Staphylococcus aureus (S. aureus) remains a major global health threat with high mortality, causing both community-associated and hospital-acquired infections. Consequently, resistance mechanisms, notably β-lactamase production and biofilm formation, are considered promising therapeutic targets. In this study, Catharanthine, a plant-derived alkaloid extracted from Catharanthus roseus (Madagascar periwinkle), is a potent adjuvant that restores the efficacy of β-lactam antibiotics against S. aureus. This study aimed to investigate resistance mechanisms, notably β-lactamase production and biofilm

Carbapenem resistance among Gram-negative bacteria remains a serious public health threat, with metallo-β-lactamase (MBL)-producing organisms increasing substantially in prevalence and few therapeutic options available to adequately address these difficult-to-treat pathogens. Cefepime-zidebactam (FEP-ZID) is a novel β-lactam/β-lactam enhancer combination that offers a mechanistically distinct approach to overcoming resistance in Gram-negative organisms. Zidebactam contributes three complementary mechanisms: inhibition of Class A and C β-lactamases, direct antibacterial activity via PBP2 inhibi

Acinetobacter baumannii has traditionally been considered a nosocomial pathogen, but cases of community-acquired A. baumannii pneumonia have increasingly been reported in tropical Asia, including Vietnam. Data on its clinical characteristics, antimicrobial resistance, and patient outcomes remain limited. This study aimed to describe the clinical profile, resistance patterns, and outcomes of community-acquired A. baumannii pneumonia in southern Vietnam. We performed a single-center retrospective cohort study at Nhan Dan Gia Dinh Hospital, a 1500-bed tertiary center in Ho Chi Minh City (January

OBJECTIVES: Antimicrobial resistance (AMR) is a serious threat to human and animal health; the emergence of 3rd-generation cephalosporin- and fluoroquinolone-resistant Enterobacterales is particularly problematic. Because Escherichia coli is the most common cause of urinary tract infections in dogs, AMR has real-world impact. The objectives of this study were to evaluate the antimicrobial susceptibility of canine urinary isolates of E. coli and to determine if AMR is increasing in Saskatchewan.
ANIMALS AND PROCEDURE: A total of 576 diagnostic isolates of canine urinary E. coli from individual

We evaluated the presence and phenotypic impact of PBP3 insertions YRIN (n = 44) and YRIK (n = 18) among 124 Escherichia coli isolates belonging to high-risk lineages selected from 1,471 isolates displaying resistance to β-lactams. PBP3 insertions were detected among 62 isolates. YRIN insertions were detected among isolates belonging to ST167 (n = 23), ST361 (n = 7), ST410 (n = 10), ST617 (n = 1), and ST648 (n = 3), while YRIK insertions were observed in ST405 (n = 9), ST410 (n = 5), and ST617 (n = 4). Aztreonam-avibactam inhibited 90.3% of all isolates from high-risk clones and 80.6% of the i

BACKGROUND: Understanding the causative microorganisms and initiating appropriate empirical antibiotics early are important in the management of knee periprosthetic joint infections (PJIs). The aim of this study was to identify trends in PJI microorganisms and antibiotic resistance profiles over 24 years to guide empirical antibiotic selection.
METHODS: This study included 487 first-episode PJIs identified between 2000 and 2023 following primary total knee arthroplasty (TKA) at 3 large tertiary hospitals. PJIs were classified using the Tsukayama classification, which is based on the timing fr

INTRODUCTION: Methicillin-resistant Staphylococcus aureus (MRSA) is a leading cause of severe infections with excess mortality. Progress with traditional antibiotics has been incremental, while resistance, persistence, tolerance, and biofilm formation continue to erode effectiveness. Parallel advances in small-molecule discovery, long-acting lipoglycopeptides, next-generation β-lactams, and non-traditional modalities such as bacteriophage lysins have renewed interest in expanding therapeutic options, though transition from promising preclinical signals to clinical benefit remains challenging.

