Leveraging high temperature for enhanced and faster detection of SARS-CoV-2 and its Omicron variant with CRISPR-AsCas12a.

CRISPR-based methods are effective for detecting SARS-CoV-2, with performance comparable to gold-standard PCR. Developing simpler, faster, and more sensitive CRISPR-based diagnostics will accelerate their adoption in field. Adjusting the temperature at which Cas proteins operate is crucial for enhancing DNA recognition kinetics, thereby improving specificity, sensitivity, and practicality. However, optimization is challenging due to the narrow temperature range for activity of wild-type Cas orthologs. Here, we show that heating the wild-type AsCas12a protein above its optimal temperature, up t

A Signal Turn-On DNA Proximity Assay That Correlates with SARS-CoV-2 Blocking Antibody Levels.

Surrogate in vitro assays have enabled the scalable testing of neutralizing antibodies to SARS-CoV-2 as a toolbox to manage the post-COVID-19 situation and future outbreaks. However, most surrogate assays use a competitive format to mimic the blocking event between the receptor binding domain (RBD) and the human angiotensin-converting enzyme 2 (ACE2) receptor. This signal "turn-off" format has a narrow dynamic range and is semiquantitative at best. In this proof-of-concept work, we introduce a signal "turn-on" dual antigen assay based on the proximity activation of a pair of RBD-oligonucleotid

Corona-Structured Magnetic Nanozymes Enable Dual-Mode Immunochromatographic Analysis for Flexible and Ultrasensitive Detection of Emerging Viruses.

Real-time and highly sensitive monitoring of pathogens in environmental and host-derived samples is critical for controlling emerging and acute infectious diseases, yet remains technically challenging. Here, we report a multifunctional nanozyme-based immunochromatographic assay (ICA) that enables rapid, universal, and multiplex detection in different scenarios through visual readouts. We constructed a corona-structured magnetic nanozyme (CSMN) with a triple signal-amplification mechanism involving "magnetic enrichment-synergistic enhancement-multiple catalytic sites," enabling efficient enrich

Psychiatric genetic liability is associated with the severity of COVID-19 and other acute respiratory infections: an observational study across five Northern European countries.

BACKGROUND: Pre-existing psychiatric disorders have been associated with the severity of acute respiratory infections, including COVID-19, particularly in hospitalized populations. However, the underlying mechanisms, especially in community-based populations, remain unknown, limiting preparedness for future pandemics. We investigated the role of genetic liability for psychiatric disorders and related traits in COVID-19 and other respiratory infection severity among individuals reporting SARS-CoV-2 testing and available respiratory symptom data. METHODS: We included population-based cohort dat

Ultrabright Quasi-Compact Quantum Dot Assembly for Sensitive and Color-Fixed Immunoassays.

Home-based immunoassay platforms constitute critical tools for the rapid screening of infectious respiratory viruses. Notably, viral loads can remain low at certain stages yet vary over several orders of magnitude during infection, and such variations are closely associated with the disease progression and transmission potential. This self-testing demand necessitates diagnostic kits that combine high sensitivity with accurate human-vision discriminability and portable terminal quantification across the entire detectable range. In response, an ultrabright label was fabricated via template-confi

Binding Free Energy Landscape of Different SARS-CoV-2 Nsp1 Variants Interacting with NXF1-RRM: Insights from Molecular Dynamics Simulations and Markov State Models.

The interaction of the SARS-CoV-2 Nsp1 protein with the host NXF1-RRM domain plays an important role in host gene expression, which influences virus replication and host immune surveillance. Thus, understanding the key factors that determine their binding affinity, such as mutations, is of great interest. In this work, we investigated the effects of three Nsp1 mutants─D33K/E36K/E37K/E41K, L27D/V28D, and R124E/K125E─on its binding to the NXF1-RRM domain using structural and energetic analysis. To quantify the affinity changes, we developed a computational framework for calculating the free ener

A yeast surface display platform for oral delivery of a SARS-CoV-2 receptor-binding domain dimer inducing mucosal and systemic immunity.

The development of scalable and stable antigen delivery systems remains a central challenge in vaccine biotechnology, particularly for oral immunization targeting mucosal immunity. Here, we report a yeast surface display (YSD)-based platform for the oral delivery of a SARS-CoV-2 receptor-binding domain (RBD) dimer. A head-to-tail tandem RBD was genetically fused to the Aga2p subunit and anchored to the cell wall of Saccharomyces cerevisiae via the Aga1p-Aga2p system, generating the YSD-RBD-dimer construct. Flow cytometry confirmed clear surface expression, with 51.1% dual-positive cells recogn

CXCL14 and Igsf8 are involved in double-stranded DNA and Alum adjuvant-mediated immune reaction.

