Regulation of immune checkpoint molecules in cancer immune evasion and therapy.

Immune checkpoint molecules are essential regulators of immune homeostasis, maintaining the balance between activation and tolerance. In cancer, tumours exploit checkpoint pathways to suppress antitumour immunity and promote progression. The advent of immune checkpoint inhibitors, particularly those that target the clinically validated PDL1-PD1 and CTLA4 axes, has transformed cancer therapy, and the LAG3 axis has recently entered clinical practice, yet most patients experience limited or transient benefit, often because the checkpoint molecules become dysregulated. Here, we examine how multila

Tamoxifen-pretreated MSC exosomes promote endocrine resistance in ER-positive breast cancer cells through the miR-137/Serpina3 axis: a mechanistic study.

BACKGROUND: Tamoxifen (TAM) resistance remains a challenge in estrogen receptor-positive breast cancer treatment. Current research suggested that mesenchymal stem cells (MSCs) derived exosomes may mediate chemoresistance, but the underlying mechanisms are unclear. This study investigates how exosomes from tamoxifen-pretreated MSCs (Tt-MSC-exos) promote tamoxifen resistance through the miR-137/SERPINA3 axis. METHODS: We isolated exosomes using ultracentrifugation. To investigate the effect of Tt-MSC-exos on TAM resistance, we co-cultured Tt-MSC-exos with ER-positive breast cancer cells and eva

A Possibility of Synthetic Lethality: Wee1 Kinase as a Promising Treating Target for Cancer.

Wee kinases, including Wee1, Myt1 (PKMYT1) and Wee2, serve as critical regulators of the DNA damage response by inhibiting the G2/M transition and have attracted increasing attention as a targetable vulnerability in TP53-deficient cancers. Wee1 is the fastest progressing member in clinical research, and its inhibitors such as AZD-1775 and ZN-c3 are under clinical evaluation. Because toxicity and resistance persist with existing agents, research is moving toward newer strategies such as protein degradation technologies. Proceeding from sequence and structure, we, for the first time, use chemoin

Pyrimidine derivatives as anticancer agents targeting kinases: design strategies, biological evaluation, and structure-activity relationship insights.

Protein kinases such as EGFR, VEGFR, PI3K, and CDKs play crucial roles in tumor progression and therapeutic resistance, making them prime targets in anticancer drug development. Among the structural scaffolds explored for designing selective kinase inhibitors, pyrimidine and its derivatives have emerged as highly privileged heterocycles due to their unique electronic properties, hydrogen-bonding capacity, and structural compatibility with kinase ATP-binding pockets. This review summarizes, recent advances in the design, synthesis, structure-activity relationship, and biological evaluation of p

NKD1 promotes the progression and 5-FU resistance of colorectal cancer via the Wnt/β-catenin signaling.

5-Fluorouracil (5-FU) resistance continues to pose a considerable barrier in the postoperative management of advanced colorectal cancer (CRC). This study identified Naked cuticle homolog 1 (NKD1) as a key factor associated with 5-FU resistance in CRC based on sequencing data from the GEO and TCGA databases. Elevated NKD1 expression correlated with adverse clinicopathological features and poor patient survival. Functional assays revealed that NKD1 overexpression elevated the IC50 of 5-FU, promoted tumor cell proliferation, and attenuated apoptosis in both drug-treated and untreated settings. Ad

Chelerythrine: a novel candidate for targeting miR-21/PTEN/PI3K/AKT in nasopharyngeal carcinoma.

BACKGROUND: Nasopharyngeal carcinoma (NPC) is characterized by high metastatic potential and therapeutic resistance, necessitating the development of novel and safe therapeutic strategies. Hyperactivation of the PI3K/AKT signaling pathway, often driven by the oncogenic microRNA-21 (miR-21) and the subsequent suppression of PTEN, plays a key role in NPC progression. Chelerythrine (CHE), a natural plant-derived alkaloid, exhibits potent anti-tumor activities; however, its precise regulatory mechanisms and translational potential in NPC remain elusive. METHODS: Network pharmacology was employed

INHBA secreted by cancer-associated fibroblasts promotes cisplatin resistance and malignant progression of ovarian cancer via ENG-mediated suppression of apoptosis and induction of EMT.

BACKGROUND: Platinum resistance remains a major cause of treatment failure in ovarian cancer (OC). Cancer-associated fibroblasts (CAFs), key components of the tumor microenvironment, play critical roles in OC progression. Inhibin subunit beta A (INHBA) and its putative receptor endoglin (ENG) have been linked to OC chemoresistance, but their functional interaction in mediating cisplatin resistance remains unclear. METHODS: Single-cell RNA sequencing (scRNA-seq) was performed to characterize CAF subsets and assess INHBA and ENG expression in OC tissues. Immunohistochemistry (IHC) and immunoflu

Reactivation of DRP1 plays a functional role in resistance to MEK inhibition in pancreatic cancer cells.