The treatment with antibiotics during the fattening of broilers has stagnated in Germany over the last years, while the national aim is to further reduce the use of antibiotics in human and veterinary medicine due to the occurrence of antibiotic-resistances in microorganisms. Therefore, the aim of this study was to identify relevant management measures in broiler farms which improve the health status of broilers and could reduce the occurrence of antibiotic-resistant, opportunistic pathogens in the broilers' environment. A survey was conducted querying existing management measures based on sev

Although protein-protein interactions (PPIs) are central to nearly all biological processes, identifying and engineering high-affinity intracellular binders remains a significant challenge due to the complexity of the cellular environment and the folding constraints of proteins. Here, we present a two-stage complementary platform that combines magnetic-activated cell sorting (MACS)-based yeast surface display with functional ligand-binding identification by twin-arginine translocation (Tat)-based recognition of associating proteins (FLI-TRAP), a bacterial genetic selection system for efficient

BACKGROUND: Pseudomonas aeruginosa remains an urgent public health threat due to the many life-threatening diseases it causes, wide dissemination of multidrug resistant strains, and ability to survive and persist on hospital surfaces. Its accessory genome, composed of genes present in a subset of strains, may provide a clue to its success as an opportunistic pathogen. Here, we aim to characterize the pool of mobile genetic elements (MGE), anti-phage defenses and antimicrobial resistance (AMR) genes in P. aeruginosa. RESULTS: Using a dataset of 1,045 complete genomes, we identified 173 plasmids

Anaerobic bacteria in the human intestinal microbiota play a pivotal role in antibiotic resistance, primarily through the production of β-lactamases. This narrative review explores the diversity of β-lactamases found in intestinal anaerobes, their functional characteristics, and clinical implications. Species of the genus Bacteroides are major producers, harboring enzymes from Ambler classes A (e.g. CepA, CfxA and CblA), B (e.g. CfiA and CcrA) and D (e.g. OXA-347), with activity ranging from cephalosporinases to carbapenemases and oxacillinases. Other gut members, including Clostridioides diff

Combined treatment with aztreonam (an established intravenous antibiotic) and avibactam (a β-lactamase inhibitor) has the potential to address the unmet need for safe and effective agents to combat infections caused by Gram-negative bacteria producing metallo β-lactamases (MBLs). Aztreonam-avibactam may also address other problematic β-lactamases, such as extended-spectrum β-lactamases and serine carbapenemases, which may be co-expressed with MBLs and contribute to a multidrug-resistant phenotype. Based on completion of two Phase 3 trials, aztreonam-avibactam was approved in Europe (2024), the

Northern public beaches are popular recreational and touristic destinations in Mauritius. However, rapid urbanization and coastal development in the region raise concerns about the microbial quality and safety of nearshore waters, which may deteriorate without effective surveillance and management. This could be exacerbated by the occurrence of marine, human pathogenic Vibrio, which may harbor genes conferring virulence and antimicrobial resistance. The aim of the study was thus to assess the microbiological safety and sanitary quality of Northern coastal seawater. The specific objectives were

Although recommendations for nutritional factors have already been studied, derived and published, nutrients like trace elements continue to be much higher supplemented in broiler feed than required. If these nutrients, such as heavy metals as trace elements, are excreted due to an oversupply, they might accumulate in the environment and cause a major impact due to the risk of antibiotic-resistant co-selection. The aim of this study was to evaluate the accumulation of these nutritional factors in environmental samples, and their influential risk for the occurrence of antibiotic-resistant oppor

Aeromonas veronii, a Gram-negative pathogen, is ubiquitous in nature and infects broad hosts, which becomes a serious threat to the public health. The cyclic adenosine monophosphate (cAMP), precisely modulated by cyclic nucleotide phosphodiesterase (PDE), regulates key cellular processes in bacteria. However, the specific function and regulatory role of PDE in A. veronii remain unclear. In this study, a Class III PDE gene named as cpdA in A. veronii WL-3 was identified for the first time, which shared 39.18%-49.19% identity with homologs from other bacteria and also contained the conserved met
