Influenza virus infects the respiratory mucosa, highlighting the importance of mucosal immunity for early protection and transmission control. Here, we evaluated whether intranasal (IN) vaccination with recombinant trimeric hemagglutinin protein from A/Michigan/45/2015 (triHA) formulated with a combined mucosal adjuvant, nanoemulsion plus IVT, an RNA-based RIG-I agonist (NE/IVT), could protect guinea pigs against heterologous A/Netherlands/602/2009 challenge and reduce viral transmission. To compare mucosal and parenteral immunization, IN triHA/NE/IVT was benchmarked against IN triHA alone, IM

Long COVID (LC) represents a failure of healthy resolution mechanisms for inflammation after SARS-CoV-2 infection. Dietary essential polyunsaturated fatty acids (PUFAs) and their downstream metabolites can orchestrate inflammation resolution, suggesting these pathways may be altered in LC. In this study, plasma samples from two independent participant cohorts totaling 106 participants (designated by chart review as LC or Recovered) were collected longitudinally in the 12 months following SARS-CoV-2 infection and analyzed by mass spectrometry for targeted PUFA metabolites. In the acute phase of

During the coronavirus disease 2019 (COVID-19) pandemic, caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), antibody-based therapies such as COVID-19 convalescent plasma (CCP) and monoclonal antibodies (mAbs) emerged rapidly. However, many CCP clinical trials failed to show consistent benefits, and mAbs lost effectiveness as new SARS-CoV-2 variants emerged. These studies consistently indicated that antibody therapies are effective when given very early in disease and in patients with delayed or suppressed immune responses. Intravenous immunoglobulin (IVIG), which is po

BACKGROUND AND OBJECTIVES: Workplace violence (WPV) committed against frontline health care workers is a worldwide concern but relatively little is known about relative rates of reported WPV events as identified through patient safety event (PSE) reports before and during the COVID-19 pandemic. This study sought to understand how the COVID-19 pandemic changed the rates and characteristics of reported WPV events committed by patients against health care workers, as captured in the PSE report system for a Mid-Atlantic health care system.
METHODS: This retrospective cohort study analyzed volunta

BACKGROUND: Life expectancy at birth has increased in Organisation for Economic Co-operation and Development countries and across the globe over the past few decades, until 2019. However, this trend was disrupted by the COVID-19 pandemic, which saw a significant decrease in life expectancy in Canada and many other countries. Few studies to date have decomposed the life expectancy gap by cause of death observed during the pandemic in a Canadian context.
DATA AND METHODS: Death counts were generated from the Canadian Vital Statistics - Death database. Life tables were created using Chiang's met

BACKGROUND: Use of rapid diagnostic tests (RDTs) may bias vaccine effectiveness (VE) estimates from test-negative design (TND) studies. We conducted individual-level simulations using national, US COVID-19 surveillance data to assess potential bias at varying likelihoods of visiting a healthcare facility and prevalence of RDT use.
METHODS: Twenty-one scenarios that varied the probabilities of visiting a healthcare facility and RDT use by vaccination status were simulated. VE was calculated as 100 × (1 - odds ratio), comparing the odds of symptomatic SARS-CoV-2 infection between vaccinated and

Post-COVID-19 pandemic Mycoplasma pneumoniae resurgences have occurred globally and have been particularly severe in Taiwan. We conducted a multicenter study to assess M. pneumoniae trends in children. Compared with the 2019 peak, 2024 data revealed a doubling of incidence (546/100,000 population), a 2.2-fold increase in intensive care unit admissions (10.9/100,000 population), and a 13.7-fold surge in central nervous system complications (6.1/100,000 population). In a 2023-2024 prospective cohort (n = 226), intensive care unit admission risk factors were neutrophils >7,000/μL (adjusted odds r

Seroprevalence surveys reveal the extent of humoral immunity against pathogens such as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and under some circumstances represent cumulative incidence of prior infection. However, antibody waning - or seroreversion - biases these estimates by reducing assay sensitivity in a time-varying manner. Because assay sensitivity decays over time, naively using serosurveys can substantially bias estimates of SARS-CoV-2 cumulative incidence and fatality rates. The Bayesian assay-specific, time-varying sensitivity adjustment developed in this paper

Nurses who served in the Vietnam War and those who cared for patients during the 2020 COVID-19 pandemic surge may be separated by decades, but they share the experience of caring for large numbers of critically injured, ill, or dying patients while risking their own lives. This article reviews the findings of two studies, one conducted in 1986 based on interviews with Vietnam War nurses, and one conducted in 2020 based on interviews with nurses during COVID-19, and explores the idea that professional nurses from different generations have similar thoughts when their lives are in danger and pat

