An Intranasal Challenge Model in African Green Monkeys (Chlorocebus aethiops) for Mild-to-Moderate COVID-19 Disease Caused by Subvariant XBB.1.5.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) primarily causes mild to moderate respiratory illness in humans, but infection can also lead to long-term complications, including chronic fatigue, respiratory and cardiac issues, or even death. In November 2021, the emergence of the highly transmissible Omicron variant marked a significant shift in the pandemic, with its subvariants rapidly spreading and continuing to evolve worldwide. The continuing introduction of Omicron subvariants underscores the need for the development of up-to-date vaccines, as well as for appropriate animal

Preclinical Evaluation of the Efficacy of α-Difluoromethylornithine and Sulindac Against SARS-CoV-2 Infection.

Despite numerous research efforts and several effective vaccines and therapies developed against coronavirus disease 2019 (COVID-19), drug repurposing remains an attractive alternative approach for treatment of SARS-CoV-2 variants and other viral infections that may emerge in the future. Cellular polyamines support viral propagation and tumor growth. Here we tested the antiviral activity of two polyamine metabolism-targeting drugs, an irreversible inhibitor of polyamine biosynthesis, α-difluoromethylornithine (DFMO), and a non-steroidal anti-inflammatory drug (NSAID), Sulindac, which have been

Impacts of adipokines in the development and consequence of COVID-19-associated mucormycosis.

Adipokines play fundamental roles in regulating inflammatory responses. Patients with COVID-19 and mucormycosis suffer from inflammatory responses leading to immune dysregulation. This study therefore focused on clarifying whether visfatin and vaspin, known adipokines with immunoregulatory effects, participate in the development and outcome of COVID-19-associated mucormycosis (CAM). Serum samples were obtained from 75 patients suffering from COVID-19 and mucormycosis and 20 non-comorbid healthy individuals. The serum levels of visfatin and vaspin were assessed by enzyme-linked immunosorbent as

Current diagnostic approach to fungal infection in the critically ill patient.

Invasive fungal infections are a major cause of morbidity and mortality in critically ill patients, with an increasing global incidence and species diversity, especially after the SARS-CoV-2 pandemic. Diagnosis relies on a combination of classical methods (microscopy, culture) and non-classical tools including biomarkers (1,3-β-D-glucan, galactomannan, mannan) and molecular assays. Fungal infections (candidiasis, aspergillosis, pneumocystosis, and mucormycosis) requires tailored diagnostic strategies based on host risk factors, epidemiology and specimen type. Combining diagnostic tests improve

Impact of COVID-19 on candidemia: Risk factors and outcomes in a southeastern US cohort.

Candidemia cases increased during the COVID-19 pandemic, likely due to increased critical care interventions. However, the pandemic's impact on candidemia risk factors and outcomes remains understudied. The objective of this study was to evaluate changes in candidemia risk factors and in-hospital mortality before and during the COVID-19 pandemic, stratified by COVID-19 status. We retrospectively analyzed adult candidemia cases (defined by Candida species isolated from blood cultures) at a single center in Albany, Georgia, USA (January 2017-May 2023). Patients were grouped as (1) pre-pandemic,

Repositioning Triazoles as Phosphodiesterase-4 Inhibitors to Suppress COVID-19 Cytokine Storms and Fungal Co-Infections via Docking and Simulation.

Severe COVID-19 cases are often characterised by a hyperinflammatory cytokine storm, which leads to immune dysregulation and increased mortality. Simultaneously, opportunistic fungal infections such as mucormycosis have been increasingly reported, especially in immunocompromised individuals. Triazole antifungals are widely used to treat such infections, but their potential immunomodulatory effects remain underexplored. This study aimed to investigate the off-target potential of commonly used antifungal triazoles itraconazole, ketoconazole, posaconazole and voriconazole against human phosphodie

Phenotypic and genotypic differences between bloodstream and non-blood Candida krusei isolates: implications for invasiveness and antifungal susceptibility.

Candida krusei is an opportunistic pathogen with intrinsic resistance to fluconazole. This study analyzed 174 C. krusei isolates from bloodstream infections (BSI, n = 115) and non-blood sources (n = 59). Biochemical profiling using the VITEK 2 YST ID Card revealed a significant difference in the TyrA reaction between BSI and non-blood isolates (P < 0.05). Antifungal susceptibility testing via CLSI broth microdilution and Sensititre YeastOne showed that BSI isolates were more susceptible to echinocandins and azoles than non-blood strains, with minor MIC discrepancies between methods. Virulence and pathogenicity of secondary fungal infections.

Fungal infections pose a significant global health threat, particularly among immunocompromised individuals facing life-threatening complications. The severity of secondary fungal infections is driven by the expression of virulence factors in immunocompromised individuals. The COVID-19 pandemic has highlighted the susceptibility of immunocompromised patients to secondary fungal infections, prompting a closer examination of the underlying mechanisms. This review provides a comprehensive overview of the virulence mechanisms of major fungal pathogens (Aspergillus, Candida, and Mucorales) in COVID

Cost-effective synthesis of a chromogenic substrate for SARS-CoV-2 Mpro and development of a miniaturized high-throughput inhibition assay.

The SARS-CoV-2 main protease (Mpro) is a critical therapeutic target for antiviral drug development. Here, we report the synthesis and application of a chromogenic substrate handmade for Mpro activity assays. The peptide-pNA conjugate was synthesized via Fmoc-based solid-phase peptide synthesis, followed by solution-phase coupling with pNA. Due to the low nucleophilicity of pNA's aromatic amino group, Oxyma was used to enhance coupling efficiency and reduce side reactions. The crude product showed a yield of 60 % and purity of 75 %. The coupling reaction was optimized and monitored by thin-lay
