The toxin from the PemIK-Sa1 toxin-antitoxin system decreases Staphylococcus aureus resistance to antibiotics.

Toxin-antitoxin (TA) systems are bacteria-specific components involved in maintaining mobile genetic elements that may carry antibiotic resistance determinants (ARDs), among other functions. PemIK-Sa1 is a plasmid-encoded TA system found in Staphylococcus aureus, a dangerous pathogen affecting humans and animals. The PemK-Sa1 toxin is a sequence-specific ribonuclease inhibited by its cognate antitoxin, PemI-Sa1. Using recombinant multidrug-resistant strains, we demonstrated that expression of the toxin reduces resistance to antibiotics from various classes, including β-lactams, fluoroquinolone

Mycoplasma hyosynoviae-associated arthritis in finisher pigs showing signs of lameness in Southwestern Norway.

Lameness in pigs is a major welfare and economic concern, with infectious arthritis being an important contributor. Mycoplasma hyosynoviae is a recognized cause of arthritis in finisher pigs globally, yet its occurrence and clinical relevance in Norway have not been systematically investigated. This small-scale study aimed to elucidate the aetiology of acute lameness in finisher pigs in southwestern Norway through systematic necropsies and microbiological analyses. Between September 2020 and April 2022, ten farms in Rogaland County with recurrent episodes of acute lameness were enrolled. Farm

A Trench Plate-Based Approach for the Selective Isolation of Metal-Cycling Bacteria.

OBJECTIVE: Several reports have suggested that IGF-I levels in patients with childhood-onset (CO) GH deficiency are lower than those observed in patients with adult-onset (AO) GH deficiency. However, these reports are unsatisfactory as there are several differences between the cohorts studied other than the timing of onset of GH deficiency; in particular, the patients were not matched for equal severity of GH deficiency. We have pursued this question further by examining the IGF-I standard deviation score (SDS) in patients with CO and AO GH deficiency, with equal degrees of severity of GH defi

Stability Study of Meropenem 50 mg/mL Eye Drops in Polypropylene Dropper Bottles.

Background/Objectives: Meropenem is a broad-spectrum carbapenem antibiotic with demonstrated efficacy against multidrug-resistant Gram-negative pathogens. Although its use as an ophthalmic formulation is off-label, growing clinical evidence supports its application in severe ocular infections such as keratitis and endophthalmitis. However, the intrinsic instability of meropenem in aqueous solutions and the absence of standardized ophthalmic preparations limit its routine use. Furthermore, no stability studies are currently available for meropenem 50 mg/mL eye drops stored in polypropylene (PP)

Single- and Double-Chain Arginine-Derived Surfactants: Antimicrobial, Antibiofilm and Synergistic Activities.

The rise in antimicrobial resistance and the resilience of microbial biofilms demand innovative strategies that combine, for instance, membrane-active agents with marketed drugs. Arginine-based surfactants are promising alternatives to conventional quaternary ammonium compounds, but comparative data on their antimicrobial, antibiofilm and modulatory activities remain limited. Five arginine-derived surfactants, the single-chain Nα-lauroyl-L-arginine methyl ester (LAM) and ethyl ester (LAE), together with their double-chain homologues LANHC3, LANHC5 and LANHC8 were evaluated against Gram-positiv

Peptidoglycan recycling is critical for cell division, cell wall integrity, and β-lactam resistance in Caulobacter crescentus.

Most bacteria possess a peptidoglycan (PG) sacculus, which is continuously remodeled during cell growth and division. The turnover products generated in this process are typically imported into the cell and reused for PG biosynthesis. While the underlying pathways have been studied intensively in gammaproteobacteria, knowledge of their presence and physiological roles in other bacterial lineages remains limited. Here, we comprehensively investigate PG recycling in the alphaproteobacterial model organism Caulobacter crescentus. Characterizing the activities of key enzymes in vitro and in vivo,

Consensus Guidance for Beta-Lactam Antibiotic Dose Individualization in Acutely Ill Patients: Endorsed by the American College of Clinical Pharmacy, European Society of Clinical Microbiology and Infectious Diseases, European Society of Clinical Microbiology and Infectious Diseases PK/PD of Anti-Infectives Study Group, Infectious Diseases Society of America, International Association of Therapeutic Drug Monitoring and Clinical Toxicology, Society of Critical Care Medicine, and the Society of Infectious Diseases Pharmacists.

Beta-lactam antibiotics (beta-lactams) are first-line treatments for most major infectious syndromes in acutely ill patients, with in vitro activity against common pathogens, demonstrated efficacy in trials, and a perceived low risk of adverse effects. The current dosing approach, informed by population-level data, tends to be simplistically reduced to a "one size fits all" dosing method which only accounts for body weight (i.e., in pediatrics) and end organ function to estimate drug clearance (e.g., estimated glomerular filtration rate); this approach results in substantial variability in obs

Beyond Carbapenemases: Chromosomal Resistance Mechanisms and Carbapenemase Type Determine Susceptibility to Novel β-Lactam/β-Lactamase Inhibitor Combinations and Cefiderocol in Carbapenem-Resistant Pseudomonas aeruginosa from China.

