SARS-CoV-2 spike protein enhances HBV replication via autophagy and aggravates inflammation in CHB patients with liver dysfunction.

This study investigated the effects of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein (S protein) on hepatitis B virus (HBV) replication and immune function in patients with chronic hepatitis B (CHB), aiming to elucidate the underlying mechanisms of S protein-HBV interaction. Through in vitro cell experiments and clinical sample analyses, S protein expression was induced in HepG2.2.15 cells, leading to enhanced HBV replication; this effect was reduced by autophagy inhibitors, confirming the critical mediating role of autophagy. Furthermore, peripheral blood mono

Molecular Simulation, Pharmacophore Mapping, and 3D QSAR Modeling on Chromene-Based SERDs.

INTRODUCTION: Estrogen receptor (ERα) is known to be a legitimate therapeutic target for the treatment of ER-positive breast cancer. Although selective estrogen receptor degraders (SERDs) like fulvestrant suppress ER signaling, their limited bioavailability challenges efficacy. Additionally, activating mutations in the ERα mediate resistance to endocrine therapy. METHODS: To elucidate the structural activity relationship within a chromene-based scaffold, we conducted pharmacophore mapping and Gaussian field-based 3D QSAR modelling. The most active analogue was docked into the ERα ligand bindi

Conquering aging-related immunosenescence and tumor immune escape.

With global population aging, senescence has emerged as a key driver of tumorigenesis. Aging-associated molecular changes, including DNA damage, telomere shortening, and epigenetic dysregulation, increase malignancy, while immunosenescence and the senescence-associated secretory phenotype (SASP), reshape the tumor microenvironment to favor immune suppression and tumor escape. Aging also impairs antigen presentation, disrupts ligand-receptor signaling, and compromises tumor suppressive pathways. In the era of immunotherapy, elderly patients face reduced efficacy and increased resistance due to

Could venom-derived therapeutics resolve treatment resistance in refractory EAC?

The incidence of esophageal adenocarcinoma is rising in Western countries. Despite advances in chemotherapy and immunotherapy, the prognosis remains poor, with an overall 5-year survival rate below 15%. A major challenge is the cancer's poor and often unpredictable response to current treatments. Animal venoms represent a promising yet underexplored source of therapeutic agents, offering millions of structurally diverse and highly potent bioactive peptides that can modulate a wide array of molecular targets. However, only a small fraction of these peptides has been pharmacologically characteri

Advanced co-delivery nanocarriers to overcome drug resistance in breast cancer: carrier-mediated synergistic treatment by chemotherapy and gene therapy.

Chemoresistance is a primary factor contributing to chemotherapy failure in breast cancer. Combining chemotherapeutic agents with nucleic acids that interfere with drug resistance-related genes can effectively overcome chemoresistance in breast cancer. However, the stark differences in key physicochemical properties such as hydrophobicity versus hydrophilicity and surface charge, along with their divergent pharmacokinetic profiles, significantly compromise the synergistic therapeutic potential. Co-delivery nanocarriers enable the encapsulation of both chemotherapeutic agents and nucleic acids

A covalent molecular glue hijacks the E3 ligase MKRN2 to degrade the ribosomal protein RPS7 and induce synthetic lethality in p53-deficient NSCLC cells.

BACKGROUND AND PURPOSE: Functional inactivation of the tumour suppressor p53 drives therapeutic resistance in non-small cell lung cancer (NSCLC). While targeted protein degradation offers a potential solution, the repertoire of ligandable E3 ligases remains limited. This study aimed to identify and characterise 2,6-didehydroxyspeperomin B (DPB) as a novel covalent molecular glue that targets p53-deficient NSCLC and to elucidate its mechanism of action involving the E3 ligase MKRN2. EXPERIMENTAL APPROACH: Cytotoxicity of DPB was assessed in p53 wild-type and p53-deficient NSCLC cells and ortho

Adjuvanted mucosal vaccination enhances protection and prevents influenza virus transmission in the guinea pig model.

Influenza virus infects the respiratory mucosa, highlighting the importance of mucosal immunity for early protection and transmission control. Here, we evaluated whether intranasal (IN) vaccination with recombinant trimeric hemagglutinin protein from A/Michigan/45/2015 (triHA) formulated with a combined mucosal adjuvant, nanoemulsion plus IVT, an RNA-based RIG-I agonist (NE/IVT), could protect guinea pigs against heterologous A/Netherlands/602/2009 challenge and reduce viral transmission. To compare mucosal and parenteral immunization, IN triHA/NE/IVT was benchmarked against IN triHA alone, IM

Long COVID is associated with lipoxygenase-catalyzed metabolism of essential polyunsaturated fatty acids.

Long COVID (LC) represents a failure of healthy resolution mechanisms for inflammation after SARS-CoV-2 infection. Dietary essential polyunsaturated fatty acids (PUFAs) and their downstream metabolites can orchestrate inflammation resolution, suggesting these pathways may be altered in LC. In this study, plasma samples from two independent participant cohorts totaling 106 participants (designated by chart review as LC or Recovered) were collected longitudinally in the 12 months following SARS-CoV-2 infection and analyzed by mass spectrometry for targeted PUFA metabolites. In the acute phase of

Pooled intravenous immunoglobulins exhibit broad binding and Fc-mediated activity against diverse SARS-CoV-2 variants.

