Metagenomic analysis of UK retail foods finds limited evidence for associations between food production method and antimicrobial resistance gene burden.

Food is produced by a range of methods including extensive (organic and free range), intensive (conventional) and wild-caught production systems. Antimicrobial use varies between different food production systems, which may affect the microbial populations as well as the prevalence and diversity of antimicrobial resistance genes (ARGs) found on food at retail. In this study, shotgun metagenomics was used to investigate the microbial and ARG composition of 25 pork, 33 beef, 33 lamb, 60 chicken, 31 salmon and 41 leafy green samples collected in Norfolk, England, and labelled as extensive, wild c

CoNS blood culture isolates from very preterm infants: antibiotic susceptibility, synergy testing and mecA gene expression.

OBJECTIVES: Previous reports suggest that neonatal sepsis with methicillin-resistant (MR) CoNS can be treated with first-generation cephalosporins. We have evaluated the in vitro effect of cefazolin and other antibiotics, synergy between β-lactams and aminoglycosides, and mecA expression in MR-CoNS isolates. PATIENTS AND METHODS: Antimicrobial susceptibility testing was performed in 167 CoNS isolates from 135 very preterm infants (<32 weeks gestation) with late-onset sepsis. Microbroth dilution chequerboard assay explored antibiotic synergy. PCR detected mecA and three aminoglycoside-modifyin

Phenotypic and genomic characterization of Castellaniella ginsengisoli, an emerging pathogen associated with disease in birds.

The environmental bacterium Castellaniella revealed its clinical relevance from case reports, causing fatal infections in animals. The lack of phenotypic and genomic characterization may have historically contributed to the scattered identification of Castellaniella in the field and in the laboratory. This pioneer study addressed this knowledge gap by performing phenotypic and genomic characterization of C. ginsengisoli isolated from 22 clinical cases of chickens. We observed that C. ginsengisoli isolates exhibited three colony morphologies on blood agar and presented flagella under transmissi

Functional characterization of a novel class A carbapenemase CAE-1 in carbapenem-resistant Pseudomonas aeruginosa clinical isolates.

Two carbapenem-resistant Pseudomonas aeruginosa (CRPA) isolates were collected, which lacked known carbapenemases but produced a β-lactamase CAE-1. CAE-1 was recently found in Comamonas aquatica and conferred resistance to penicillins and cephalosporins. In this study, we aim to know whether CAE-1 was responsible for the carbapenem resistance in the CRPA isolates and its enzyme hydrolyzing characteristics. Both CRPA clinical isolates exhibited minimal inhibitory concentrations (MICs) of 8 µg/mL for imipenem and meropenem, with a positive result in the modified Hodge test for meropenem. The bla

Utilizing whole genome sequencing data for machine learning driven prediction of antibiotic resistance in Escherichia coli.

BACKGROUND: Antimicrobial resistance (AMR) is an escalating global public health threat that substantially undermines the effectiveness of standard anti-infective therapies and increases the risk of adverse clinical outcomes. METHODS: We analyzed whole-genome sequencing (WGS) data from 1952 Escherichia coli isolates with AST phenotypes. Gene, single-nucleotide polymorphisms (SNPs), and k-mer features were extracted to train machine-learning classifiers for predicting resistance to 10 common antibiotics. Model performance was evaluated using the area under the receiver operating characteristic

Antimicrobial use contributes to resistance gene enrichment across cattle groups on commercial dairy farms.

Antimicrobial use (AMU) in agricultural systems is frequently linked to antimicrobial resistance (AMR). Yet, the scale at which AMU reshapes host-associated resistomes remains unclear. This gap arises, in part, from the scarcity of farm-level AMU data from commercial production systems. Here, we combine detailed AMU records from commercial dairy farms with metagenomic analyses of bovine fecal resistomes from calves, lactating cows, sick cows, and cull cows. At a broad level, resistome profiles were similar regardless of farm AMU. Resistance associated with historically common antibiotics, such

Combating New Delhi metallo-beta-lactamase-1 (NDM-1): an integrated review of inhibitor development and next-generation therapeutic platforms.