BACKGROUND: In RAS-mutant tumours, ERK phosphorylates the mitochondrial fission GTPase DRP1 to promote mitochondrial fission. DRP1 activity is tumour-promoting in pancreatic and other RAS-driven cancers, but its role in therapeutic resistance is unknown. METHODS: We developed a panel of patient-derived pancreatic cancer cell lines resistant to the MEK inhibitor trametinib. We used immunofluorescence imaging, in vitro growth assays and orthotopic xenografts to determine the role of DRP1 in trametinib resistance. RESULTS: We find that trametinib-resistant cells exhibit increased expression and

Cascade-responsive HA-decorated liposome/mesoporous silica nanoplatform co-delivering BMF-219 and 5-FU enhances gastric cancer chemotherapy through Menin- and p53-associated apoptotic signaling.

BACKGROUND: Our preliminary study found that Men1 is an oncogene and a potential therapeutic target in gastric cancer (GC). Chemotherapy remains a common therapeutic regimen for GC, but is frequently limited by multidrug resistance and poor intratumoral coordination of combination drugs. Targeting Menin may enhance chemotherapy sensitivity, but efficient co-delivery of Menin inhibitors and chemotherapeutics is hindered by the profound spatiotemporal pharmacokinetic decoupling of the drugs in vivo. METHODS: We first evaluated the synergistic antitumor effect and Menin/p53-associated mechanism

Inhibition of DLX6 sensitizes lung adenocarcinoma to cisplatin via GPX4-dependent ferroptosis.

BACKGROUND: Lung adenocarcinoma (LUAD) is a leading cause of cancer-related mortality worldwide, with limited treatment options due to inherent chemotherapy resistance. Ferroptosis, an iron-dependent form of regulated cell death, has recently emerged as a promising strategy to overcome this resistance. Inducing ferroptosis may sensitize LUAD cells to chemotherapy, but its molecular regulators and therapeutic potential remain largely unexplored. This study aims to investigate the role of Distal-less homeobox 6 (DLX6) in regulating ferroptosis and its contribution to LUAD progression. METHODS:

Role of chromatin remodeling and immunopharmacology in cancer therapy.

Epigenetic modifications play a crucial role in cancer, influencing cellular physiology, extracellular matrix (ECM) remodeling, and immune responses. With growing emphasis on chromatin remodeling and the epigenetic regulation of immunity, these regulatory pathways and related players have become focal points of oncological investigations. Epigenetic manipulation of immune system components has emerged as a promising strategy in cancer treatment. Several FDA-approved "epi-drugs" target the epigenome to modulate immune responses and offer new opportunities across different cancers. DNA repair pa

Harnessing tumor-associated exosomal aminopeptidase N as a dual-function tool for monitoring and tackling chemoresistance in gastric cancer.

Chemoresistance remains a major obstacle to effective treatment in gastric cancer (GC), with a notable scarcity of clinically applicable biomarkers and therapeutic targets. In this study, we employed an integrated proteomic approach analyzing paired serum samples from GC patients at disease control and progressive disease stages during chemotherapy, exosomes from chemoresistant and chemosensitive GC cells, and corresponding whole-cell proteomes to identify tumor-associated exosomal drivers of chemoresistance. Cross-dataset integration identified aminopeptidase N (APN/CD13) as selectively enric

Elevated CHAF1A Suppresses Type I Interferon Production and Causes Immunotherapy Resistance in Esophageal Squamous Cell Carcinoma.

In esophageal squamous cell carcinoma (ESCC), chemoradiotherapy potentiates the effects of immune checkpoint inhibitors by activating the tumor-intrinsic innate immune response. However, ESCC cells frequently suppress this activation, which contributes to the high rates of immunotherapy resistance (70%-80%) observed clinically. Thus, identifying intracellular suppressors of this innate immune response remains an unmet critical need. Herein, through multiomics analyses, we identify the chromatin assembly factor CHAF1A as a suppressor of the tumor-intrinsic innate immune response in ESCC. We fou

PPP2R2C promotes aerobic glycolysis and mediates pemetrexed resistance in lung adenocarcinomas by upregulating TCP1-dependent protein folding of glycolytic enzymes.

BACKGROUND: Lung adenocarcinoma (LUAD) remains lethal primarily due to its tendency for early metastasis and the universal development of chemotherapy resistance. The results of this study delineate a coherent signaling axis, originating from the upregulation of PPP2R2C expression to metabolic reprogramming, which ultimately influences the malignant progression and chemotherapy resistance of LUAD. METHODS: GEPIA analysis was used to evaluate PPP2R2C expression in NSCLC. Following shRNA-mediated knockdown, cell proliferation, migration, apoptosis, and glycolytic activity were assessed in NCI-H

MAP2K6 reprograms NAD+ metabolism via the HMGA1-NAMPT axis to sensitize ovarian cancer cells to PARP inhibition.

PARP inhibitors (PARPi) selectively target cancers with homologous recombination deficiency (HRD), yet emerging evidence suggests that additional determinants beyond BRCA/HRD status modulate PARPi response. Here, by integrating unbiased proteomic profiling with systematic drug-response phenotyping across 11 ovarian cancer cell lines, we identify MAP2K6 as a previously unrecognized regulator of PARP1 activation and an important determinant of PARP inhibitor sensitivity in BRCA-wild-type ovarian cancer. MAP2K6 promotes global PARP1-mediated PARylation and enhances cellular responsiveness to the
