Mechanometabolic endothelial senescence as a translational framework for cardiometabolic vascular disease and vascular ageing.

Cardiometabolic vascular disease (CMVD) is used in this Review as an operational umbrella for vascular injury arising from interacting metabolic, renal and cardiovascular stress, rather than as a universally standardised diagnosis. We examine mechanometabolic endothelial senescence as a framework linking local haemodynamic vulnerability with systemic cardiometabolic load. Evidence from human vascular tissue, clinical vascular-function studies, animal models, flow-controlled systems and endothelial-cell experiments indicates that low or oscillatory shear, matrix stiffness, hyperglycaemia, dysli

School-based predictive contributors to six-minute walk test performance across pubertal stages: an explainable machine learning study in Tunisian adolescents.

BACKGROUND: The six-minute walk test (6MWT) provides a practical measure of submaximal functional exercise capacity in adolescents and reflects the integrated response of cardiovascular, respiratory, and musculoskeletal systems during walking. Traditional age-based linear models inadequately capture nonlinear, maturation-dependent interactions among anthropometric, behavioral, and physiological factors. This study aimed to (i) identify stage-specific predictive contributors to 6MWT performance using supervised machine learning, (ii) improve interpretability through explainable artificial intel

Steric control of signaling bias in the immunometabolic receptor GPR84.

Biased signaling in G protein-coupled receptors offers therapeutic promise, yet rational design of biased ligands remains challenging due to limited mechanistic understanding. Here, we report a molecular basis for controlling signaling bias at the immunometabolic receptor GPR84. We identify three structurally-matched ligands (OX04529, OX04954, and OX04539) with varying steric profiles that exhibit comparable Gi protein activation but markedly different β-arrestin recruitment capacities. A high-resolution cryo-EM structure of GPR84-Gi in complex with OX04529, complemented by molecular dynamics

Lightweight SwiM-UNet with multi-dimensional adaptor for efficient on-device medical image segmentation.

For medical image segmentation, transformer-based models have demonstrated superior performance. However, their high computational complexity remains a significant challenge. In contrast, Mamba provides a more computationally efficient alternative, though its segmentation performance is generally inferior to that of transformers. This study proposes a novel lightweight hybrid model based on U-Net, named SwiM-UNet, which represents the first Mamba–transformer hybrid model specifically designed for processing three-dimensional data. Specifically, efficient TSMamba (eTSMamba) blocks are incorpora

Detecting and understanding the dynamics of blood coagulation on a material's surface: An optical reflectance approach.

Despite the technological advancement in blood-contacting biomedical devices, issues related to thrombosis remain a persistent challenge. These devices not only include implants such as artificial heart valves and stents but also surgical tools and instruments. This makes hemocompatibility an important parameter to be considered before developing any material for a blood-contacting device. The two oldest methods, including the mechanical tilting method and the free-hemoglobin method, lack temporal accuracy and quantitative analysis. Our work gives an accurate and quantitative analysis to measu
