Estrogen-Regulated Proline-Rich Acidic Protein 1 in Endometrial Epithelial Cells Affects Embryo Implantation by Regulating Mucin 1 in Mice.

This study explores the regulatory role of proline-rich acidic protein 1 (Prap1) effects on embryo implantation. A high estrogen model and ovariectomized mice were employed to demonstrate that estrogen regulates Prap1 expression. Uterine tissues were collected from E 1.5 (the presence of a vaginal plug was recorded as embryonal day 0.5, E 0.5) to E 7.5 to detect the Prap1 expression pattern in early pregnancy using qRT-PCR. Embryo adhesion was assessed through uterine perfusion of PRAP1 protein and Prap1 overexpression in endometrial epithelial cells (EECs). The data showed that Prap1 expressi

Fibroblast growth factors and endometrial decidualization: models, mechanisms, and related pathologies.

The onset of pregnancy is marked by the formation of a zygote, while the culmination of gestation is manifested by the delivery of a fetus. Meanwhile, a successful pregnancy entails a meticulously coordinated sequence of events from embryo implantation to sustained decidualization of the uterus to placental development and childbirth. The decidual reaction, a pivotal process occurring within the endometrium during pregnancy, is finely regulated by sex steroids and cytokines. Notably, fibroblast growth factors (FGFs), particularly FGF2, play a critical role in this physiological cascade. Dysreg

Kidney transplant from donation after circulatory death: a novel pre-transplant combined clinico-pathological predictive score.

BACKGROUND: Kidney donation after circulatory death is a valid alternative to donation after brain death. However, ischemia-reperfusion injury can damage the graft and make it non-viable. The Karpinski score, currently used to evaluate kidney quality, does not account for this type of damage and is, therefore, insufficient for predicting expected functional recovery. METHODS: Sixty-seven kidney biopsies from donors  after circulatory death and seventy-two kidney biopsies from donors after brain death were analyzed histologically. A Karpinski score was assigned to all samples. Additionally, bi

A Diagnosis of Maturity-onset Diabetes of the Young Type 5 Provides Clarity and Broadens Reproductive Options.

Maturity-onset diabetes of the young type 5 (MODY5) is a rare form of monogenic diabetes caused by variants and deletions of the HNF1B gene. It is associated with a range of features including cystic renal disease, urogenital anomalies, and neurodevelopmental delay. Pregnancies in the presence of HNF1B gene variants and deletions have a significant risk of adverse maternal and fetal outcomes. Accurate diagnosis allows for genetic counseling, individualized therapies, and reproductive testing options. We report the case of a woman who was recognized as having MODY5 in her first pregnancy. The f

Osteopontin expression in prepubertal and adult dog testes and the effect of slow-release deslorelin implants (Suprelorin® 4.7 mg).

Osteopontin (OPN) is a highly phosphorylated glycoprotein expressed in several cells, tissues, and tissue fluids, including the male reproductive system. Recent studies have indicated that OPN may be a potential fertility marker in male dogs. However, OPN expression and localization during testicular growth are still unclear, and the effect of pharmacological castration on testicular OPN expression in male dogs has not been studied to date. This study aimed to investigate and compare the expression and protein immunolocalization of OPN in the prepubertal (PRE) and adult dog (AD) testes, while

The role of leptin in mammalian oocyte developmental competence and pre-implantation embryo development.

Leptin is primarily involved in energy homeostasis and has been implicated in fertility. Leptin- and leptin receptor-deficient mouse models have demonstrated that expression of leptin signalling in the central nervous system is essential for ovulation. However, evidence from ruminants and other species gathered from models with physiological leptin signalling suggests that modulation of leptin may not play a major role in the attainment of ovulation in post-pubertal individuals, despite influencing ovarian steroidogenesis and folliculogenesis. In vitro studies indicate that leptin concentratio

The Role of β-Core Fragment hCG in Embryo Implantation and Early Pregnancy.

Human chorionic gonadotropin (hCG) is a pregnancy biomarker, and five forms of this hormone are involved in female physiological regulation. β-core fragment hCG (bcf-hCG) is a fragment of hCG whose biological role in female reproduction has not been completely elucidated. This study aimed to investigate its role in embryo implantation and maintenance of a pregnancy-supportive environment. We analyzed the protein expression pattern of bcf-hCG in the intrauterine environment during early pregnancy by performing western blotting and immunohistochemistry with a monoclonal anti-bcf-hCG antibody. We

Modulation of uterine responses in dogs: The divergent effects of semen and embryonal signals on steroidogenic receptors and selected immune system-related factors.

Although dogs lack the classical pregnancy recognition via embryo-derived anti-luteolytic signaling, precise embryo-maternal communication remains essential in establishing pregnancy. While previous studies confirmed uterine responses to pre-attachment embryos versus semen-exposed non-pregnant controls, they did not include non-mated, non-pregnant controls. Following the hypothesis that embryo-derived signaling differs from the responses induced by semen, the present project included three groups of dogs (d. 10-12 of pregnancy/corresponding dioestrus): non-mated, mated and pregnant, and mated

In vivo 3D imaging of ovarian cancer outgrowth in transgenic mouse model with optical coherence tomography.

