in Plasma-Cell Neoplasms: Italian Experts' Recommendations.

OBJECTIVES: Using suboptimal imaging modalities on the detection of bone and extra-bone myeloma lesions affects disease status recognition and early treatment decisions. Moreover, a systematic search of the evidence was not provided alongside the recommendations. The objective of this study was to assess the current real world imaging practices in Italy, influencing factors, and barriers to adopting international imaging guidelines for MGUS and MM (reported as plasma cell disorders, PCD) and to develop evidence-based, country-specific recommendations for imaging assessment of PCD patients. Clinical relevance of tissue copper, selenium, and cadmium alterations in colorectal cancer.

Imbalances in essential elements and altered levels of toxic elements are thought to play a role in colorectal cancer (CRC) development. This study assessed the concentrations of selected elements in CRC tissues and explored their associations with clinical parameters. Paired samples of tumor tissue and adjacent noncancerous tissue were collected from 62 patients with confirmed CRC. After microwave-assisted acid digestion, ten elements (Al, Cr, Mn, Fe, Ni, Cu, Zn, Se, Sr, Cd) were quantified using ICP-MS. Statistical analyses and machine-learning methods were applied to identify features discr

Single-cell profiling reveals reprogrammed hierarchy and disrupted immune-stromal ecosystem in TP53-mutated AML.

BACKGROUND: TP53-mutated acute myeloid leukemia (AML) represents one of the most adverse-risk subtypes of AML, yet the mechanisms underlying its resistance and relapse remain poorly defined. METHODS: We performed single-cell RNA sequencing on bone marrow samples from 30 de novo AML patients (11 TP53-mutated, 19 TP53-wild-type) and systematically analyzed leukemic, immune, and stromal compartments to delineate differentiation trajectories, transcriptional heterogeneity, and microenvironmental remodeling. We also performed in vitro assays to validate ferroptosis resistance, leukemia-T cell dysf

Long-term Outcomes of 3-dimensional-printed Cutting Guides for Long Bone Sarcoma Resection and Intercalary Allograft Reconstruction: An Updated Case Series.

BACKGROUND: The use of 3-dimensional (3D)-printed cutting guides for resection of long bone sarcoma may offer advantages over traditional free-hand or navigational osteotomy, including improved margin control and reconstruction accuracy. We evaluated long-term surgical and oncologic outcomes of limb salvage procedures using 3D-printed cutting guides, with updated follow-up from our previously published case series and the addition of new cases focusing on margin status, bony union, and local recurrence. MATERIALS AND METHODS: We retrospectively reviewed 9 patients from our surgical database w

Prognostic impact of clinico-genetic characteristics and donor types in NPM1-mutated AML undergoing allogeneic hematopoietic stem cell transplantation.

NPM1-mutated acute myeloid leukemia (AML) is a distinct entity with specific biological and clinicopathological features. Prognosis varies depending on the associated mutations and clinical characteristics. This study evaluated the prognostic factors of NPM1-mutated AML in patients who underwent allogeneic hematopoietic stem cell transplantation (allo-HSCT). We enrolled 139 patients with NPM1-mutated AML who underwent allo-HSCT. The median age at allo-HSCT was 47 years with a median follow-up of 1,134 days. The 3-year cumulative incidence of relapse (CIR), leukemia-free survival (LFS), and ove

Comprehensive geriatric assessment (CGA) scale does not effectively identify elderly AML patients suitable for intensive induction chemotherapy - a multicenter study from Polish Adult Leukemia Group.

Standard scales, such as ECOG Performance Status, Hematopoietic Cell Transplantation Comorbidity Index (HCT-CI), and Activities of Daily Living (ADL), are commonly used to stratify patients with acute myeloid leukemia (AML) for intensive chemotherapy (IC). Comprehensive geriatric assessment (CGA) may further identify patients at risk of treatment-related toxicity, but it is underutilized due to time constraints. We aimed to evaluate the predictive relevance of CGA domains in elderly AML patients selected for IC using ECOG, HCT-CI, and ADL. In this prospective study, patients aged 60–75 were el

Salivary and urinary microRNAs as non-invasive biomarkers of minimal residual disease in pediatric acute lymphoblastic leukemia.

The measurement of minimal residual disease (MRD) in bone marrow serves as a significant predictor of relapse in acute lymphoblastic leukemia (ALL). MicroRNAs, which are secreted in urine and saliva, typically reflect the cellular state and function, thereby demonstrating potential as non-invasive biomarkers for predicting MRD status. This study aims to evaluate the utility of circulating microRNAs in saliva and urine as MRD biomarkers in pediatric patients with ALL. The study cohort comprised 106 patients under 18 years of age diagnosed with ALL at a national pediatric referral hospital in Li

Kinetics of De Novo Bone and Bone Marrow Niche Formation With Hybrid Click Cryogels.

