TasTY: Development and evaluation of a taste test for quantifying taste sensitivity among preschool children-test-retest reliability, application, and comparison between children with and without prior SARS-CoV-2 infection.

UNLABELLED: Taste dysfunction gained attention during the coronavirus disease 2019 (COVID-19) pandemic; however, validated objective tests for preschool children remain limited. We aimed to develop a feasible, reliable test quantifying taste sensitivity among children aged 3-6 years and to compare performance between children with and without prior SARS-CoV-2 infection. We included 68 symptom-free children (median age 5.1 years; interquartile range (IQR) 3.9-5.9): 36 without known SARS-CoV-2 infection (control group, CG) and 32 with confirmed prior infection (SARS-CoV-2 infection group, C19-G)

Multiplexed Nucleic Acid Detection Using Sequence-Specific Cleavage Mediated by Repair Endonucleases.

Rapid, portable detection of multiple nucleic acid targets is essential for infectious disease surveillance and precision oncology. CRISPR-based diagnostics have set a high bar for sensitivity and single-base specificity, yet their reliance on collateral nuclease activity complicates multiplexing, integration with amplification, and point-of-care deployment. Here, we present TIMBER (templated incision mediated by endonucleases of repair), a non-CRISPR platform that achieves comparable performance without nonspecific nuclease activity. TIMBER uses a repair endonuclease to cleave probes containi

Highly effective viral RdRP translocation hindrance by the remdesivir nucleotide.

Nucleoside/nucleotide analogs (NAs) typically interfere with substrate selection and catalysis steps of polymerase nucleotide addition cycle (NAC), thereby affecting polymerase catalytic efficiency. In contrast, their interference with post-chemistry translocation is rarely considered in rational design, partly due to a lack of direct and quantitative evidence for such interference. Remdesivir (RDV) is an adenosine analog with broad-spectrum antiviral activity against diverse RNA virus families and has been approved for COVID-19 treatment. RDV is capable of inducing a characteristic "delayed"

Effect of telemedicine-delivered lifestyle interventions in older adults during the COVID-19 pandemic: a nationwide two-arm randomized trial (PROACTIVE-19).

Older adults are at increased risk of severe COVID-19 outcomes, yet randomized evidence on structured lifestyle interventions in this context remains limited. We aimed to evaluate the effects of a personalized lifestyle intervention on health-related behaviors and adverse outcomes among adults aged ≥ 60 years during the COVID-19 pandemic. PROACTIVE-19 was a nationwide, open-label, randomized trial initiated in Hungary in March 2020, during the first wave of the COVID-19 pandemic. Participants aged ≥ 60 years were randomly assigned (1:1) to general (group A) or personalized (group B) lifestyle

Nationwide survey of testing practices for respiratory viruses in Australia in the post COVID-19 pandemic era.

BACKGROUND: In the wake of the coronavirus disease 2019 (COVID-19) pandemic, there have been dramatic changes in the testing protocols for respiratory illness. Laboratory identification of respiratory pathogens causing clinical symptoms has become more important in the post-COVID-19 era, both for epidemiological and clinical reasons. This is particularly the case for respiratory syncytial virus (RSV) where a number of preventatives are at the point of clinical translation. The laboratory diagnosis is one of the critical elements in understanding the disease burden of respiratory pathogens and

BACKGROUND: Since the outbreak of the COVID-19 pandemic, few studies have been carried out on the prevalence of the severe acute respiratory syndrome of the coronavirus 2 (SARS-CoV-2), particularly in groups of patients with comorbidity factors. Moreover, the question of whether HIV increases the risk of contracting SARS-COV-2 or its severity has been the subject of some research. OBJECTIVE: To determine the prevalence of SARS-COV-2 and estabilish the relationship between HIV infection and SARS-CoV-2 infection in female sex workers. METHODOLOGY: Descriptive cross-sectional study of female se

Disseminated BCG disease and recurrent pulmonary infections in a child with trisomy 21 carrying a novel BACH2 variant of uncertain significance: a 5-year case report.

BACKGROUND: Trisomy 21 is associated with combined immune dysregulation, including lymphopenia, impaired responses to vaccines, and heightened susceptibility to respiratory infections, driven in part by triplication of chromosome 21-encoded interferon receptor genes. BACH2 haploinsufficiency is a rare monogenic immunodeficiency characterized by lymphopenia, hypogammaglobulinemia, and susceptibility to infections and autoimmune diseases. Co-occurrence of these two conditions has not been previously described. CASE PRESENTATION: We report a 5-year longitudinal course in a Kenyan girl with triso

Applying normalized p-control charts for early detection of local COVID-19 outbreaks.

The COVID-19 pandemic underscored the urgent need for early detection of local outbreaks to enable better resource allocation and effective mitigation. Outbreak identification largely depended on local infection rates, which were highly variable and influenced by daily testing volumes. Effective pandemic management requires accurate, adaptive methods that reflect the dynamic and localized nature of these rates, ensuring timely and appropriate interventions. This study seeks to improve the early detection and localization of COVID-19 outbreaks through Statistical Process Control (SPC) and healt

Pandemic Politics: Developing Vaccines in Canada's Anti-vax Heartland.

The COVID-19 pandemic in Saskatchewan is a story of extremes. Through most of 2020, low case numbers, effective policy, and broad agreement among government officials, health care workers, and the public sustained a consensus that strict measures and vaccination offered the best responses; however, that consensus collapsed. The province shifted from a focal point for Canadian vaccine development to a staging ground for an anti-vaccine movement that took root on social media and then in street protests. Premier Scott Moe moved from a pro-vaccine, pro-consensus stance to a confrontational positi

Therapeutic mechanistic framework of mesenchymal stem cell-derived extracellular vesicles in neurodegenerative diseases.

Neurodegenerative diseases are a spectrum of progressive disorders unified by chronic neuroinflammation, oxidative and mitochondrial stress, and accumulation of misfolded proteins that ultimately drive neuronal dysfunction and loss. Despite recurring convergence on these pathogenic processes, therapeutic progress has remained limited, underscoring the need for interventions capable of targeting convergent pathogenic pathways rather than single downstream features. Mesenchymal stromal/stem cells (MSCs) have emerged as promising therapeutic candidates through their paracrine actions that modulat

Comparative serum cytokine and chemokine profiles across tuberculosis, latent TB, HIV, and COVID-19 infections in Namibian adults.

BACKGROUND: The devastating pandemic of coronavirus disease 2019 (COVID-19) highlighted the importance of early detection and global collaboration in combating infectious diseases. Namibia is one of the countries that bears a high burden of tuberculosis (TB) and human immunodeficiency virus (HIV) infection. The overlap of these diseases underscores the need to better understand host immune responses, which play a critical role in disease pathogenesis and may differ across infections. This study aimed to profile selected cytokines and chemokines among Namibian individuals with TB, latent TB inf

COVID-19 severity and early inflammatory biomarkers in patients with immune-mediated inflammatory rheumatic diseases: a multicenter cohort study.

Patients with immune-mediated inflammatory rheumatic diseases (IMIRDs) may be more vulnerable to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection because of underlying immune dysregulation. However, it remains unclear which factors determine coronavirus disease 2019 (COVID-19) severity in this population, especially in Eastern European settings. We conducted a retrospective, multicenter observational study including 211 patients with IMIRDs diagnosed with COVID-19 between 01.03.2020 and 31.12.2024. We collected clinical, demographic, and laboratory data within the first 7