SIGNIFICANCE: Peritoneal dissemination is the major mechanism of how ovarian cancer (OC) spreads. It features tumor outgrowths in the form of multicellular spheroids, their detachment from the primary site, and their implantation in the peritoneal cavity. To understand this process, analyzing the outgrowths at their native locations within the female reproductive system is essential. However, in vivo study of the OC outgrowths remains unattainable primarily due to the lack of in vivo imaging approaches to probe such small tumor structures at a high resolution. AIM: We address this technical c

Oocyte maturation triggers in high-responders: a report on 1,217 consecutive cycles.

ABSTRACT: Using a large patient series, we aimed to evaluate, in a high-responder population, the effect of triggering oocyte maturation with human choriogonadotropin (hCG) or with a gonadotropin-releasing hormone agonist (GnRHa). We analyzed data from 683 intended fresh embryo transfer cycles (ETCs) and 534 frozen-thawed embryo transfer (FET) cycles. The rates of ovarian hyperstimulation syndrome (OHSS), and the embryological, clinical, and newborn outcomes were compared. Considering the type of oocyte maturation trigger and embryo destination, cycles were divided into five groups. Cycles usi

Deciphering immune tolerance in allogeneic pig pregnancy.

Embryo transfer (ET) has transformed swine biotechnology, enabling genetic advancements and disease control. However, its success remains inconsistent, partly due to immune-mediated challenges at the maternal-fetal interface. This study explores the immunological environment of hemi-allogeneic pregnancies (via artificial insemination, AI) versus allogeneic pregnancies (via ET) in pigs during the critical implantation phase. Sows were categorized into groups based on pregnancy type and fetal counts, reflecting varying outcomes. Endometrial immune cell populations, including T lymphocytes, regul

Molecular Basis of Impaired Decidualization in the Eutopic Endometrium of Endometriosis Patients.

Endometriosis is a chronic gynecological disorder characterized by the presence of endometrial tissue outside the uterine cavity. A common feature of this pathology is the impaired decidualization of endometrial stromal cells, a critical process that prepares the uterus for embryo implantation. This decidualization defect has been mechanistically linked to progesterone resistance in endometriotic lesions. However, the presence and underlying mechanisms of decidualization defects in the eutopic endometrium of women with endometriosis remain controversial. The aim of the present study is to inte

Role of uterine NK cells in pregnancy complication.

Remodeling of blood vessels and angiogenesis play a critical role in the pregnancy process, particularly at the implantation site. Both the innate and adaptive immune systems, especially uterine natural killer (uNK) cells, are involved in this process. During the first trimester, uNK cells constitute approximately 70 % of decidual leukocytes and are differentiated from CD34+ progenitor cells. In comparison to peripheral blood NK cells, uNK cells secrete specific cytokines that promote tissue remodeling while exhibiting lower cytotoxic activity. Any disturbance in the function of uNK cells or d

Outcomes of Ultimpro Testing Implementation for Patients With Unexplained Recurrent Implantation Failure: A Single-Center Retrospective Cohort Study.

PROBLEM: A proportion of patients with unexplained recurrent implantation failure (uRIF) may have endometrial immune dysregulation amenable to treatment. Ultimpro (Matrice lab Innove, Paris, France) is an endometrial immunophenotype test which identifies the absence or presence and type of immune dysregulation, proposing personalized protocol adjustments for a future embryo transfer (ET). METHOD OF STUDY: 2.5-year single-center retrospective cohort analysis of 121 patients who underwent Ultimpro testing as a pilot study (January 2021-May 2023). Group 1 included 93 patients with uRIF, and Grou

Self-healing, degradable, and implantable tissue-like sensor for monitoring of abnormal bladder activity.

Real-time monitoring of bladder activity is essential for assessing and diagnosing lower urinary tract dysfunction. However, traditional clinical monitoring methods that rely on repeatedly invasive catheters can cause considerable physical and psychological discomfort for patients. In this study, we present a tissue-like implantable sensor that enables real-time bladder activity monitoring while eliminating the need for repeated invasive procedures. The sensor incorporates a tissue-like antifouling, self-healing, and biodegradable zwitterionic elastomer through strategic molecular integration

Distinct microbiome profiles on vaginally inserted polypropylene midurethral mesh slings compared to vaginal, urinary, and skin microbiomes.

Midurethral slings are widely used in the treatment of stress urinary incontinence in women. However, little is known about the microbiomes that develop on these implants, their relationship to the urinary and vaginal microbiomes, or their potential role in mesh-related complications. In this study, we characterized the microbiomes of explanted midurethral slings and examined associations with clinical complications. Seventy-four women provided a total of 397 samples, including explanted mesh, urine, and swabs from the vagina and groin or suprapubic skin. Participants were categorized into cli

Effects of reversine and proTAME treatment on chromosome segregation during mouse oocyte maturation.

Aneuploidy in oocytes is a leading cause of implantation failure, miscarriage and congenital disorders. During meiosis, proper timing of chromosome segregation is regulated by the spindle assembly checkpoint (SAC) and the anaphase-promoting complex/cyclosome (APC/C). However, how pharmacological manipulation of these regulatory pathways affects aneuploidy remains incompletely understood. In this study, we investigated whether SAC inhibition by reversine induces aneuploidy in mouse oocytes and whether partial inhibition of APC/C by proTAME can alleviate these errors. Germinal vesicle (GV) oocyt