Compromised bone marrow niches following irradiation limit hematopoietic stem cell transplantation (HSCT) success by delaying immune reconstitution. Strategies to rebuild functional marrow environments are essential to support hematopoietic stem cell (HSC) maintenance. However, the fundamental relationship between bone formation and bone marrow niche development, and how these processes are modulated by key biological variables, remains poorly understood. Here, we present an alginate-gelatin cryogel platform that provides sustained BMP-2 release for ectopic osteogenesis and use it to systemati

Matching-Adjusted Indirect Comparison of Olutasidenib and Ivosidenib in Isocitrate Dehydrogenase 1-Mutated Relapsed/Refractory Acute Myeloid Leukemia.

INTRODUCTION: Olutasidenib and ivosidenib are isocitrate dehydrogenase 1 (IDH1) inhibitors approved for relapsed/refractory (R/R) IDH1 mutant (IDH1m) acute myeloid leukemia (AML). METHODS: A matching-adjusted indirect comparison estimated relative treatment effects using registrational Phase I/II data for olutasidenib (Study 2102-HEM-101; individual patient data) and ivosidenib (Study AG120-C-001; study-level data) since a head-to-head trial is unlikely. Beyond readthrough: ataluren restores mitochondrial function and reduces oxidative stress in FANCA-mutated cells via mTOR-DRP1 modulation.

Fanconi anemia (FA) is a rare inherited bone marrow failure syndrome characterized by genomic instability, mitochondrial dysfunction, and oxidative stress. While the therapeutic potential of ataluren, a translational readthrough-inducing drug, has been investigated in FA cells carrying nonsense mutations, its broader metabolic impact remains unclear. Here, we demonstrate that ataluren (tested at 2.5, 5, and 10 μM) modulates cellular energy metabolism and redox homeostasis in FA lymphoblasts harboring either nonsense or missense mutations in the FANCA gene. At low doses (2.5 μM for 72 h), atalu

Real-World Analysis of Outcomes of Venetoclax+Azacitidine Versus 7+3 Induction in Acute Myeloid Leukemia.

BACKGROUND: Venetoclax plus azacitidine (V+A) is standard for older, intensive-ineligible patients with acute myeloid leukemia (AML). Its expanding use in younger, curative-eligible adults lacks comparative evidence against conventional 7+3 induction, raising uncertainty about potential survival compromise. METHODS: We performed an age-stratified, retrospective comparative effectiveness study using de-identified TriNetX electronic health records (2018-2025). Adults receiving first-line V+A or 7+3 were propensity-matched 1:1 within prespecified age groups (18-59; ≥60) by demographics, comorbid

Chronic graft-versus-host disease: Current situation and unmet needs - A European position statement.

Chronic graft-versus-host disease (cGVHD) remains the leading cause of late morbidity and non-relapse mortality after allogeneic hematopoietic cell transplantation, despite major advances in transplant techniques and supportive care. This European position statement provides a comprehensive and forward-looking synthesis of the evolving biology, epidemiology, diagnosis, and management of cGVHD, while highlighting critical unmet needs that impede progress. cGVHD arises from a complex interplay of immune dysregulation, aberrant tissue repair, and progressive fibrosis, resulting in a heterogeneous

Peripheral Lymphocyte Dynamics in the Immune Microenvironment of Multiple Myeloma During Autologous Stem Cell Transplantation.

Background and objective Autologous stem-cell transplantation (ASCT) remains the standard of care for multiple myeloma (MM), yet it is not curative. Over the past decade, there has been growing interest in immunotherapy-based strategies, highlighting the need for predictive biomarkers to tailor post-ASCT treatment. Characterization of immune status via peripheral lymphocyte immunophenotyping is a critical step in this process. This study aimed to characterize peripheral blood lymphocyte immunophenotypes in MM patients undergoing ASCT at baseline, during stem cell mobilization, and on day +21 p

Heterogeneous survival outcomes in molecularly defined acute myeloid leukemia with myelodysplasia-related changes.

Acute myeloid leukemia with myelodysplasia-related changes (AML-MRC) is a high-risk AML subtype defined by cytogenetic or genetic changes. We evaluated the relevance of AML-MRC classifications in a contemporary cohort of intensively treated patients with AML. We retrospectively reviewed 496 consecutive patients receiving induction chemotherapy for AML between 2016 and 2023 at a single center. Patients were categorized using both World Health Organization 2017 (MRC-2017) and International Consensus Classification 2022 (MRC-2022) criteria. Outcomes were compared using χ2 or Kaplan-Meier.