New Delhi metallo-β-lactamase-1 (NDM-1), which has emerged globally, exhibits resistance to almost all β-lactam antibiotics, including carbapenems, posing a major challenge to modern antibiotic therapy. Deep sequencing has revealed the evolution of several new NDM variants (some of the variants are more thermostable than NDM-1) due to mutations that increase their zinc-binding and catalytic efficiency and establish a selective pressure on the enzymes. In addition to enzymatic breakdown, NDM-mediated resistance is exorbitantly exhibited via porin loss, efflux pump activity, biofilm formation, a

Clinical and Epidemiological Landscape of Antimicrobial Resistance and Virulence in Streptococcus Species.

Species of the genus Streptococcus constitute important pathogens in human and veterinary medicine, being responsible for a wide spectrum of infections ranging from mild illnesses to severe invasive pathologies. Although β-lactams continue to be effective against most species, the global increase in resistance to macrolides, lincosamides, tetracyclines, and, in some cases, reduced susceptibility to penicillin represents a growing challenge for antimicrobial therapy. This review synthesizes the clinical and epidemiological landscape of antimicrobial resistance in the main Streptococcus species,

Optimizing Infection Risk in Primary Total Knee Arthroplasty: A Narrative Review of Current Concepts, Controversies, and Evidence Gaps.

Periprosthetic joint infection (PJI) remains one of the most serious complications following primary total knee arthroplasty (TKA), despite its relatively low incidence. Prevention requires addressing multiple interacting factors related to patient characteristics, microbial burden, antimicrobial prophylaxis, operative technique, and postoperative wound management. The objective of this review is to synthesize contemporary evidence on strategies for reducing infection risk in primary TKA, with particular emphasis on clinically actionable interventions, areas of controversy, and current evidenc

Chemical residues in agricultural land in Germany between 2022 and 2024 - prevalence of selected antibiotic resistance genes in soil.

Growing concern about antibiotic resistance genes (ARGs) in agricultural soils, particularly due to application of organic fertilizers-has increased the need to better understand environmental reservoirs and mechanisms of antimicrobial resistance and their potential risks to human and animal health. In this study, soils from 51 agricultural sites across Germany were sampled between 2022 and 2024 aiming to determine the antibiotic resistance occurrence and pharmaceutical product contamination in the soils, and to evaluate the influence of different fertilization practices (mineral, organic, or

Concurrence of antibiotic resistance genes in plasmid genomes shape environmental resistomes.

Horizontal transfer of plasmid-associated antibiotic resistance genes (ARGs) plays a pivotal role in environmental antibiotic resistance dissemination. Here, we characterized ARG concurrence patterns in plasmid genomes and examined plasmid-associated ARGs across 106 environmental metagenomes. Approximately half of known ARG subtypes (257) occurred in plasmid genomes, and nearly one-quarter of plasmids carried ARGs, including "super plasmids" harboring over 20 ARG subtypes spanning 10 antibiotic categories. Aminoglycoside resistance genes (AmRGs) exhibited the highest concurrence frequency (CF)

Spread potential and architectural variant of a multiple-drug resistant Escherichia coli co-producing lipopolysaccharide-modifying determinants and extended-spectrum β-lactamases.

BACKGROUND: Abuse of β-lactam antibiotics and its spread of drug resistance challenged the applicability of antimicrobial agents, enabling polymyxins as a last-line treatment for anti-infections of multi-drug resistant (MDR) pathogen. Polymyxin is applicable to herb breeding/growth prompting and combatting Gram-negative bacteria by combining other antimicrobials by membrane dysfunction, permeability damage, vesicle-vesicle contacts and oxidative stress, with its increasingly ubiquitous resistance. However, the complex mechanisms and high transmission risk of polymyxin resistance pose a severe