During the coronavirus disease 2019 (COVID-19) pandemic, caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), antibody-based therapies such as COVID-19 convalescent plasma (CCP) and monoclonal antibodies (mAbs) emerged rapidly. However, many CCP clinical trials failed to show consistent benefits, and mAbs lost effectiveness as new SARS-CoV-2 variants emerged. These studies consistently indicated that antibody therapies are effective when given very early in disease and in patients with delayed or suppressed immune responses. Intravenous immunoglobulin (IVIG), which is po

Heterogeneity of MGMT expression in corticotroph adenomas and mechanism of acquired temozolomide resistance.

CONTEXT: Temozolomide (TMZ) can be an effective medical treatment for aggressive pituitary tumors. In case of disease recurrence following TMZ treatment, however, treatment with the drug generally does not control tumor regrowth. OBJECTIVE: This work aimed to better understand the mechanisms of resistance of corticotroph tumors to TMZ in the context of heterogeneity of methylguanine-DNA methyltransferase (MGMT) expression. METHODS: We performed immunohistochemical analysis of the MGMT expression pattern in 25 corticotroph tumors to evaluate intratumoral heterogeneity. In addition, we created

Application of small interfering RNAs and drug combination for pancreatic cancer therapy.

In recent years, pancreatic cancer has become one of the major causes of death among cancers. It has been confirmed that changes in the expression of various genes are important factors associated with the occurrence and development of pancreatic cancer. Small interfering RNAs (siRNAs) are 21-25nt ribonucleotides that target specific DNA sequences and reduce their expression levels. An siRNA delivery system is required as a common and effective method for cancer treatment. Silencing the expression of critical genes (K-Ras, GJB4, SIX4, RARγ, etc) via siRNAs can effectively block the development

Decomposing the decline in life expectancy at birth by cause of death during the COVID-19 pandemic in Canada.

BACKGROUND: Life expectancy at birth has increased in Organisation for Economic Co-operation and Development countries and across the globe over the past few decades, until 2019. However, this trend was disrupted by the COVID-19 pandemic, which saw a significant decrease in life expectancy in Canada and many other countries. Few studies to date have decomposed the life expectancy gap by cause of death observed during the pandemic in a Canadian context. DATA AND METHODS: Death counts were generated from the Canadian Vital Statistics - Death database. Life tables were created using Chiang's met

Differences in SARS-CoV-2 rapid diagnostic testing usage by COVID-19 vaccination status can bias vaccine effectiveness estimates against symptomatic SARS-CoV-2 infection from a test-negative design.

BACKGROUND: Use of rapid diagnostic tests (RDTs) may bias vaccine effectiveness (VE) estimates from test-negative design (TND) studies. We conducted individual-level simulations using national, US COVID-19 surveillance data to assess potential bias at varying likelihoods of visiting a healthcare facility and prevalence of RDT use. METHODS: Twenty-one scenarios that varied the probabilities of visiting a healthcare facility and RDT use by vaccination status were simulated. VE was calculated as 100 × (1 - odds ratio), comparing the odds of symptomatic SARS-CoV-2 infection between vaccinated and

Emergence of Novel Macrolide-Resistant Mycoplasma pneumoniae Sequence Type 3 Subclades in Children after COVID-19 Pandemic, Taiwan.

Post-COVID-19 pandemic Mycoplasma pneumoniae resurgences have occurred globally and have been particularly severe in Taiwan. We conducted a multicenter study to assess M. pneumoniae trends in children. Compared with the 2019 peak, 2024 data revealed a doubling of incidence (546/100,000 population), a 2.2-fold increase in intensive care unit admissions (10.9/100,000 population), and a 13.7-fold surge in central nervous system complications (6.1/100,000 population). In a 2023-2024 prospective cohort (n = 226), intensive care unit admission risk factors were neutrophils >7,000/μL (adjusted odds r

Estimating COVID-19 cumulative incidence from seroprevalence surveys accounting for time-varying seroreversion: A fully Bayesian methodology.

Seroprevalence surveys reveal the extent of humoral immunity against pathogens such as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and under some circumstances represent cumulative incidence of prior infection. However, antibody waning - or seroreversion - biases these estimates by reducing assay sensitivity in a time-varying manner. Because assay sensitivity decays over time, naively using serosurveys can substantially bias estimates of SARS-CoV-2 cumulative incidence and fatality rates. The Bayesian assay-specific, time-varying sensitivity adjustment developed in this paper

Clinical Nursing in Times of War and a Pandemic.

Nurses who served in the Vietnam War and those who cared for patients during the 2020 COVID-19 pandemic surge may be separated by decades, but they share the experience of caring for large numbers of critically injured, ill, or dying patients while risking their own lives. This article reviews the findings of two studies, one conducted in 1986 based on interviews with Vietnam War nurses, and one conducted in 2020 based on interviews with nurses during COVID-19, and explores the idea that professional nurses from different generations have similar thoughts when their lives are in danger and pat

Prevalence of alpha-synuclein seeding activity in the Olfactory Mucosa following SARS-CoV-2 infection: results from a pilot study.

Olfactory dysfunction (OD) is a hallmark of SARS-CoV-2-infection where it is believed to result from neuroinflammation induced by viral invasion via the olfactory mucosa (OM). Hyposmia also affects the majority of patients with Parkinson's disease (PD) where it is caused by α-synuclein (aSyn) pathology in the olfactory system, and aSyn-seeding-activity can be detected in the OM using aSyn-seeding-amplification-assays (aSyn-SAA). The present pilot study aimed to investigate whether SARS-CoV-2-infection induces local aSyn-misfolding and -aggregation in OM samples using aSyn-SAA. We conducted an