The chemokine CXCL14 is known to bind CpG oligonucleotides and promote their uptake by dendritic cells and macrophages, thereby activating TLR9 in endosomes/lysosomes. However, it remains to be clarified whether CXCL14 also interacts with double-stranded DNA (dsDNA) and promotes its intracellular transportation. In this study, we demonstrate that CXCL14 forms a complex with dsDNA, and that this complex is internalized via immunoglobulin superfamily (Igsf) proteins. Furthermore, we found that stimulation with aluminum salt particles (Alum) adjuvant, which is involved in dsDNA-mediated innate im

Plasma-activated solutions alleviate intestinal mucosal injury induced by pathogen-host interactions by modulating redox imbalance.

BACKGROUND: SARS-CoV-2 spike binds angiotensin-converting enzyme 2 (ACE2), but the mechanisms linking spike exposure to intestinal injury remain incompletely understood. We investigated whether plasma-activated solutions (PAS) alleviate spike-associated intestinal injury by regulating antioxidant signaling. METHODS: Intestinal injury was induced by recombinant spike receptor-binding domain exposure in humanized ACE2 mice pretreated with dextran sulfate sodium. Caco-2 and HCT116 cells were used for in vitro experiments. PAS was administered orally to mice or applied as plasma-activated culture

Lipid Nanoparticle Surface Engineering with Heparosan Polysaccharides Enhances mRNA Efficacy via Reduced Endosomal Retention.

In this study, we developed a library of mRNA lipid nanoparticles (LNPs) containing clinically used ionizable lipids. We surface-modified these nanoparticles with heparosan polysaccharides and systematically compared their performance with conventional poly(ethylene glycol) (PEG)-modified formulations. Our heparosan (HEP)-coated LNPs increased mRNA expression in dendritic and macrophage cell lines by up to ∼10-fold. Mechanistic studies using live-cell imaging and intracellular nanoparticle trafficking analysis revealed that HEP-modified LNPs exhibited reduced colocalization with late endosomal

Hydrolytic Disruption of Viral Envelopes for Broad-Adaptive Antiviral Protection by an Esterase-Mimicking Nanozyme based on Copper Imidazole-Carboxylate Coordination.

Viral infections pose a persistent challenge to global public health and cause millions of infections and substantial mortality each year. There is therefore an urgent need for effective antiviral strategies to limit the transmission of viruses such as influenza A virus and SARS-CoV-2. In this work, we report a copper imidazole-carboxylate coordinated nanozyme (Cu-ImCar) with high esterase-mimicking activity and potent antiviral performance. Mechanistic studies reveal that Cu-ImCar nanozyme catalyzes the hydrolysis of lipid components in the viral envelope, leading to structural disruption and

Simultaneous determination of serotonin and its precursor tryptophan in small volumes of human serum using HPLC-ECD.

Serotonin (5-HT) is not only a key neurotransmitter in the brain but also plays an important role in peripheral tissues e.g. in the gastrointestinal (GI) tract. The multitude of processes in which 5-HT is involved explains why recent studies have linked altered 5-HT levels to Long Covid (LC) and fatigue. LC is a post-viral syndrome which can occur after a SARS-CoV-2 infection with various symptoms, amongst others including fatigue. We present an optimized bioanalytical HPLC method with electrochemical detection (ECD) for the simultaneous detection of 5-HT and its precursor tryptophan (W) in sm

6-Substituted 7-Deazapurine Ribonucleosides Capable of Non-canonical Base Recognition as Inhibitors of Respiratory RNA Viruses.

Most nucleoside antiviral analogues mimic canonical Watson-Crick base pairing to increase their likelihood of recognition and incorporation by replicative enzymes into a growing viral nucleic acid strand. Herein, we investigated whether non-canonical base recognition principles based on either shape complementarity or non-hydrogen bond interactions could also be exploited to generate novel small-molecule antivirals. Thus, the exogenous amino group of 7-deazaadenosine was systematically replaced with various 6-alkyl and 6-alkynyl substituents to assess their impact on antiviral activity against