Olfactory dysfunction (OD) is a hallmark of SARS-CoV-2-infection where it is believed to result from neuroinflammation induced by viral invasion via the olfactory mucosa (OM). Hyposmia also affects the majority of patients with Parkinson's disease (PD) where it is caused by α-synuclein (aSyn) pathology in the olfactory system, and aSyn-seeding-activity can be detected in the OM using aSyn-seeding-amplification-assays (aSyn-SAA). The present pilot study aimed to investigate whether SARS-CoV-2-infection induces local aSyn-misfolding and -aggregation in OM samples using aSyn-SAA. We conducted an

This paper describes lessons learned from a partnership between the First Nations Health and Social Secretariat of Manitoba (FNHSSM) and researchers at the University of Manitoba to develop a community-led COVID-19 modelling platform. The envisioned tool would allow First Nations leaders to input local data and generate outputs to support decision-making using a stochastic, age-stratified agent-based model of COVID-19 transmission. Although the platform was not fully operationalized during the project, several important lessons emerged. First, the COVID-19 pandemic placed significant demands o

Cross-reactive SARS-CoV-2-specific antibodies could compromise the specificity of MERS-CoV serology, potentially leading to misclassification of infection history. SARS-CoV-2 exposure induced MERS-CoV antibodies primarily targeting the conserved S2 domain, with occasional neutralizing capacity. Combining MERS-CoV S1 binding with neutralization, however, limited the influence of SARS-CoV-2-induced cross-reactivity on MERS-CoV serology.

We analyzed 919 Streptococcus pyogenes isolates from laboratory-confirmed streptococcal toxic shock syndrome cases in Japan during 2023-2024. Post-COVID-19 resurgence was driven by expansion of M1UK (49.8%) and emerging M113SNPs (37.4%) lineages, replacing ancestral M1global. M113SNPs were associated with younger patient age and higher death rates in unadjusted analyses.

The (large) season-to-season variability and limited dynamical history make the forecasting of infectious diseases a challenging problem. Here, we examine the extent to which advances in data-driven dynamical modeling can provide accurate predictions by benchmarking the performance of one such method, Hankel dynamic mode decomposition (DMD), on the 2024-2025 influenza, respiratory syncytial virus (RSV), and COVID-19 seasons in the United States and Canada. Using Hankel-DMD, we generated weekly forecasts that were submitted to the Center for Disease Control and Prevention's (CDC) FluSight Forec

Passive disease surveillance systems may systematically undercount burden in low-resource minority neighborhoods, yet fine-scale evidence of this phenomenon remains limited. We examined spatiotemporal patterns of reported COVID-19 cases across Miami-Dade County's diverse neighborhoods and investigated which socioeconomic and demographic characteristics predicted areas of elevated and suppressed case rates. In an ecological, cross-sectional analysis of 491,769 geocoded cases across 1,831 census block groups from March 2020 to May 2021, we applied negative binomial regression with spatial eigenv

Health social workers practicing in hospital settings were confronted with numerous personal and professional challenges during the COVID-19 pandemic. Resiliency among frontline health social workers was important for effective practice, retention, and workforce sustainability during the COVID-19 pandemic. The Transactional Model of Stress and Coping was used to better inform how frontline health social workers responded during the pandemic and to identify risk and protective factors for professional social work resiliency. This study examined 163 health social workers employed across several

INTRODUCTION: Anaerobic threshold (AT), a crucial indicator of submaximal exercise capacity and cardiorespiratory function, has been reported to be impaired in individuals with long COVID. However, the predictors of reduced AT during exercise in this population and its impacts on patient-reported outcomes, including sleep quality and health-related quality of life (HRQL), remain unknown. This study aims to identify factors associated with low AT and examine its relationship with patient-reported outcomes.
OBJECTIVE: To investigate the predictors of low AT and compare patient-reported outcomes

Surface functionalization plays a decisive role in the performance of biosensors, as it governs the efficiency and stability of biomolecule immobilization at the sensor interface and, consequently, the overall performance of the biosensing platforms. In this work, we present a comparative study of three organosilane chemistries - APTES, APDMS, and CPTES - applied to a SiO2-terminated 1D photonic crystal able to sustain Bloch surface waves and designed to operate as optical biosensors in both label-free and fluorescence-enhanced modes. Each chemistry was evaluated through a standardized label-f

The envelope (E) protein is a highly conserved structural protein that plays key roles in viral assembly, membrane curvature, and pathogenesis. The E-protein transmembrane (TM) domain oligomerizes into a pentameric ion channel, which contributes to the activation of the NLRP3 inflammasome that is directly linked to acute respiratory distress syndrome (ARDS). Therefore, targeted disruption of E-protein oligomerization represents a powerful therapeutic strategy to attenuate viral virulence. In this study, we report the first DNA aptainhibitors to block E-protein oligomerization. The lead aptainh