BACKGROUND: β-lactam/β-lactamase inhibitor combinations (BL/BLIs) and siderophore cephalosporin serve as critical polymyxin alternatives for carbapenem-resistant Pseudomonas aeruginosa (CRPA). However, systematic multicenter clinical data from China and the resistance mechanisms underlying non-susceptibility to these agents remain poorly defined. METHODS: We collected 442 CRPA isolates from 10 hospitals across eight Chinese provinces (2017-2025), determined minimum inhibitory concentrations (MICs) for cefiderocol (FDC), ceftazidime-avibactam (CZA), imipenem-relebactam (IMR), and ceftolozane-t

Frequency of CIT, EBC and DHA flocks (families) of AmpC beta-lactamases in clinical isolates of Klebsiella pneumoniae collected from hospitalized patients in North of Iran.

BACKGROUND: Klebsiella pneumoniae exhibits a marked propensity for acquiring beta-lactam resistance genes. Among these, AmpC beta-lactamases are able to hydrolyze a broad spectrum of antibiotics including first- through third-generation cephalosporins, cephamycins, and aztreonam. The CIT, EBC, and DHA families are among the most clinically significant plasmid-mediated AmpC variants in this pathogen. Consequently, this study aimed to investigate the prevalence of genes encoding these specific enzymes among clinical isolates of K. pneumoniae in northern Iran. METHODS: One hundred clinical isola

The mecillinam resistome in Klebsiella pneumoniae: how resistance to a one-target β-lactam triggers a diversity of responses.

Antimicrobial resistance is a silent pandemic responsible for 1.14 million deaths worldwide in 2021, with a major contribution from Klebsiella pneumoniae. β-lactams are the most commonly used antibiotics in humans, and there is an urgent need to characterize the resistance mechanisms to these antibiotics in Enterobacterales other than Escherichia coli. Mecillinam is a narrow-spectrum β-lactam targeting a single penicillin-binding protein, PBP2. Pivmecillinam, its oral prodrug, is used as a first-line treatment for uncomplicated urinary tract infections. It has been used for decades in Europe b

High in vitro activity of aztreonam/avibactam against NDM-producing Enterobacterales with emerging resistance in Escherichia coli in the United States Mid-Atlantic region.

UNLABELLED: New Delhi metallo-β-lactamase (NDM)-producing Enterobacterales represent a critical antimicrobial resistance threat with limited therapeutic options. Aztreonam/avibactam, which combines the stability of aztreonam against metallo-β-lactamases with the inhibition of serine β-lactamases by avibactam, is a promising therapeutic candidate. However, emerging resistance associated with ftsI (penicillin-binding protein 3 [PBP3]) insertions and expression of AmpC β-lactamases has been described. We evaluated 95 NDM-producing Enterobacterales isolates submitted to the Maryland Department of

Brevibacterium casei: An Unusual Cause of Peritoneal Dialysis-Associated Peritonitis Managed by Catheter Salvage.

Peritoneal dialysis (PD)-associated peritonitis remains the leading infectious complication of the technique and a major cause of catheter loss and transfer to haemodialysis. Although it is usually caused by common Gram-positive organisms, rare pathogens such as Brevibacterium casei are occasionally implicated. These coryneform bacteria are easily mistaken for skin contaminants, since they are part of the commensal flora, and may require molecular methods for identification. They are typically resistant not only to first-line beta-lactams but also to third-generation cephalosporins, clindamyci

Association of minimum inhibitory concentration with outcome in patients with Stenotrophomonas maltophilia bacteraemia treated with empirical levofloxacin.

Stenotrophomonas maltophilia is intrinsically resistant to carbapenems and most β-lactam antibiotics. Empiric therapy for bloodstream infections often includes either trimethoprim-sulfamethoxazole or levofloxacin. However, there are currently no European Committee on Antimicrobial Susceptibility Testing (EUCAST) breakpoints available for interpretation of susceptibility testing for levofloxacin. We examined the relationship between levofloxacin minimum inhibitory concentration (MIC) and clinical outcomes in S. maltophilia bacteraemia. We retrospectively examined 79 consecutive patients with S.

In vivo evolution of resistance to contemporary β-lactam/β-lactamase inhibitor combinations during treatment of a KPC-producing Serratia marcescens infection.

Klebsiella pneumoniae carbapenemases (KPCs) are a family of serine β-lactamases that confer broad antibiotic resistance by hydrolyzing virtually all β-lactam (BL) agents. Contemporary β-lactamase inhibitors (BLIs) such as avibactam were developed to neutralize the activity of KPCs and other clinically important carbapenemases. Ceftazidime-avibactam (CZA), a BL/BLI combination in which the cephalosporin ceftazidime is protected from KPC-mediated hydrolysis, demonstrated improved outcomes in early clinical use. However, CZA-resistant isolates soon emerged. Herein, we describe a challenging clini

Modulation of macrophage survival during Klebsiella pneumoniae infections.

Klebsiella pneumoniae, a facultative anaerobe and Gram-negative bacterium, has high clinical significance in public health concerns. K. pneumoniae is given its name in recognition of the German-Swiss microbiologist Edwin Klebs. Carl Friedlander first identified K. pneumoniae as a bacterium isolated from the lungs of individuals who had died from pneumonia. K. pneumoniae is a prominent member of the Enterobacteriaceae family, an opportunistic pathogen, and one of the pathogens sweeping the world in the antibiotic resistance pandemic. This organism accounts for approximately one-third of all Gra
